Synergistic action on hypnosia: Yinao capsules with pentobarbital sodium of threshold and sub-threshold dosages

BACKGROUND： Sedative and hypnotic drugs could cure insomnia in a dependent manner, and traditional Chinese medicine has some superiority in treating insomnia. OBJECTIVE： To observe the synergistic action of yinao capsules with pentobarbital sodium in threshold and sub-threshold dosages in hypnosia and sedation. DESIGN： A completely randomized grouping design and control experiment. SETTING： Pharmacological laboratory, College of Pharmacy, Henan College of Traditional Chinese Medicine. MATERIALS： Totally 200 grade II Kunming mice of 18 - 21 g, either male or female, were used. Yinao capsules, main ingredients of which were turtleback glue, thinleaf milkwort root, Chinese magnoliavine fruit, mythic fungus, tangshen, ginseng and grassleaf sweetflag rhizome, were offered by Chinese-American Huayi Pharmacy, Co.,Ltd. （ratified number： 040901）; Kangnaoshuai capsules, main ingredients of which were prepared rehmannia root, tuber fleeceflower root, ginseng, membranous milkvetch root, thinleaf milkwort root, Fushen, grassleaf sweetflag rhizome, spine date seed, lecithin, barbary wolfberry fruit, pueraria root, vitamin E, etc., were produced by Shijiazhuang Siyao, Co.,Ltd. （ratified number： 040964）; pentobarbital sodium was produced by China Medicine （Group） Shanghai Chemical Reagent, Co,.Ltd. （Ratified number： 030816）. ZZ-6 mice spontaneous activity apparatus was produced by Chengdu Taimeng Science and Technology, Co.,Ltd. METHODS： The experiment was carried out in the Animal Experimental Center, Henan College of Traditional Chinese Medicine from October to December in 2005. （1） Influence of Yinao capsules on the spontaneous activity of mice： Fifty mice were randomly divided into five groups with 10 mice in each group： Mice in the large, middle and small dosages of Yinao capsules groups were intragastrically infused with Yinao capsules suspension （1.36, 0.68, 0.34 g/kg）; Those in the Kangnaoshuai capsules group were infused with Kangnaoshuai suspension （1.12 g/kg）; Those in the control group were given physiologic saline o

Effect of different anesthesia methods on erythrocyte immune function in mice

Objective:To explore effect of different anesthesia methods and different anesthetics on erythrocyte immune function in mice.Methods:The mice were anesthetized by isoflurane and ether inhalation,and also under intraperitoneal anesthesia with sodium pentobarbital and chloral hydrate,Blood was collected from the ventro-cardinal vein.Automatic blood cell analyzer was used for routine blood examination,and the canthine oxidase method was used to measure the superoxide dismutase(SOD)activity.Lipid peroxidation product malondialdehyde(MDA)was measured with TBA,and glutathione peroxidase(GSH-Px)was measured with DTNB,and then the effect of different anesthesia methods and different anesthetics on erythrocyte immune function in mice was observed.Result:Hct level of chloral hydrate intraperitoneal injection group was significantly higher than the other three groups(Ppentobarbital sodium group were significantly higher than the other three groups(Ppentobarbital intraperitoneal injection group were significantly lower than the other two groups;RBC-C 3bRR and RBC-ICR of the chloral hydrate and sodium pentobarbital intraperitoneal injection group were significantly lower than the other two groups.Conclusions:Different drugs can induce changes in immune function of mice at different levels.Isoflurane and ether have less damage to animal body,while chloral hydrate and sodium pentobarbital intraperitoneal injection have a certain inhibitory effect on the animal body respiratory system and can cause greater damage to the body.Therefore,the reasonable selection and control of anesthetics are very important in order to avoid the experimental errors caused by anesthesia.

Background Anesthetics such as pentobarbital are generally applied before sacrificing animals,but their effect on stress responses has not been elucidated.The present study aimed at revealing the effect of intraperitoneal（ip）pentobarbital injection on the stress responses in rat before decapitation.Methods 69 Sprague Dawley male rats were randomly divided into 7 groups:chronic unpredictable stresses（CUS） rats sacrificed with direct decapitation（CUSD,n=10）,CUS rats sacrificed following ip injection of pentobarbital before decapitation（CUSP,n=10）,acute electric foot shock（FS） rats sacrificed with direct decapitation（FSD,n=10）;FS rats sacrificed following ip injection of pentobarbital before decapitation（FSP,n=10）,control rats sacrificed with direct decapitation（CTRD,n=10）,control rats sacrificed following ip injection of pentobarbital before decapitation（CTRP,n=10）,and control rats sacrificed with ip injection of saline before decapitation（CTRS,n=9）.Plasma concentrations of corticosterone（CORT）,testosterone（T）and estradiol（E2） and hypomalamic mRNA levels of corticotrophin-releasing hormone（CRH）,vasopressin（AVP） and oxytocin（OXT）,all of which are stress-related molecules,were measured.Results CUSD and FSD groups both showed significantly increased plasma CORT,T and E2 levels compared with the CTRD group（P ＜ 0.01） while these significant changes disappeared in CUSP or FSP rats compared with the CTRP group.In the hypothalamus,differences between CUSD or FSD group and the CTRD group in the CRH-,AVP- or OXT-mRNA expression did not change significantly following ip injection of pentobarbital before decapitation.In addition,ip injection of saline showed similar effects as ip injection of pentobarbital.Conclusion ip injection of pentobarbital before decapitation may significantly affect the chronic or acute stress responses,especially for plasma parameters,which seems to be caused by the injection itself,rather than by pentobarbital.

OBJECTIVE Our previous studies indicated that L-type calcium channel blocker diltiazem could potentiate pentobarbital-induced hypnosis through serotonergic system.In view of the important role of dorsal raphe nucleus(DRN) on the sleep regulation and the pharmacological actions of calcium channel blocker,we presumed that Ca in the DRN may play an important role in sleep regulation in pentobarbital treated rats.Therefore,we investigated whether the Ca modulation in DRN would affect the sleep parameters in pentobarbital treated rats.METHODS L-type Ca channel antagonist diltiazem,agonist BAY-K-8644,Ca chelator EGTA and CaCl was microinjected into the DRN and the sleep parameters was detective by electroencephalogram(EEG).RESULTS Perfusion of the agents attenuating Ca function,such as diltiazem(5 or 20 nmol) or EGTA(3 or 6 pmol) into DRN significantly increased pentobarbital(35 mg·kg,i. p.)-induced total sleep(TS),non-rapid eye movement(NREM) sleep and the slow wave sleep(SWS) ratio in NREM sleep.On the contrary, the DRN injection of the agents improving Ca function,such as BAYk-K-8644(10 nmol) or CaCl(50 or 100 nmol) significantly reduced pentobarbital(35 mg·kg,i.p.)-induced TS,NREM sleep,rapid eye movement(REM) sleep and REM sleep ratio in TS without influence on SWS.CONCLUSION These results suggested that the suppression of Ca function in DRN could increase NREM sleep including SWS,and the elevation of Ca function could reduce both NREM and REM sleep in pentobarbital treated rats.

腓肠肌内注射HRP后,用生物化学法测定坐骨神经、L;节段背根和腹根神经的HRP含量。在戊巴比妥连续全身麻醉大鼠的HRP含量明显低于不麻醉的大鼠,而肌肉不活动（TTX中毒和切腱）大鼠神经组织中的HRP含量无甚变化。刺激神经不能改变麻醉大鼠的HRP含量。上述结果提示:除麻醉剂造成的肌肉不活动因素外,戊巴比妥对大鼠骨胳肌HRP的轴突摄取和逆行传送具有另外的抑制作用。已有研究报道:破伤风和单纯性疱疹病毒脑炎都是由于它们的毒素或病毒,通过外周神经摄取然后逆行传送到各级中枢而致病的。
