Considerable data have shown that plasminogen activator (PA) may play an important role in the mechanism of ovulation. A recent report argued that PA is not a primary proteoIytic enzyme for follicular rupture because a dose of indomethacin that blocked ovulation did not inhibit ovarian PA content. To further clarify the specific role of PA in the ovulation, we have carefully examined the effect of indomethacin on the secretion of PA in granulosa and theca-interstitial (TI) cells following hCG or GnRH agonist administration in PMSG-primed hypophysectomized immature rats. We have also studies in vitro action of this compound on PA secretion in granulosa cells. The results indicate that indomethacin can only suppress hCG- and GnRH agonist-induced PA secretion, but not suppress the ovarian content of these enzymes.

Objective To compare the efficacy of low dose recombinant tissue-type plasminogen activator (rt-PA) thrombolysis with primary coronary stenting after acute myocardial *** Of 261 patients with first acute myocardial infarction, 131 were given low dose rt-PA intravenous thrombolysis, and 130 primary coronary *** The age, time from onset of chest pain to hospital presentation and infarct location between these two groups were comparable. The patency rate of the infarct-related artery (IRA) in patients in the thrombolysis group was significantly lower than that of patients in the primary stenting group (P0.05).Conclusion Comparing with low dose rt-PA thrombolytic therapy after acute myocardial infarction, primary coronary stenting has a higher patency rate of the IRA, better cardiac function and shorter hospitalization time.

Effects of forskolin on progesterone and plasminogen activator production in pseudopregnant ratcorpora lutea was investigated using isolated in vitro perfused *** andplasminogen activator production were measured on day 1,8 and 18 of *** results indicated:different concentrations of forskolin(100,200,400 and800 μg)administered to ovaries on the 8th day of pseudopregnancy caused elevation of progesteronesecretion in a dose-dependent *** 8 hours of perfusion,PA contents increasedsignificantly in ovaries treated with *** exogenous PA-urokinase(800 U)added to theperfusion solution,progesterone secretion increased significantly as compared to control group andremained on high level throughout the perfusion *** exerting no apparent effects in lowdosage(5 mM),AMCHA,a PA inhibitor,administered in higher dosage(10 and 15 mM)led tomarked reduction in PA activity and progesterone secretion as compared to control *** causes significant elevation of level of progesterone secretion and PA activity inpseudopregnant rat ovary perfused in *** PA seems to regulate progesterone secretion in theperfused rat corpora luteum.

[Objective]To investigate the thrombolytic and antiplatelet effects of a novel plasminogen activator from the venom of Gloydius brevicaudusviper（GBV-PA）in vitro and in vivo.[Methods]Thrombolytic experiments were performed in rabbit models of ear vein thrombosis and carotid artery thrombosis,and in dog model of acute cerebral *** of thrombus formation was evaluated in rat inferior vena cava thrombosis model and ferric chloride-induced arterial *** vitro,we assayed the antithrombotic effect of GBV-PA on rabbit blood clots,euglobulin lysis time（ELT）of rabbit plasma,and on ADP-induced platelet aggregation.[Results]GBV-PA intravenous administration significantly reduced vascular recanalization times of rabbit ear veins thrombosis and thrombus weight of rabbit carotid artery *** arterial recanalization rates were doseand time-dependently improved after administration of GBV-PA in canine acute cerebral infarction *** length and weight was significantly reduced by GBV-PA both in rat inferior vena cava and ferric chloride-induced arterial thrombosis *** formation of blood of rabbits they were administrated with GBV-PA was also ***-PA radically reduced plasma ELT of rabbit’s blood ***-induced platelet aggregation was inhibited by GBV-PA in a dose-dependent manner with a half-maximal inhibitory concentration of 19.9 μg/mL.[Conclusion]This study demonstrates that GBV-PA is a thrombolytic and antiplatelet *** has significant antithrombotic effects on various in vitro and in vivo experimental models of *** mechanisms that underline its antithrombotic effects were related to GBV-PA’s capabilities of increasing fibrinolytic activities and inhibition of platelet aggregation.

Tumor invasion and metastasis is a highly complicated, multi-step phenomenon. In the complex event of tumor progression, tumor cells interact with basement membrane and extracellular matrix components. Proteolytic enzymes(proteinases) are involved in the degradation of extracellular matrix, but also in cancer invasion and metastasis. The four categories of proteinases(cysteine-, serine-, aspartic-, and metalloproteinases) are named and classified according to the essential catalytic component in their active site. We and others have shown that proteolytic enzymes play a major role not only in colorectal cancer(CRC) invasion and metastasis, but also in malignant transformation of precancerous lesions into cancer. Tissue and serumplasma antigen concentrations of proteinases might be of great value in identifying patients with poor prognosis in CRC. Our results, in concordance with othersindicate the potential tumor marker impact of proteinases for the early diagnosis of CRC. In addition, proteinases may also serve as potential target molecules for therapeutic agents.

We have demonstrated that prolactin inhibits gonadotropin-induced ovulation in PMSG-primed *** number of ova in oviducts considerably decreased in the group of hCG plus prolactin (PRL)(19.7 + 4.9) as compared with that of hCG alone (31.7 + 6.7). PRL inhibition of hCG-inducedovulation in mice may be through decreasing the ovarian plasminogen activator (PA) activity on onehand, and inhibiting the preovulatory increase in estrogen (E) secretion on the other.

Objective To construct a new kind of recombinant vector containing human tissue-type plasminogen activator(t-PA) cDNA for studing the feasibility of enhancing fibrinolytic activity by transplantation of genetic engineering cells. Methods We recombinated human tPA cDNA with expression vector pcDNA3. 1 (+) by using the method of molecular cloning, sequenced the plasmid DNA, and cut the plasmid by using enzymes. Results The plasmid pcDNA3.1(+)tPA was divided into 5.4Kb and 2.0Kb segments respectively by using Kpn 1 and Xba I, into 78bp, 414bp, 622bp, 2.0Kb, and4.2Kb segments respectively by tsing Pst l, into 472bp and 6.9Kb segments respectively by using EcoR I. Sequencing result showed that it is the whole human tPA cDNA. Conclusion The new kind of recombinant expression vector could serve as new tools and methods for preventing thrombogenic diseases.
