A novel fibrinolytic enzyme from Cordyceps militaris was purified and partially characterized for the first time, which was designated C. Militaris fibrinolytic enzyme（CMase）. This extracellular enzyme from C. militaris was isolated by ammonium sulphate fraction, and purified to electrophoretic homogeneity using gel filtration chromatography. The apparent molecular mass of the purified enzyme was estimated to be 27.3 kDa by SDS-PAGE. The optimum pH and temperature for the enzyme activity were pH 6.0 and 25℃, respectively. In the presence of metal ions such as Mg2+and Fe2+ions the activity of the enzyme increased, whereas EDTA and Cu2+ ion inhibited the enzyme activity. Interestingly the N-terminal amino acid sequences of the enzyme is extremely similar to those of the trypsin proteinases from insects, and has no significant homology with those of the fibrinolytic enzyme from other medicinal mushroom. In conclusion, C. militaris produces a strong fibrinolytic enzyme CMase and may be considered as a new source for thrombolytic agents

Squalene synthase catalyzes the condensation of two molecules of farnesyl diphosphate to produce squalene, the first committed precusor for sterol, brassinosteroid, and triterpene biosynthesis. A squalene synthase gene, designated Io SQS, was isolated from Inonotus obliquus, a medicinal mushroom produces a plethora of bioactive triterpenes. The Io SQS cDNA was found to contain an open reading frame of 1,476 bp, encoding a protein of 491 amino acids with a calculated molecular mass of 55.85 kDa. The Io SQS genomic DNA sequence consisted of 1,813 bp and contained four exons and three introns. The restriction fragment polymorphisms revealed by Southern blot analysis suggested that IoSQS was a single copy gene. Promoter analysis indicated that the 5’ upstream region of IoSQS possessed various potential elements associated with physiological and environmental factors. The expression pattern of IoSQS in different stages and under MeJA treatment were correlative with the accumulation of total triterpenoids, and was consistent with the predicted results of Io SQS promoter region. The N-terminal 466 residues of the hydrophilic sequence was expressed as a His-tagged protein in E. coli, and the resultant bacterial crude extract was incubated with farnesyl diphosphate and NADPH, squalene was detected in vitro reaction mixture by HPLC analysis. These results suggest that IoSQS enzyme is involved in squalene production in I. obliquus.

Ganoderma is a genus of medicinally and economically important mushrooms in the family Ganodermataceae.Ganoderma species are popular medicinal mushrooms and their health benefits are well-documented.Ganoderma is a cosmopolitan genus that is widely distributed in both tropical and temperate regions.This genus is characterized by its unique laccate or non-laccate species with double-walled basidiospores.Here,we report on eight collections of G.gibbosum collected during surveys in Kunming,Yunnan Province,China.The specimens are described and illustrated based on macro-and micro-morphological characteristics.Total DNA of the eight G.gibbosum strains were extracted using the Biospin Fungal Extraction Kit following manufacturer protocol.Amplification of the Internal Transcribed Spacer(nrITS)region was carried out using ITS5/ITS4 primers and LROR/LR5 for the nuclear ribosomal large subunit 28S rDNA gene(LSU).Phylogenetic analysis with closely related species to G.gibbosum showed that all eight collections grouped with G.gibbosum with 100%bootstrap support.Phylogenetic similarity and morphological variations within the eight collections of G.gibbosum are discussed.

Ganoderma lucidum sensu lato,is an ancient Chinese mushroom commonly known as“Reishi”or“Linghzi”which has been used for nutritional and therapeutic purposes.Reishi is a basidiomycete widely distributed in temperate and tropical regions.Among 2000 classes of Reishi,only 6 types are commonly presented to be effective for therapeutic purpose.These 6 types were known as black,red,blue,yellow,white and purple,respectively.Main constituents of Reishi are Polysaccharides,Triterpenoids,Sterols and Proteins.Reishi has been clinically used as immune system modulator,antiviral,cytotoxic agent,antibacterial,hepatoprotective,anti-inflammatory and also in the treatment of neurological disorders.The wide clinical scope of Reishi has influenced to name it in English with the nickname of“the mushroom of immortality”.

Identification of Common Edible and medicinal mushrooms by DNA Barcoding

Objective:To identify common edible and medicinal mushrooms using the internal transcribed spacer(ITS) region.Method:A total of eighty-five samples belonging to forty-three species were used in this study.ITS regions were amplified and sequenced.All ITS regions were analyzed using BLAST and MEGA 5.10 methods.Results:Forty species were identified correctly via BLAST method whereas forty-one species were distinguished via Neighbor joining(NJ)tree.All the species could be distinguished by BLAST combined with NJ tree and ITS sequence characteristics.Conclusion:Forty-three common species of edible and medicinal mushrooms were effectively identified using DNA barcoding technology based on ITS region.

The medicinal Fungi had played an important role in traditional Chinese medicine and have been used to treat different diseases in China for a long time. In recent years, many studies have been undertaken worldwide,and have ascertained that components of the mushroom possessed various therapeutic functions including immunomodulating activity, antitumor, antidiabetes and hepatoprotective ability and so on. It was found that many bioactivities of medicinal Fungi are relative with the polysaccharide. The study was focused on chemical structure and bioactivity of polysaccharides from 7 different important medicinal fungi, Ganoderma lucidum,Ganoderma sinense, Hericium erinaceus, Pleurotus eryngii,Coprinus comatus,Phellinus igniarius,Tremella aurantialba,Flammulina velutiper. 22 homogeneous polysaccharides were purified from hot water and alkali extracts of the fruiting body or spores using a series of chromatographic steps. Their chemical structures were elucidated by sugar analysis, methylation analysis and NMR. These polysaccharides was classified toβ-glucan,α-glucan, Fucogalactan, Fucoglucogalactan, Galactoglucan, acidic polysaccharides. Conformation of a 1,3/1,6-glucan from G. lucidum was studied by HPSEC-MALLS-RI system and Laser light scattering method;it was found conformation of the glucan is triple helix in solution. The bioactivity of these polysaccharides were found to induce macrophage activation and B or T cell activation, stimulate the proliferation of lymphocytes,stimulate the secretion of cytokines and immunoglobulins of lymphocytes and inhibit the Sarcoma S180 tumor growth. A sulfated polysaccharide deritive of Ganoderma lucidum, GLPss58, was found to possess the anti-inflammatory properties by inhibited the L-selectin – s Tyr/sLeX binding of lymphocytes. Our research results indicated that the structure and bioactivity of polysaccharides from medicinal fungi was diversity, it is difficult to obtain the clear structure-functional relationship of polysaccharides from medicinal fungi.
