A simple and sensitive gas chromatography(GC)-based method was developed for the analysis of monosaccharide composition of purified polysaccharides in Ganoderma atrum (***).The polysaccharide was firstly hydrolyzed to give the constituent monosaccharides, which were subsequently derived into acetylated *** separation of the derivatized monosaccharides was efficiently carried out by GC,using a capillary column packed with a (5%phenyl)methylpolysiloxane stationary *** concentrations of monosaccharides were determined based on the measured peak area *** stable derivatives of the most common monosaccharides of these glycoconjugates(Ara,Rha,Xyl,Fuc,Gal,Man,GIc,and Gala)could be separated and quantitatively and reproducibly determined with a high degree of sensitivity level in the presence of acetyl inositol as the inner *** method fulfilled the established criteria for *** limits of detection(LOD)and quantification(LOQ)were down to 0.013 mg/mL and 0.043 mg/mL *** intermediary precision values(expressed as RSD) were less than 10%.The mean recovery of the method was 100+3%,with the RSD values of less than 5%.In order to prove the validity of the above method,high performance liquid chromatography(HPLC)was also used to compare with *** determined concentrations of monosaccharides obtained from the two chromatographic methods were found close to each other within acceptable error *** study demonstrated that the developed method could normally be recommended for oligosaccharide analysis,and could also be used in the reproducible quantification of monosaccharides in biological and biochemical laboratories,where HPLC chromatography or MS method may not be readily available.

高效液相色谱法测定四种植物多糖中单糖方法的建立

采用高效液相色谱法测定四种植物多糖的单糖组成。用超声辅助提取大豆、香菇、木耳、枸杞多糖，通过三氟乙酸水解后，用1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生–液相色谱法检测，并选用专属性，精密度，灵敏度，线性范围和加标回收率对此方法进行验证，最后对大豆、香菇、木耳、枸杞多糖中单糖的组成进行分析。结果表明，待测定的各单糖组分在1.03~550 μg/mL范围内呈良好的线性关系，且各组分分离度大于1.5，检出限大于2.01 μg/mL，定量限大于6.43 μg/mL，精密度与稳定性的相对标准偏差均小于4%，加标回收率在98.0%~101.4%之间，验证结果显示该方法简单快速、准确可靠。通过应用此方法测定大豆、香菇、木耳、枸杞多糖的单糖组成，结果显示大豆多糖属于以半乳糖为主的杂多糖，且半乳糖含量为53.40%；香菇和枸杞多糖属于以葡萄糖为主的杂多糖，所含的葡萄糖含量分别为45.74%和21.07%；木耳多糖属于以D-甘露糖为主的杂多糖，且D-甘露糖含量为18.64%。

COMPARISON OF POLYSACCHARIDES ISOLATED FROM THE MYCELIA OF A CULTIVATED STRAIN OF PORIA COCOS GROWN IN DIFFERENT LIQUID CULTURE MEDIA

Mycelium of a cultivated strain of Poria cocos was grown by submerged fermentation in a liquid mediumcontaining corn steep liquor with orbital shaking. Six polysaccharides coded as ac-PCM1, ac-PCM2, ac-PCM3-Ⅰ andⅡ, ac-PCM4-Ⅰand Ⅱ were isolated from the myelium by extracting with 0.9% NaCl aqueous solution, hot water, 0.5 mol/L NaOHaqueous solution and 88% formic acid. Exo-polysaccharide was obtained from the culture medium and coded as *** monosaccharide composition and molecular weights of these polysaccharides were characterized by using infraredspectroscopy, gas chromaography, elemental analysis, ^(13)C-NMR, viscometry and light scattering. The results indicated thatac-PCM0, ac-PCM1 and ac-PCM2 are heteropolysaccharides containing glucose, galactose, mannose and fucose, and ac-PCM3-Ⅰ and ac-PCM3-Ⅱ mainly consist of D-glucose. The content of the glucose in the polysaccharides increased with theisolation progress. Remarkably, α-glucan and β-glucan coexisted in the extract by NaOH aqueous solution (ac-PCM3), andcould be separated by chemical methods. The protein in the ac-PCM polysaccharides cultured from the medium containingcorn steep liquor was higher than that in the ab-PCM from the medium with bran extract. Therefore, the polysaccharidesfrom Poria cocos mycelia cultured in different media have different chemical composition, molecular weights and conformations.

Anti-hyperglycemic effect of the polysaccharide fraction isolated from mactra veneriformis

Total macromolecule extract was obtained from the soft body of Mactra veneriformis by the coupling techniques of decoction and alcohol *** extract was deproteinized with an ion exchange column,and resulted in the purifying of the crude polysaccharide *** was found by chemical analysis that the crude polysaccharide part is composed of abundant polysacchar-ides(>95%)and few proteins(monosaccharide,glucose,was detected from its hydrolytes by thin-layer chromatography,indicating that the polysaccharides might be analogs of *** anti-hyperglycemia effects of the crude polysaccharide part were preliminarily investigated using several pharmacolo-gical methods in normal and diabetic *** experimental results showed that the crude polysaccharide fraction exhibited proper glycemia inhibition activity,and 300 mg/kg-weight dose has the optimal effect among all the studied *** is concluded that the crude polysaccharide fraction can be explored as a novel health product that possesses potential as an anti-hyperglycemic agent.

In this paper,a water-soluble polysaccharide named as CPC-1 was isolated from the hot water extract of Cyclocarya paliurus(Batal.)Iljinskaja,which is a well-known native medicinal plant in china that used against various types of illnesses related to the hyperlipoidemia, hypertensive and immune *** process of isolating and purifying the water-soluble polysaccharide from Cyclocarya paliurus(Batal.)Iljinskaja,was achieved through hot water extraction,filtration,ethanol precipitation,deproteination,decolourization and a combination of anion-exchange chromatography on DEAE-Sephadex A-25 and gel permeation chromatography over Sephacryl *** homogeneity of the polysaccharide CPC-1 was proved by gel filtration chromatogrophy on Sepharose CL-6B,with a molecular weight of 1.2×106 Da determined by high-performance gel permeation chromatography(HPGPC).The analysis of monosaccharide composition in the polysaccharide CPC-1 from Cyclocarya paliurus(Batal.)Iljinskaja by gas chromatography revealed that it was composed of L-rhamnose,L-arabinose,D-xylose, D-mannose,D-glucose and D-galactose in the molar ratios of 10.64:31.23:3.23:8.72:30.16: *** characterization of the polysaccharide CPC-1 were also elucidated.

Ultrasonic-assisted technique was employed to extract the crude polysaccharide（TMP） from Tricholoma matsutake fruiting ***-factor test and orthogonal experimental design（L9（3）3） were investigated to obtain the optimal extraction *** showed that the optimal parameters wereas follows:ultrasonic temperature 40℃,ultrasonic time 50 min and water to raw material ratio 25 ml/*** novel polysaccharide fractions（TMP30,TMP60 and TMP80） were isolated and purified from TMP by stepwise alcohol *** monosaccharide compositions and percentage were determined by HPAEC-PAD and FT-IR ***,their in vitro antioxidant activities were investigated in terms of reducing power assay and scavenging rate of 2,2-diphenyl-1-picrylhydrazyl（DPPH） radicals and hydroxyl *** order of various fractions on antioxidant activities was TMP80 ＞ TMP ＞ TMP60 ＞ *** findings suggested that the novel polysaccharide fractions from ***,especially TMP80,could be promising active macromolecules for biomedical use.
