Genome constitution for Musa beccarii (Musaceae) varieties

Musa beccarii *** var. beccarii and Musa beccarii *** var. hottana H?kkinen were described earlier in Acta Phytotaxonomica et Geobotanica based on their morphological characteristics. In order to distinguish the genomes between the two M. beccarii varieties and M. coccinea Andrews, we have now studied them with the Inter- Retrotransposon Amplified Polymorphism (IRAP) marker analyses. The high levels of IRAP polymorphism detected in this study showed that M. beccarii var. beccarii and M. beccarii var. hottana are two distinct varieties. Additional IRAP bands found in M. beccarii var. beccarii indicated that M. beccarii var. hottana might be evolutionarily more ancient than M. beccarii var. beccarii because LTR retrotransposons are not transpositionally removed.

Genome-wide analysis.of Plant-specific Dof Transcription Factor Family in Tomato

The Dof （DNA binding with One Finger） family encoding single zinc finger proteins has been known as a family of plant-specific transcription factors. These transcription factors are involved in a variety of functions of importance for different biological processes in plants. In the current study, we identified 34 Dof family genes in tomato （Solanum lycopersicum L.）, distributed on 11 chromosomes. A complete overview of SlDof genes in tomato is presented, including the gene structures, chromosome locations, phylogeny, protein motifs and evolution pattern. phylogenetic analysis.of 34 SlDof proteins resulted in four classes constituting six clusters. In addition, a comparative analysis.between these genes in tomato, Arabidopsis （Arabidopsis thaliana L.） and rice （Oryza sativa L.） was also performed. The tomato Dof family expansion has been dated to recent duplication events, and segmental duplication is predominant for the SlDof genes. Furthermore, the SlDof genes displayed differential expression either in their transcript abundance or in their expression patterns under normal growth conditions. This is the first step towards genome-wide analyses of the Dof genes in tomato. Our study provides a very useful reference for cloning and functional analysis.of the members of this gene family in tomato and other species.

New ancient DNA sequences suggest high genetic diversity for the woolly mammoth (Mammuthus primigenius)

Partial DNA sequences of cytochrome b gene (mtDNA) were successfully retrieved from Late Pleistocene fossil bone of Mammuthus primigenius collected from the Xiguitu County ( Yakeshi), Inner Mongolia Autonomous Region and from Zhaodong, Harbin of Heilongjiang Province in northern China. Two ancient DNA fragments (109 bp and 124 bp) were authenticated by reproducible experiments in two different laboratories and by phylogenetic analysis.with other Elephantidae taxa. phylogenetic analysis.using these sequences and published data in either separate or combined datasets indicate unstable relationship among the woolly mammoth and the two living elephants, Elephas and Loxodonta . In addition to the short sequences used to attempt the long independent evolution of Elephantidae terminal taxa, we suggest that a high intra-specific diversity existed in Mammuthus primigenius crossing both spatial and temporal ranges, resulting in a complex and divergent genetic background for DNA sequences so far recovered. The high genetic diversity in the extinct woolly mammoth can explain the apparent instability of Elephantidae taxa on the molecular phylogenetic trees and can reconcile the apparent paradox regarding the unresolved Elephantidae trichotomy.

Identification and Expression of Floral Organ Homeotic Genes from Alpinia oblongifolia (Zingiberaceae)

Current understanding of the classical ABC model of floral development has provided a new set of characters to evaluate floral evolution. However, what is still lacking is a clear assessment of this genetic program across monocots. Here, to investigate the evolution of members of class A and B genes in monocots, we report the sequence characteristic and transcript expression of three new MADS-box genes in Alpinia oblongifolia Hayata. Sequence and phylogenetic analysis.reveals that these genes are FUL-like and AP3-1ike. Therefore, they were termed AoFL1, AoFL2 and AoAP3. AoFL1 contains the FUL motif, but AoFL2 lacks this motif. Their expression revealed by in situ hybridization may reflect the ancestral function of FUL-like genes in the specification of inflorescence and floral meristems. The AoAP3 gene contains two conserved motifs, the PI-derived and paleoAP3 motifs. The AoAP3 transcripts located to the corolla and stamen, and hybridization signals were detected in the central whorl. These expression patterns suggest that the functions of homologous organ identity genes are diversified in A. oblongifolia. The implications of these findings on the conservation of homologous gene function are discussed.
