methyl-β-cyclodextrin enhanced biodegradation of polycyclic aromatic hydrocarbons and associated microbial activity in contaminated soil

The contamination of soils by polycyclic aromatic hydrocarbons （PAHs） is a widespread environmental problem and the remediation of PAHs from these areas has been a major concern. The effectiveness of many in situ bioremediation systems may be constrained by low contaminant bioavailability due to limited aqueous solubility or a large magnitude of sorption. The objective of this research was to evaluate the effect of methyl-β-cyclodextrin （MCD） on bioaugmentation by Paracoccus sp. strain HPD-2 of an aged PAH-contaminated soil. When 10% （W/W） MCD amendment was combined with bioaugmentation by the PAH-degrading bacterium Paracoccus sp. strain HPD-2, the percentage degradation of total PAHs was significantly enhanced up to 34.8%. Higher counts of culturable PAH- degrading bacteria and higher soil dehydrogenase and soil polyphenol oxidase activities were observed in 10% （W/W） MCD-assisted bioaugmentation soil. This MCD-assisted bioaugmentation strategy showed significant increases （p 〈 0.05） in the average well color development （AWCD） obtained by the BIOLOG Eco plate assay, Shannon-Weaver index （H） and Simpson index （λ） compared with the controls, implying that this strategy at least partially restored the microbiological functioning of the PAH-contaminated soil. The results suggest that MCD-aided bioaugmentation by Paracoccus sp. strain HPD-2 may be a promising practical bioremediation strategy for aged PAH-contaminated soils.

AcMNPV and BmNPV are the two most widely used baculoviruses for the production of foreign proteins,and a number of modifications have been made to increase the quantity and quality of baculovirus ***,we provide a new method to improve baculovirus expression productivity without modifying the viral vector.MβCD,a chemical that is used to deplete cholesterol,enhanced baculovirus infection and foreign protein expression productivity.A low concentration of MβCD,with the optimal application concentration of 0.25 mM and the appropriate preincubation time range from 10 min to 120 min,can efficiently enhance expression levels in both the AcMNPV and BmNPV expression *** study provides a new method for baculovirus research and for application in biopharmaceutics for heterologous protein expression.

Synthetic phosphoethanolamine(Pho-s)is a monophosphoester ester with anti-inflammatory and pro-apoptotic *** chloridrate(MC)is a histamine H1 receptor blocker that is also able to inhibit cellular ***,MC does not inhibit cellular respiration in isolated mitochondria such as antimycin and ***-β-cyclodextrin(MβCD)belongs to theβ-cyclodextrin family,which is capable of removing cholesterol from the plasma *** aim of this study was to evaluate the proliferative effects of meclizine chloridrate and methyl-β-cyclodextrin compounds associated with synthetic phosphoethanolamine in a triple-negative human breast tumor line,MDA-MB-231 Cell viability of the tumor line and normal cells FN1 was evaluated by MTT colorimetric test;the production of free radicals was determined by lipoperoxidation(LPO)test;and the percentage of cell cycle phases and proliferative index was evaluated by flow *** viability demonstrated a significant decrease with the treatments of MβCD,MC and Pho-s associated with *** production of free radicals decreases significantly in all *** addition,a significant increase of DNA fragment and decrease in G0/G1 cell cycle phase were observed in cellular percentage with concentrations of 20 and 30 mM of Pho-s in association with MC and MβCD,respectively.

Insight into the role of cholesterol in modulation of morphology and mechanical properties of CH0-K1 cells: An in situ AFM study

Cholesterol plays a significant role in the organization of lipids and modulation of membrane dynamics in mammalian cells. However, the effect of cholesterol depletion on the eukaryotic cell membranes seems controversial. In this study, the effects of cholesterol on the topography and mechanical behaviors of CHO-K1 cells with manipulated membrane cholesterol contents were investigated by atomic force microscopy (AFM) technique. Here, we found that the depletion of cholesterol in cell membranes could increase the membrane stiffness, reduce the cell height as well as promote cell retraction and detachment from the surface, whereas the cholesterol restoration could reverse the effect of cholesterol depletion on the membrane stiffness. Increased methyl-β-cyclodextrin levels and incubation time could significantly increase Young’s modulus and degree of stiffing on cell membrane and cytoskeleton. This research demonstratede importance of cholesterol in regulating the dynamics of cytoskeleton-mediated processes. AFM technique offers excellent advantages in the dynamic monitoring of the change in membranes mechanical properties and behaviors during the imaging process. This promising technology can be utilized in studying the membrane properties and elucidating the underlying mechanism of distinct cells in the near-native environment.

Cholesterol synthesis inhibition or depletion in axon regeneration

Cholesterol is biosynthesized by all animal cells. Beyond its metabolic role in steroidogenesis, it is enriched in the plasma membrane where it has key structural and regulatory functions. Cholesterol is thus presumably important for post-injury axon regrowth, and this notion is supported by studies showing that impairment of local cholesterol reutilization impeded regeneration. However, several studies have also shown that statins, inhibitors of 3-hydroxy-3-methylglutaryl-Co A reductase, are enhancers of axon regeneration, presumably acting through an attenuation of the mevalonate isoprenoid pathway and consequent reduction in protein prenylation. Several recent reports have now shown that cholesterol depletion, as well as inhibition of cholesterol synthesis per se, enhances axon regeneration. Here, I discussed these findings and propose some possible underlying mechanisms. The latter would include possible disruptions to axon growth inhibitor signaling by lipid raft-localized receptors, as well as other yet unclear neuronal survival signaling process enhanced by cholesterol lowering or depletion.

Deposition of amyloid in the brain is a critical step in the pathogenesis of Alzheimer＇s disease. The endocytosis of β-amyloid peptide （Aβ） is an important factor among the many factors that contribute to the genesis of amyloid deposits. Since cholesterol participates in many important physiological processes, the present work investigated the relationship between the cellular cholesterol content and the endocytosis of the exogenic Aβ, and found that reduction of the cholesterol content by methyl-β-cyclodextrin could reduce the endocytosis of AI3. The study indicates that the endocytosis of Aβ is partly mediated by cholesterol.

Caveolin-1(Cav-1) is a resident protein of Caveolae,a subset of membrane microdomains enriched in cholesterol and ***-1 has been proved to bind with many signal molecules such as growth associated protein-43(GAP- 43),synaptophysin(Syn) and synaptosomal-associated protein 25(SNAP25) through the Caveolin binding *** studies indicate that during the process of active axon growth and synapse formation,Cav-1 plays an important role in regulating the rebuilding of neuron membrane. To investigate the role of Cav-1 in plasticity of central nervous system,PC12 cells were used for neuron model and treated by 1.5 mg/mL cholesterol binding drug,methyl-β-cyclodextrin (MβCD),for 24 h to down-regulate the expression of *** then the expression of GAP- 43,an internal determinant of neuron development and regeneration,was examined by Western *** results showed that 1.5 mg/mL MβCD successfully led to a down-regulation of Cav-1 expression (p<0.05) and GAP-43 expression was concomitantly attenuated(p<0.05 ) in PC12 cells,which suggested that Cav-1 might be involved in the regulation of GAP-43 expression and played an important role in the process of neuron development and regeneration.
