The complete cDNA and deduced amino acid sequences of the ribosomal proteins S23(AmphiS23)and L35(AmphiL35)from amphioxus Branchiostoma belcheri tsingtauense were identified inthis study.AmphiS23 cDNA is 546 bp long and encodes a protein of 143 amino acids.It has a predictedmolecular mass of 15,851 Da and a pI of 10.7.AmphiL35 cDNA comprises 473 bp,and codes for a proteinof 123 amino acids with a predicted molecular mass of 14,543 Da and a pI of 10.8.AmphiS23 shares morethan 83% identity with its homologues in the vertebrates and more than 84% identity with those in theinvertebrates.AmphiL35 is more than 63% identical to its counterparts in the vertebrates and more than52% identical to those in the invertebrates.Southern blot analysis demonstrated the existence of 1-2 copiesof the S23 gene and 2-3 copies of the L35 gene in the genome of amphioxus B.belcheri tsingtauense.Thisis in sharp contrast to the presence of 6-13 copies of the S23 gene and 15-17 copies of the L35 gene in therat genome.It is clear that the housekeeping genes like S23 and L35 underwent a large-scale duplication inthe vertebrate lineage,reinforcing the gene/genome duplication hypothesis.

In this study, we describe the partial genomic organization of ribosomal protein S7 gene isolated from the mosquito Anopheles stephensi. Initially a 558 bp partial cDNA sequence was amplified as precursor mRNA sequence containing 223 bp long intron. 5＇ and 3＇ end sequences were recovered using end specific rapid amplification of cDNA ends （RACE） polymerase chain reaction. The full-length cDNA sequence was 914 nucleotide long with an open reading frame capable of encoding 192 amino acid long protein with calculated molecular mass of 22174 Da and a pI point of 9.94. protein homology search revealed 〉75% identity to other insect＇s S7 ribosomal proteins. Analysis of sequence alignment revealed several highly conserved domains, one of which is related to nuclear localization signal （NLS） region of human rpS7. Interestingly, intron nucleotide sequence comparison with A. gambiae showed a lesser degree of conservation as compared to coding and untranslated regions. Like this, early studies on the genomic organization and cDNA/ Expressed sequence tag analysis （EST） could help in genome annotation ofA. stephensi, and would be likely to be sequenced in the future.

Although ribosomal proteins are known for playing an essential role in ribosome assembly and protein translation,their ribosomeindependent functions have also been greatly appreciated.Over the past decade,more than a dozen of ribosomal proteins have been found to activate the tumor suppressor p53 pathway in response to ribosomal stress.In addition,these ribosomal proteins are involved in various physiological and pathological processes.This review is composed to overview the current understanding of how ribosomal stress provokes the accumulation of ribosome-free ribosomal proteins,as well as the ribosome-independent functions of ribosomal proteins in tumorigenesis,immune signaling,and development.Wealso propose the potential of applying these pieces of knowledge to the development of ribosomal stress-based cancer therapeutics.

Ribosomes are important organelles for synthesizing proteins in cells.They are composed of ribosomal RNA and more than 80 ribosomal proteins.It is well known that an essential function of ribosomal proteins is to participate in protein translation.In addition,ribosomal proteins also perform extra-ribosomal functions,such as participating in DNA replication,transcription,and damage repair,regulating cell growth,proliferation,apoptosis,and transformation.In recent years,studies have shown that alterations in ribosomal protein synthesis or function can lead to various hematologic diseases,including Diamond-Blackfan anemia,5q-syndrome,Shwachman-Diamond syndrome,and other blood system diseases.Moreover,abnormal expressions of specific ribosomal protein genes have been reported in many malignant tumors.In this review,we elaborated on the changes in ribosomal proteins in hepatocellular carcinoma and colorectal,prostate,gastric,esophageal,and other cancers and discussed the relationship between ribosomal proteins and the occurrence of hematologic disorders and cancers.

Chloroplasts （plastids） possess a genome and their own machinery to express it. Translation in plastids occurs on bacterial-type 70S ribosomes utilizing a set of tRNAs that is entirely encoded in the plastid genome. In recent years, the components of the chloroplast translational apparatus have been intensely studied by proteomic approaches and by reverse genetics in the model systems tobacco （plastid-encoded components） and Arabidopsis （nucleus-encoded components）. This work has provided important new insights into the structure, function, and biogenesis of chloroplast ribosomes, and also has shed fresh light on the molecular mechanisms of the translation process in plastids. In addition, mutants affected in plastid translation have yielded strong genetic evidence for chloroplast genes and gene products influencing plant develop- ment at various levels, presumably via retrograde signaling pathway（s）. In this review, we describe recent progress with the functional analysis of components of the chloroplast translational machinery and discuss the currently available evidence that supports a significant impact of plastid translational activity on plant anatomy and morphology.

Kinesin is a superfamily of microtubule-based motor proteins that move directionally along microtubules and involved in various essential biological processes in plant development.However,few kinesins have been functionally identified in plant embryo and seed.Their function in embryogenesis and seed formation is largely unknown.Here,we report NtKRP,a novel motor protein of kinesin-12 family with unique structure.NtKRP predominantly expresses in the tissues where cells are actively dividing,especially in embryo and embryonic root during seed germination.NtKRP RNAi lines displayed a reduction of cell number in meristematic zone and reduced length of embryonic root.The size of matured embryo also obviously reduced and this resulted in smaller seeds,the reduced seed yield as well.Furthermore,we identified a 60S ribosomal protein NtRPL17 as NtKRP interacting protein.NtRPL17was universally,but preferentially expressed in embryo and active-dividing cells.The phenotypes of NtRPL17 RNAi lines strongly resemble that of NtKRP RNAi lines,indicating they functions in the same pathway.Further experiments verified that NtKRP and CDKA:1 were regulated as a couple of co-expressed genes in vivo.CDKA;1 that possess kinase activity could bind to NtKRP at its phosphorylation sites,suggesting NtKRP activity could be regulated by CDKA;1 phosphorylation.Accordingly,we show evidences for that cell division delayed at G2-M transition phase,which led to the cell number decreased in embryos of NtKRP-and NtRPLl 7-silenced lines.We also revealed mat the tail domain of NtKRP that is usually involved in binding cargo lacks of consensus CDKA;1 phosphorylation sites,but can also interact with CDKA;1.This unique feature implies its specific regulatory mechanism of the novel NtKRP compared to other members in kinesin-12 family.NtKRP probably first carries its regulator CDKA;1 to the targeting sites and is then regulated by CDKA;1 phosphorylation for playing its active role in embryogenesis.

ribosomal proteins(RP) has been reported as a central player in the translation system,and are required for the growth and maintenance of all cell kinds.RP genes form a family of homologous proteins that express in the stable pattern and were used for phylogenetic analysis.Here we constructed a cDNA library of alpaca skin and 13,800 clones were sequenced.In the cDNA library,RP genes from skin cDNA library of alpaca were identified.Then 8 RP genes were selected at random and built the Phylogenetic trees from the DNA sequences by using parsimony or maximum likelihood methods for molecular and evolutionary analysis of ribosomal proteins.The results showed that the 42 RP genes of alpaca have been expressed in alpaca skin.They were highly conserved.The phylogeny inferred from all these methods suggested that the evolutionary distances of alpaca RP genes were closer to rat.
