P2X receptors are a family of cation channels expressed in neurons and nonneuronal *** play a variety of roles including synaptic transmission,muscle contraction,neuronal secretion and inflammation response. Security breach:peripheral nerves provide unrestricted access for toxin delivery into the central nervous system

Objective Glaucoma is the second leading cause of irreversible blindness worldwide. Elevated intraocular pressure（IOP） is the most important risk factor for developing glaucoma. Current treatments for glaucoma are focused on the reduction of IOP, such as medications and surgery. However, in many patients the disease still processes after reduction of IOP. Therefore novel therapeutic strategies focused on the protection of retina ganglion cells（RGCs） is urgent to develop. Brain derived neurotrophic factor（BDNF） is known to protect RGCs in vitro and in vivo. BDNF binds to two types of neurotrophin receptors, neurotrophin receptor p75 and tyrosine kinase receptor, trk B. Both receptors have been implicated in neurotrophin trafficking. Active transport along the axon is crucial to the neuron. Recent progress suggested that retrograde transport correlate with rapidly progressive neuronal cell death. Therefore, our study focused on the effective of BDNF in retrograde transport and axonal intact. Methods The rats were deeply anesthetized and a 29-G needle was injected into the anterior chamber with sterile saline, the IOP was elevated to 130 mm Hg for 45 min. BDNF/PBS solution（500 ng） was injected into the intravitreal after 6 hours of the ischemia-reperfusion. To investigate the impairment of the retrograde transport and the intact of axons, Di D and fluorogold（FG） were injected into the superior colliculus. The animals would sacrificed after 7 days. Optic nerves were embedded in epon resin. Semithin sections were stained with 1% Toluidine Blue. RGC axons were counted at 1 mm from the optic nerve head（anterior optic nerve） and 1 mm from optic chiasm（posterior optic nerve）. Non overlapped 24 squares were counted in each section. Results（1） In the ischemia-reperfusion group, the density of FG labeled RGCs decreased significantly compared with the control group after 3 weeks, 6 weeks and 9 weeks（59.83%, 44.91% and 22.76%, respectively）. We explore the hypothesis that a potential explanation for the initiation of motor neuron disease is an unappreciated vulnerability in central nervous system defense,the direct delivery of neurotoxins into motor neurons via peripheral nerve retrograde *** further suggests a mechanism for focal initiation of neuro-degenerative diseases in general,with subsequent spread by network degeneration as suggested by the Frost-Diamond *** propose this vulnerability may be a byproduct of vertebrate evolution in a benign aquatic environment,where external surfaces were not exposed to concentrated neurotoxins.

Despite serotonin’s and FMRF-amide’s wide distribution in the nervous system of invertebrates and their importance as neurotransmitters,the exact roles they play in neuronal networks leaves many *** mapped the presence of serotonin and FMRF-amide-immunoreactivity in the central nervous system and eyes of the pond snail Lymnaea stagnalis and interpreted the results in connection with our earlier findings on the central projections of different peripheral *** the chemical nature of the intercellular connections in the retina of *** is still largely unknown,we paid special attention to clarifying the role of serotonin and FMRF-amide in the visual system of this snail and compared our findings with those reported from other *** least one serotonin-and one FMRF-amidergic fibre were labeled in each optic nerve,and since no cell bodies in the eye showed immunoreactivity to these neurotransmitters,we believe that efferent fibres with somata located in the central ganglia branch at the base of the eye and probably release 5HT and FMRF-amide as *** labelling revealed retrograde transport of neurobiotin through the optic nerve,allowing us to conclude that the central pathways and serotonin-and FMRF-amide-immunoreactive cells and fibres have different locations in the CNS in *** chemical nature of the fibres,which connect the two eyes in ***,is neither serotoninergic nor FMRF-amidergic.
