The pyramid representation of the functional network using resting-state fMRI

Background resting-state functional magnetic resonance imaging(RS-fmri)has been proved to be a useful tool to study the brain mechanism in the quest to probe the distinct pattern of inter-region interactions in the brain.As an important application of RSfmri,the graph-based approach characterizes the brain as a complex network.However,the network is susceptible to its scale that determines the trade-off between sensitivity and anatomical variability.Objective To balance sensitivity and anatomical variability,a pyramid representation of the functional network is proposed,which is composed of five individual networks reconstructed at multiple scales.Methods The pyramid representation of the functional network was applied to two groups of participants,including patients with Alzheimer’s disease(AD)and normal elderly(NC)individuals,as a demonstration.Features were extracted from the multi-scale networks andwere evaluated with their inter-group differences between AD andNC,aswell as the discriminative power in recognizing AD.Moreover,the proposed method was also validated by another dataset from people with autism.Results The different features reflect the highest sensitivity to distinguish AD at different scales.In addition,the combined features have higher accuracy than any single scale-based feature.These findings highlight the potential use ofmulti-scale features asmarkers of the disrupted topological organization in AD networks.Conclusion We believe that multi-scale metrics could provide a more comprehensive characterization of the functional network and thus provide a promising solution for representing the underlying functional mechanism in the human brain on a multi-scale basis.

Asthma is a serious health problem that involves not only the respiratory system but also the central nervous system.Previous studies identified either regional or network alterations in patients with asthma,but inconsistent results were obtained.A key question remains unclear:are the regional and neural network deficits related or are they two independent characteristics in asthma?Answering this question is the aim of this study.By collecting resting-state functional magnetic resonance imaging from 39 patients with asthma and 40 matched health controls,brain functional measures including regional activity(amplitude of low-frequency fluctuations)and neural network function(degree centrality(DC)and functional connectivity)were calculated to systematically characterize the functional alterations.Patients exhibited regional abnormities in the left angular gyrus,right precuneus,and inferior temporal gyrus within the default mode network.Network abnormalities involved both the sensorimotor network and visual network with key regions including the superior frontal gyrus and occipital lobes.Altered DC in the lingual gyrus was correlated with the degree of airway obstruction.This study elucidated different patterns of regional and network changes,thereby suggesting that the two parameters reflect different brain characteristics of asthma.These findings provide evidence for further understanding the potential cerebral alterations in the pathophysiology of asthma.

Regional homogeneity abnormalities in patients with tensiontype headache:a resting-state fMRI study

Tension-type headache（TTH） is the most prevalent type of primary headache. Many studies have shown that the pathogenesis of primary headache is associated with fine structural or functional changes. However, these studies were mainly based on migraine. The present study aimed to investigate whether TTH patients show functional disturbances compared with healthy subjects. We used restingstate functional magnetic resonance imaging（f MRI） and regional homogeneity（Re Ho） analysis to identify changes in the local synchronization of spontaneous activity in patients with TTH. Ten patients with TTH and 10 age-, gender-, and education-matched healthy controls participated in the study. After demographic and clinical characteristics were acquired, a 3.0-T MRI system was used to obtain restingstate f MRIs. Compared with healthy controls, the TTH group exhibited significantly lower Re Ho values in the bilateral caudate nucleus, the precuneus, the putamen, the left middle frontal gyrus, and the superior frontal gyrus. There was no correlation between mean Re Ho values in TTH patients and duration of TTH, number of attacks, duration of daily attacks, Visual Analogue Scale score, or Headache Impact Test-6 score. These results suggest that TTHpatients exhibit reduced synchronization of neuronal activity in multiple regions involved in the integration and processing of pain signals.

Epilepsy is a disease characterized by abnormal spontaneous activity in the brain.resting-state functional magnetic resonance imaging（RS-fmri） is a powerful technique for exploring this activity.With good spatial and temporal resolution,RS-fmri is a promising approach for accurate localization of the focus of seizure activity.Although simultaneous electroencephalogram-fmri has been performed with patients in the resting state,most studies focused on activation.This mini-review focuses on RS-fmri alone,including its computational methods and its application to epilepsy.

Theory and prospective risk factor studies indicate that retrained eaters characterized by less efficient response inhibition are vunerable for future onset of binge eating and bulimic pathology.Previous imaging studies investigating restrained eaters(REs) were based mainly on task-related functional Magnetic Resonance Imaging(fmri) and little is known about resting state eural activity underlying restrained eating.To illuminate this issue,we investigated resting state fmri differences between samples of REs(n=22) and unrestrained eaters(UREs)(n=30) using regional homogeneity(ReHo) analysis.Samples were equated on body mass index(BMI) and caloric deprivation levels(i.e.,14 hours±2.1 h since last evening meal before undergoing the fmri.Correlation analyses were performed between the ReHo index and response inhibition evaluated by stop signal reaction time(SSRT) within each sample.Compared with UREs,REs exhibited increased ReHo in brain regions associated with food reward(i.e.,orbitofrontal cortex (OFC),dorsallateral prefrontal cortex(dlPFC),attention(i.e.,lingual gyrus,cuneus,inferior parietal lobule) and somatosensory region dysfunction(i.e.,paracentral lobule,anterior insula). In addition,ReHo values for the left dlPFC were negatively correlated with SSRT while positive correlations were found between ReHo values in the left insula and SSRT.In concert with previous studies,these results suggest neural bases that explain why dieting to maintain or lose weight often fails or increases risk for binge eating among REs.

Background:Schizophrenia(SCZ)is a complex psychiatric disorder associated with widespread alterations in the subcortical brain structure.Hemispheric asymmetries are a fundamental organizational principle of the human brain and relate to human psychological and behavioral characteristics.We aimed to explore the state of thalamic lateralization of SCZ.Methods:We used voxel-based morphometry(VBM)analysis,whole-brain analysis of low-frequency fluctuations(ALFF),fractional amplitude of low-frequency fluctuations(fALFF),and resting-state seed-based functional connectivity(FC)analysis to investigate brain structural and functional deficits in SCZ.Also,we applied Pearson’’s correlation analysis to validate the correlation between Positive and Negative Symptom Scale(PANSS)scores and them.Results:Compared with healthy controls,SCZ showed increased gray matter volume(GMV)of the left thalamus(t=2.214,p=0.029),which positively correlated with general psychosis(r=0.423,p=0.010).SCZ also showed increased ALFF in the putamen,the caudate nucleus,the thalamus,fALFF in the nucleus accumbens(NAc),and the caudate nucleus,and decreased fALFF in the precuneus.The left thalamus showed significantly weaker resting-state FC with the amygdala and insula in SCZ.PANSS negative symptom scores were negatively correlated with the resting-state FC between the thalamus and the insula(r=-0.414,p=0.025).Conclusions:Collectively,these results suggest the possibility of aberrant laterality in the left thalamus and its FC with other related brain regions involved in the limbic system.

Selective Aberrant Functional–Structural Coupling of Multiscale Brain Networks in Subcortical Vascular Mild Cognitive Impairment

Subcortical vascular mild cognitive impairment(svMCI)is a common prodromal stage of vascular dementia.Although mounting evidence has suggested abnormalities in several single brain network metrics,few studies have explored the consistency between functional and structural connectivity networks in svMCI.Here,we constructed such networks using resting-state f MRI for functional connectivity and diffusion tensor imaging for structural connectivity in 30 patients with svMCI and 30 normal controls.The functional networks were then parcellated into topological modules,corresponding to several well-defined functional domains.The coupling between the functional and structural networks was finally estimated and compared at the multiscale network level(whole brain and modular level).We found no significant intergroup differences in the functional–structural coupling within the whole brain;however,there was significantly increased functional–structural coupling within the dorsal attention module and decreased functional–structural coupling within the ventral attention module in the svMCI group.In addition,the svMCI patients demonstrated decreased intramodular connectivity strength in the visual,somatomotor,and dorsal attention modules as well as decreased intermodular connectivity strength between several modules in the functional network,mainly linking the visual,somatomotor,dorsal attention,ventral attention,and frontoparietal control modules.There was no significant correlation between the altered module-level functional–structural coupling and cognitive performance in patients with svMCI.These findings demonstrate for the first time that svMCI is reflected in a selective aberrant topological organization in multiscale brain networks and may improve our understanding of the pathophysiological mechanisms underlying svMCI.

Background Although postpartum depression(PPD)and non-peripartum major depressive disorder(MDD)occurring within and outside the postpartum period share many clinical characteristics,whether PPD and MDD are the same or not remains controversial.Methods The current study was devoted to identify the shared and different neural circuits between PPD and MDD by resting-state functionalmagnetic resonance imaging data from 77 participants(22 first-episodic drug-naleMDD,26 drug-nale PPD,and 29 healthy controls(HC)).Results Both the PPD andMDD groups exhibited higher fractional amplitude of low-frequency fluctuation(fALFF)in left temporal pole relative to the HC group;the MDD group showed specifically increased degree centrality in the right cerebellum while PPD showed specifically decreased fALFF in the left supplementary motor area and posterior middle temporal gyrus(pMTG_L),and specifically decreased functional connectivities between pMTG and precuneus and between left subgeneual anterior cingulate cortex(sgACC_L)and right sgACC.Moreover,sgACC and left thalamus showed abnormal regional homogeneity of functional activities between any pair of HC,MDD,and PPD.Conclusions These results provide initial evidence that PPD and MDD have common and distinct neural circuits,which may facilitate understanding the neurophysiological basis and precision treatment for PPD.

Background:As one of the leading causes of global disability,major depressive disorder(MDD)places a noticeable burden on individuals and society.Despite the great expectation on finding accurate biomarkers and effective treatment targets of MDD,studies in applying functional magnetic resonance imaging(fmri)are still faced with challenges,including the representational ambiguity,small sample size,low statistical power,relatively high false positive rates,etc.Thus,reviewing studies with solid methodology may help achieve a consensus on the pathology of MDD.Methods:In this systematic review,we screened fmri studies on MDD through strict criteria to focus on reliable studies with sufficient sample size,adequate control of head motion,and a proper multiple comparison control strategy.Results:We found consistent evidence regarding the dysfunction within and among the default mode network(DMN),the frontoparietal network(FPN),and other brain regions.However,controversy remains,probably due to the heterogeneity of participants and data processing strategies.Conclusion:Future studies are recommended to apply a comprehensive set of neuro-behavioral measurements,consider the heterogeneity of MDD patients and other potentially confounding factors,apply surface-based neuroscientific network fmri approaches,and advance research transparency and open science by applying state-ofthe-art pipelines along with open data sharing.
