O6-methylguanine DNA methyltransferase(MGMT), a DNA repair enzyme, has been reported in some congenital malformations, but it is less frequently reported in neural tube defects. This study investigated MGMT mRNA expression and methylation levels in the early embryo and in different embryonic stages, as well as the relationship between MGMT and neural tube defects. Spina bifida aperta was induced in rats by a single intragastric administration of all-trans retinoic acid on embryonic day(E) 10, whereas normal control rats received the same amount of olive oil on the same embryonic day. DNA damage was assessed by detecting γ-H2 A.X in spina bifida aperta rats. Real time-polymerase chain reaction was used to examine mRNA expression of MGMT in normal control and spina bifida aperta rats. In normal controls, the MGMT mRNA expression decreased with increasing embryonic days, and was remarkably reduced from E11 to E14, reaching a minimum at E18. In the spina bifida aperta model, γ-H2 A.X protein expression was increased, and mRNA expression of MGMT was markedly decreased on E14, E16, and E18. Bisulfite sequencing polymerase chain reaction for MGMT promoter methylation demonstrated that almost all CpG sites in the MGMT promoter remained unmethylated in both spina bifida aperta rats and normal controls, and there was no significant difference in methylation level between the two groups on either E14 or E18. Our results show that DNA damage occurs in spina bifida aperta rats. The mRNA expression of MGMT is downregulated, and this downregulation is independent of promoter DNA methylation.

AIM To explore the relationship between the configuration changes of the nuclear matrix-intermediate filament system in cancer cell induced by retinoic acid and the malignantphenotypic reversion of cancer *** The human gastric adenocarcinomacell line MGc80-3 cells were induced with 10-6mol/ L retinoic acid and subouItured at cover slipstrip and gold grids. The cells were treated byselective extraction methOd and prepared for whole mount electron microscopy *** samples were examined respectively withscanning and transmission electron *** The nuclear matrix filaments andintermediate filaments in MGC80-3 cells wererelatively few and scattered, not welldistributed and arranged irregularly. The nuclear lamina was ununiformly thick and compact,connected to the nuclear matrix filaments and intermediate filaments relaxedly. However, thetwo kinds of filaments were abundant and welldistributed, different in slender and thick form and interweaved into a regular network in the cells induced by 10-6 mol/ L RA. The nuclear matrix filaments and intermediate filaments were connected closely by the thin and compact fiber-Iike lamina, and interlaced into a regularnetwork throughout the whole cell *** The NM-IF system in MGc80-3cells had undergone a restorational changesimilar to those of normaI cells after RAinducement. This alternation is an importantmorpholOgical and functional expression to themalignant phenotypic reversion of cancer cells.

Treatment of the human esophageal cancer cell line EC8712 with retinoic acid (RA) stopped the cell growth significantly and gave rise to terminal differentiation of the cells characterized by increased expression of involucrin gene. Two cDNA libraries were constructed from the parental and RA-treated cells respectively. Repeated subtractive hybridization of single-stranded plasmid DNA prepared from pooled colonies of cDNA library of the parental cells with cDNA probe generated from the RA-treated cells exhausted sequences common to both libraries of the cell. The unhybridized cDNA probe represented, therefore, the genes activated after RA-treatment. By using these enriched cDNAs as probe to screen the cDNA library constructed from the RA-treated cells thirty-nine positive colonies were obtained, of which two were specifically due to RA-induction. One of these two cDNA clones, designated as pRA538, has undergone further analysis and shown differentiation-inducing effect on parental cancer cells. A novel strategy for cloning genes involved in terminal differentiation of cancer cells is developed.

retinoic acid (RA) plays an important role in development of vertebrate embryos. We demonstrate impacts of exogenous RA on the formation of neural crest cells in zebrafish using specific neural crest markers sox9b and crestin. Treatment with all-trans RA at 10-7 mmol/L at 50% epiboly induces sox9b expression in the forebrain and crestin expression in the forebrain and midbrain, resulting in significant increase of pigment cells in the head derived from the cranial neural crest. In addition, RA treatment induces expression of sox9b and crestin in the caudal marginal cells of the neuroectoderm during early segmentation. Earlier commitment of these cells to the neural crest fate in the posterior margins leads to abnormal development of the posterior body, probably by preventing mingling of ventral derived and dorsal-derived cells during the formation of the tailbud.

Objective To observe the prevention and therapeutic effects of tanshitone(TAN)on retinoic acid induced osteoporosis in *** The mice osteoporosis was induced by given retinoic acid intragasttrically for two *** histomorphological features of bone were observed and biochemical indexes in serum(Ca,P,ALP,TRAP,E2,BGP)were determined after mice were given TAN at the dose of 40,80,160 mg·kg-1 *** Tanshitone can induce high conversion of *** levels of P,ALP,TRAP and BGP in the TAN groups were lower than the model group,while the E2 level was higher than the model *** Tanshitone can prevent the loss bone in the experimental *** mechanism may be that it improves the level of estrogenic hormone and inhibits the high bone turnover.

Combination of all trans retinoic acid with butyric acid and its prodrugs markedly enhancing differentiation of human acute promyelocytic leukemia NB 4 cells

Objective To use NB 4, an authentic human acute promyelocytic leukemia cell line, as well as the marrow cells from patients with acute promyelocytic leukemia (APL), containing the PML/RARα fusion gene and fused protein to examine the growth inhibition and cytodifferentiation induced by all trans retinoic acid (ATRA), butyric acid (BA) and its prodrug tributyrin (TB) either as a single agent or in *** NB 4 and APL cells were cultured in presence of ATRA, BA and TB respectively either as a single agent or in combinations at various concentration ratio. Cell growth was measured and myeloid differentiation was determined by morphology and the percentage of positive nitroblue tetrazolium reduction (NBT) on consecutive days over the whole process of culture. Results NB 4 cells can be induced by ATRA alone and synergistically induced by the combinations of BA or TB with ATRA to differentiate. The synergy was reflected by a remarkable decrease in the effective concentration of ATRA required in the combinations in comparison with it as a sole agent. The combinations also shortened the time for the cells to reach the same level of maturation as that needed for ATRA alone. The potentiation on ATRA induced differentiation of NB 4 cells seemed depending on an appropriate concentration ratio of each inducer in the combinations and the time of action. A preliminary result of in vitro induction of primarily cultured leukemic cells from APL patients by the combined inducers was promising. Conclusion The combinations of ATRA with BA or TB at an appropriate ratio may improve the clinical outcome of differentiation therapy for APL patients.
