polymorphism of P66 in Borrelia Burgdorferi Strains in China

Objective To study the polymorphism in P66 and its human B-cell epitopes of Borrelia burgdorferi strains in China.Methods Polymerase chain reaction(PCR)and sequencing were used to obtain the P66 sequences of59 Chinese B.burgdorferi.Then the sequences were analyzed by MEGA 5.10 software and compared with the human B-cell epitope sequences from the Immune Epitope Database(IEDB)based on the reference strain of each genotype.Results Results showed that genetic and amino acid diversity presented in the 66 kD protein of all 59 Chinese strains,especially in Borrelia garinii(B.g)and Borrelia afzelii(B.a)strains.B.g strains were divided into three subclusters and two scattered strains JC1-7 and JC2-2 according to the amino acid sequences of P66.The P66 sequences of 15 Xinjiang strains represented by XI91-12 in the B.g subcluster1,changed from CAA to TAA codon at 508 aa position,resulting in early termination.Bases A and C were inserted at sequence position 1523 bp of strains FP1,LB20,LB21,and SZ21 in the B.a genotype,which resulted to early termination at position 511 aa.G base was inserted at 438 bp of LIP94-11 strain,which led to early termination at position 172 aa.Conclusion In P66 of 59 Chinese strains,polymorphisms were widely distributed.More importantly,the P66 amino acid sequences of B.g strains had a certain regional character.One of the characteristics of Xinjiang B.g isolates might be the variation at the 508 aa location in 15 Xinjiang B.g strains,which may be related to the strains’pathogenicity in this area.

polymorphism of OAS2 rs739901 C/A Involves the Susceptibility to EV71 Infection in Chinese Children

This study aimed to assess the relationship of OAS2 rs739901 5＇-flanking C/A polymorphisms with the susceptibility to Enterovirus-71 （EV71） infection. We investigated 294 hand-foot-mouth disease （HFMD） Chinese children with EV71 infection （165 mild cases and 129 encephalitis cases）. The improved multiplex ligation detection reaction （iMLDR） technique was used to test the genotypes. In EV71-infected patients, the CA genotype distribution （P=0.007）, A allele frequency （OR 1.32, 95% CI 1.0-1.7, P=0.034） and CA＋AA carriage frequency （P=0.003） of OAS2 rs739901 5＇-flanking were obviously elevated as compared with controls, but there were no statistically significant differences between mild cases and encephalitis cases. In EV71-infected patients, the counts of white blood cells （P=0.034） and blood glucose concentrations （P=0.042） were raised in A carriers （CA＋AA）. Among different genotypes of encephalitis cases, the contents of cerebrospinal fluid （CSF） showed no significant differences. IFN-7 levels in EV71-infected patients were higher than those in controls （mild group vs. control group, P〈0.01; encephalitis group vs. control group, P〈0.001）. In encephalitis cases, IFN-7 levels were reduced （P〈0.05） in A carriers compared to CC genotype, however, there were no significant differences between genotypes CA and AA （ P=0.226）. These findings suggest that OAS2 rs739901 5＇-flanking C/A genetic polymorphisms involve the susceptibility to EV71 infection, and A allele might be a risk factor of the susceptibility to EV-71 infection.

Genomic landscape of mitochondrial DNA insertions in 23 bat genomes:characteristics,loci,phylogeny,and polymorphism

The transfer of mitochondrial DNA to the nuclear genome gives rise to the nuclear DNA sequences of mitochon-drial origin(NUMTs),considered as a driving force in genome evolution.In this study,NUMTs in 23 bat genomes were investigated and compared systematically.The results showed that NUMTs existed in 22 genomes except for Noctilio leporinus,suggesting that mitochondrial fragment insertion in the nuclear genome was a common event in bat genomes.However,remarkable variations in NUMTs number,cumulative length,and proportion in the nu-clear genome were discovered across bat species.Further orthologous NUMT loci analysis of the Phyllostomidae family indicated that the NUMTs insertion events in bat genomes were homoplasy-free.The NUMTs were mainly inserted into the intergenic regions,particularly,co-localized with repetitive sequences(especially transposable el-ements).However,several NUMTs were inserted into genes,some of which were in the exon region of functional genes.One NUMT in the genome of Pteropus alecto surprisingly matched with cDNA of ATP8B3 that provided evidence of NUMTs with coding function.Phylogenic analysis on NUMTs originating from COXI and COXII loci highlighted 2 clusters of Yinpterochiroptera and Yangochiroptera for Chiroptera.Seven NUMTs from Rhinolophus ferrumequinum were ampliﬁed,and the sequencing results conﬁrmed the reliability of the NUMT analysis.One of them was polymorphic for the presence or absence of the NUMT insertion,and each genotype of NUMT loci showed a distinct regional distribution pattern.The information obtained in this study provides novel insights into the NUMT organization and features in bat genomes and establishes a basis for further studying of the evolution of bat species.

polymorphism of D-mannitol:Crystal structure and the crystal growth mechanism

The polymorphism of D-Mannitol(mannitol) is reviewed in this paper. It was found that the structure of the stable form is consistent in most literatures, but different authors have given different information about the two metastable forms. Therefore the commonly used nomenclature of mannitol was summarized based on the crystal unit cell parameters with the help of X-ray powder diffraction. Moreover, the crystal growth mechanism of mannitol polymorphs was summarized. Considering the lack of kinetic data for the metastable form especially, a reported method was attempted to apply to δ mannitol in an aqueous cooling crystallization process based on the induction time previously measured, and it was identified that the growth of the δ form follows the two-dimensional(2D) nucleationmediated mechanism. The results also indicate that the method based on induction time and supersaturation should have the potential to be expanded to the metastable polymorphs for the growth property study in a bulk system.

polymorphism dependent triplet-involved emissions of a pure organic luminogen

polymorphism has been frequently used in tuning the singlet emissions of pure organic dyes. The modulation of triplet-involved emissions, particularly room temperature phosphorescence(RTP),however, is scarcely reported. Herein, polymorphism is reported to tune the triplet-involved emissions of 2 CZBZL, a newly designed pure organic luminogen consisting of twisted benzil and two planar carbazole moieties. Other than the conventional modulation through changing molecular conformation and packing, vibration can also finely tune the triplet-involved emissions. Besides prompt fluorescence(PF),polymorph B with relatively extended conformation emits thermally activated delayed fluorescence(TADF), whereas the others(A, C–E) with similarly more twisted conformations generate predominant RTP or simultaneous DF and RTP. These results demonstrate the fascinating chance to regulate the tripletinvolved emissions through controlling conformation and vibration.

Precise Synthesis of Organic Heterostructures via the Synergy Approach of polymorphism and Cocrystal Engineering for Optical Applications

Organic heterostructures that can integrate the physiochemical properties of two or more substances have atracted widespread attention in the field of optoelectronics.However,the epitaxial growth process between different organic material molecules is unpredictable,and the precise synthesis of organic heterostructures is stll particularly challenging.Herein,through the synergy approach of polymorphism and cocrystal engineering,a series of organic branched heterostructures have been successfully prepared.Interestingly,by simply adjusting the solvent ratio,different crystalline phases of perylene microplates can be epitaxilly grown on the perylene-OFN cocrystal microwires,which provides a feasible approach for the preparation of organic heterostructures.Meanwhile,these as-prepared organic branched heterostructures can realize multi-channel and multi-color emission,and act as optical logic gates,which promotes the multifunctional integrated optoelectronics.

CTLA-4+49 A/G polymorphism and the Risk of Lung Cancer:a Meta-analysis

Background and objective:Lung cancer is one of the malignant tumors.Gene mutations associated with cellular immune function and regulating the activation and proliferation of immune cells.Several publications have explored the relationship between cytotoxic T lymphocyte antigen-4(CTLA-4)+49 adenine(A)/guanine(G)polymorphism and susceptibility of lung cancer,but the results remain controversial.Thus,we performed this meta-analysis to derive a more comprehensive estimation of the relationship.Methods:All articles addressed lung cancer and polymorphisms of CTLA-4 were searched from the Pub Med,EMBASE databases published up to June 29,2019.Odds ratios(ORs)with 95%confidence intervals(CIs)were used to assess the strength of association.Publication bias of relevant studies was examined via Begg’s test and funnel plots.Results:The meta-analysis included 8 case-control studies covering 4,430 lung cancer patients and 5,198 healthy controls from September 2008 to April 2020.The overall eligible data indicated that CTLA-4+49 A/G polymorphisms did not correlate with the elevated lung cancer risk in all genetic comparison models(dominant model:OR=1.037,95%CI:0.925-1.161;recessive model:OR=0.968,95%CI:0.888-1.055;allele model:OR=0.992,95%CI:0.933-1.054;homozygous model:OR=0.980,95%CI:0.857-1.121;heterozygous model:OR=1.023,95%CI:0.906-1.154).In further stratified analyses,CTLA-4+49 A/G polymorphism was found to be significantly associated with susceptibility to NSCLC in these models(dominant model:OR=1.404,95%CI:1.074-1.836;allele model:OR=1.273,95%CI:1.034-1.565;homozygous model:OR=1.553,95%CI:1.044-2.310;heterozygous model:OR=1.308,95%CI:1.062-1.611).Conclusion:CTLA-4+49 A/G polymorphism were not associated with the risk of lung cancer but might be a risk factor only in NSCLC.

Chronic hepatitis B virus(HBV)infection is one of the major causes of hepatocellularcarcinoma(HCC),and the HBV X(HBx)gene plays a critical role in the molecular pathogenesis ofHBV-related HCC.We have investigated whether there are particular HBx gene mutations associated withHCC in patients from southern China.The HBx gene was examined in 51 paraffin-embedded tumor tissuesamples from patients with HCC and 25 serum samples from the HBV carrier by nested polymerase chainreaction(PCR),single-stranded conformational polymorphism and heteroduplex analysis.The HBx geneswith potentially important mutations from tumor tissue samples were cloned,sequenced and aligned withthe published HBx gene sequence.HBV genotypes in tumor tissue samples were analyzed by nested PCR.Analyses of HBx gene polymorphism showed that 31.3% of HBx gene fragments in tumor tissue sampleshad a special pattern.A common deletion at nt 382-400 of the HBx gene accompanied by 29 point mutationswas detected in four randomly selected tumor tissue samples with this pattern which caused a frame-shiftin the HBx open reading frame with a new stop codon at nt 1818,resulting in an HBx polypeptide chaintruncated at the C end in these cases.Among the four randomly selected samples,three were HBV genotypeB,and one was not detected by our present assay.In another tumor tissue sample,amplification of thefull-length HBx gene yielded a shorter fragment.Sequencing of this fragment revealed a 264 bp deletionbetween nt 1577 and 1840 of the HBV gene.These results suggest that HBx gene mutation occurs frequentlyin HCC samples,and the deletion at nt 382-400 of the HBx gene might play a role in carcinogenesis of HCCin southern China.

Intraspecific DNA methylation polymorphism in the non-edible oilseed plant castor bean

Investigation of the relationships of phenotypic and epigenetic variations might be a good way to dissect the genetic or molecular basis of phenotypic variation and plasticity in plants. Castor bean(Ricinus communis L.), an important non-edible oilseed crop, is a mono-species genus plant in the family Euphorbiaceae. Since it displays rich phenotypic variations with low genetic diversity, castor bean is a good model to investigate the molecular basis of phenotypic and epigenetic variations. Cytosine DNA methylation represents a major molecular mechanism of epigenetic occurrence. In this study, epigenetic diversity of sixty landrace accessions collected worldwide was investigated using the methylationsensitive amplification polymorphism(MSAP) technique. Results showed that the epigenetic diversity(based on the polymorphism of DNA methylated loci) exhibited a medium variation(Ne=1.395,He=0.242,I=0.366) at the population level though the variation was great, ranging from 3.80% to 34.31% among accessions. Both population structure analysis and the phylogenetic construction(using the neighbor-joining criteria) revealed that the two main clades were identified, but they did not display a distinct geographic structure. After inspecting the location of polymorphic methylated loci on genome we identified that the polymorphic methylated loci occur widely in nuclear and organelle genomes. This study provides new data to understand phenotypic and epigenetic variations in castor bean.

Impact of CYP3A4*1G polymorphism on Fentanyl Analgesia Assessed by Analgesia Nociception Index in Chinese Patients Undergoing Hysteroscopy

Background:The clinical efficacy of fentanyl for pain control differs greatly across individuals.The purpose of this study was to investigate the impact of CYP3A4*1G polymorphism including wild-type homozygote (CYP3A4*1/*1,GG),mutant heterozygote (CYP3A4*1/*1G,GA),and mutant homozygote (CYP3A4*1G/*1G,AA)on fentanyl analgesia in Chinese patients undergoing hysteroscopy by the assessment of analgesia nociception index (ANI). Methods:A total of 200 gynecologic patients scheduled for elective hysteroscopy under general anesthesia at Peking University People's Hospital from May to December in 2017 were enrolled in this study.Venous blood was withdrawn for genotyping of CYP3A4*1G before operation.Fentanyl 1μg/kg was administered preoperatively followed by target-controlled infusion of propofol for induction and maintenance.Intraoperative analgesic efficacy of fentanyl was assessed by ANI monitoring at T0(entering room),T1(cervical dilation), T2(start of cervical aspiration),and T3(end of cervical aspiration)time points.The duration of propofol infusion and total dosage of propofol were recorded as well. Results:The patients were divided into three groups according to CYP3A4*1G polymorphism,including 143 in GG group,47in GA group, and 10 in AA group.There was no significant difference in clinical demographics among three groups.The frequency of CYP3A4*1G variant alleles accounted for 16.8%and the distribution of variant alleles was consistent with Hardy-Weinberg equilibrium.Using a multilevel model,ANI values at T1(63.81±19.61),T2(63.63±17.82),and T3(65.68±17.79)were significantly lower than that at T0 (77.16±12.93)in the study population (F =23.50,P polymorphism associated with the analgesic efficacy of intraoperative fentanyl in the patients undergoing hysteroscopy under general anesth
