By using anti-ras p21 mouse monoclonal antibody(McAb),SCI-oncogema 1,the au-thors examined immunohistochemical staining in pancreatic *** percentage ofpositive staining was 55.8%(24/43)and was related to either histopathological grade or *** statistical analysis of the correlation between the positive staining of anti-ras p21and patient prognosis with Klaplan-Meier curve and Log-rant test,the positive staining casesshowed comparatively better prognosis than the negative ones,suggesting that ras p21 expres-sion may be important in the early stage of pancreatic carcinoma.

OBJECTIVE To investigate the status of DNA methylation in gastric *** We analysed methylation pattern of c-myc, c-Ha-ras oncogenes by southern blot hybridization. DNA from cancerous, paracancerous and non-cancerous area of surgically resected samples in 21 cases of advanced gastric cancer were digested withMspI/HpaII and hybridized with the two genomic32P labelled probes. In addition, the level of total genomic DNA methylation was measured by incubating DNA with3H-S-Adenosylmethionine (3H-SAM) in the presence of a methylase which methylates all the cytosine residues that are in the double CpG (cytosine-guanine).RESULTS The results indicated that both c-myc and c-Ha-ras oncogene fragments containing CCGG sequence were hypomethylated in DNA samples from cancerous (10/21 and 5/10) and paracancerous (13/21 and 4/10) areas. Moreover, the level of total genomic DNA methylation in cancerous tissue was significantly lower than that in non-cancerous and paracancerous mucosa tissues (Poncogenes fragments containing CCGG sequence was abnormal in gastric mucosa tissue from gastric cancerous and paracancerous areas.

Embryonic stem cells (ES-5) were transfected with PSV 2-neo-EJ-Ras and the cell colonies were chosen in the selection medium containing Geneticin. Southern-blot analysis showed that all the transfected cells possessed the whole EJ-Ras DNA. Northern-blot analysis demonstrated that the selected clones express higher level of Ras mRNA than the control. When the transfected ES-5 cells were injected into the flanks of nude mice, they formed bigger tumors and more stem cell nests were found. The chimeric ability of the transfected ES-5 cells decreased when the cells were injected into the blastocysts which were then transferred back into the uterine of foster mother. We propose that the over-expression of EJ-Ras in ES-5 cells increase the proliferation ability but decreases the chimeric potential of cell.
