Is there hybridization between 2 species of the same genus in sympatry?——The genetic relationships between Anoplophora glabripennis,Anoplophora chinensis,and putative hybrids

Anoplophora glabripennis(Asian longhorn beetle,ALB)and Anoplophora chinensis(Citrus longhorn beetle,CLB)are native forest pests in China;they have become important international quarantine pests.They are found using the same Salix aureopendula host tree of Cixi,Zhejiang province,China.On this host tree,we collected additional beetles that appeared to be morphologically intermediate between ALB and CLB.By using a stereoscope,we observed that there were several bumps on the base of the elytra,which was inconsistent with ALB,which typically has a smooth elytral base,but was more like CLB,which has numerous short tubercles on the elytral base.Given their sympatry and intermediate morphology,we hypothesized that these may represent ALB×CLB hybrids.We studied the genomic profiles for 46 samples(ALB,CLB,and putative hybrids)using genotyping-by-sequencing(GBS)providing a reduced representation of the entire genome.Employing principal component analyses on the 163 GBS-derived single nucleotide polymorphism data,we found putative hybrids tightly clustered with ALB,but genetically distinct from the CLB individuals.Therefore,our initial hybrid hypothesis was not supported by genomic data.Further,while mating experiments between adult ALB and CLB were successful in 4 separate years(2017,2018,2020,and 2021),and oviposition behavior was observed,no progeny was produced.Having employed population genomic analysis and biological hybridization experiments,we conclude that the putative hybrids represent newly discovered morphological variants within ALB.Our approach further confirmed the advantage of genome-wide information for Anoplophora species assignment in certain ambiguous classification cases.

The effects of prey lipid on female mating and reproduction of a wolf spider

As predators,the macronutrients spiders extract from their prey play important roles in their mating and reproduction.Previous studies of macronutrients on spider mating and reproduction focus on protein,the potential impact of prey lipid content on spider mating and reproduction remains largely unexplored.Here,we tested the influence of prey varying in lipid content on female mating,sexual cannibalism,reproduction,and offspring fitness in the wolf spider Pardosa pseudoannulata.We acquired 2 groups of fruit fly Drosophila melanogaster that differed significantly in lipid but not protein content by supplementing cultural media with a high or low dose of sucrose on which the fruit flies were reared(HL:high lipid and LL:low lipid).Subadult(i.e.,1 molt before adult)female spiders that fed HL flies matured with significantly higher lipid content than those fed LL flies.We found that the mated females fed with HL flies significantly shortened pre-oviposition time and resulted in a significantly higher fecundity.However,there was no significant difference in female spiders varying in lipid content on other behaviors and traits,including the latency to courtship,courtship duration,mating,copulation duration,sexual cannibalism,offspring body size,and survival.Hence,our results suggest that the lipid content of prey may be a limiting factor for female reproduction,but not for other behavioral traits in the wolf spiders P.pseudoannulata.

Light is one of the key environmental factors for insects to survive.Artificial light sources different from natural environmental light can cause light stress in insects.Yellow and green light stress can interfere with the diurnal rhythm of nocturnal moths and their mating,oviposition,and adult longevity.The scarabaeid beetles Anomala corpulenta Motschulsky and Holotrichia parallela Motschulsky are widely distributed,and they are very harmful underground pests.In order to clarify the effects of light stress on their behaviors,individuals of both species were exposed to yellow light(565-585 nm)and green light(525-545 nm),with different light intensity gradients of yellow light in a laboratory setting.The short-term light stress treatment of A.corpulenta and H.parallela was carried out at night.The number of beetles emerging per half an hour was recorded,and mating pairs and feeding activity in 24 h were counted.The results showed that yellow and green light stress significantly changed the rhythm and reduced the rate of beetle emergence in the two beetle species investigated.Also,the peak emergence activity was delayed and the feeding and mating activities were significantly reduced.When treated with different intensities of yellow light,it was found that the rate of emergence of A.corpulenta under 10 lx was close to the control groups.The rate of emergence in H.parallela was significantly lower than the control groups before 0:00,in the 60 lx and 110 lx treatment groups,but after 0:00,the emergence rate of H.parallela was significantly higher in the 60 lx and 110 lx treatment groups than other treatments.However,the emergence rhythms in the three light intensity treatment groups are basically the same as in the control groups.The feeding amount and mating beetles in the three light intensity treatment groups were significantly lower than in the control groups.There were no significant differences in the three treatments.The results show that light stress above 10 lx significantly interferes with the behavioral a

Camellia oleifera Abel.is an important economic tree species of southern China.In this study,we evaluated the mating system and genetic diversity of a series of cultivars of C.oleifera‘Ruan Zhi’.A total of 159 individuals from the progenies of four cultivars were tested by simple sequence repeat molecular markers.Results reveal that 11 pairs of primers showed polymorphism and their polymorphism information content value was greater than 0.73,suggesting that these primers could be used to identify the genetic diversity of open-pollinated populations.The average number of effective alleles(Ne=4.88)was significantly different from the average number of alleles(Na=12.18),and their distribution in the sample population was not uniform.The average observed heterozygosity(Ho=0.96)was greater than the average expected heterozygosity(He=0.79),and the population heterozygote was excessive.Shannon index was 1.84 and populations showed high genetic diversity.As regards to the mating system,the multilocus outcrossing rate was 0.996,and the single locus 0.866.These results indicate a high degree of outcrossing by C.oleifera‘Ruan Zhi’.We recommend selecting individuals for high genetic gain from the progenies of cultivars because of outcrossing characteristics and genetic diversity for application to germplasm conservation and promotion.

Emergence,mating and Oviposition Behavior of the Chinese Population in Pink Bollworm,Pectinophora gossypiella (Lepidoptera:Gelechiidae)

The pink bollworm, Pectinophora gossypiella （Lepidoptera： Gelechiidae） is one of the most serious lepidopteran pests of cotton in the world. This pest invaded China at the onset of the 20th century, possibly through repeated introductions from several different locations worldwide. In this paper, we describe different behavioral parameters of this Hubei P. gossypiella strain under laboratory and field conditions. Using an infra-red video recorder, we observed （nocturnal） emergence, flight and mating activities, and oviposition patterns. Moth emergence started from 13：00 and continued up till 23：00. Under laboratory and field conditions, 2-7-d-old moths initiated flight around sunset, peaked 50 rain later and gradually declined until 04：00. Although mating started immediately after darkness （i.e., 20：00）, mating behavior was most intense from 23：00 to 03：40. Oviposition also showed distinct time-related patterns, with approx. 70% eggs laid between 20：00 and 22：00. In the studies of the relationship between flight and oviposition, the duration of flight had an effect on oviposition. The 1-d-old moths flown for 6 and 12 h began oviposition earlier than the unflown ones, and the 6-h flight had no effect on the egg production. However, the longevity of the flown moths was shortened after flight. In addition, the peaks of oviposition for the flown moths were advanced 2-3 d. The age when the moths flied affected the oviposition of adult moths. The earlier the moths started to fly after emergence, the greater the fecundity they had. The average egg production of moths flown for 24 h was less than that of non-flown moths which were of the same age. Both flight and delay in mating could advance the oviposition peak. Our study provides detailed insights in nocturnal activities and the reproductive biology of local P. gossypiella populations, which could be employed to fine-tune current pest management programs in China.

The genetic diversity, individual relatedness and possible mating system of an isolated population of the Cyprinid species Megalobrama pellgrini in upper reaches of the Changjiang (Yangtze) River, China

Megalobrama pellegrini is a cyprinid fish endemic to upper reaches of the Changjiang(Yangtze)River,China,which is also an important economic species in the local area.In recent years,resources of this species have decreased sharply and its conservation has drawn great attention.In the present study,we collected 120 individuals from the Longxi River,a tributary isolated from the main channel of the Changjiang River,where M.pellegrini is still relatively abundant.Using two different molecular markers,mitochondrial cytochrome b(cyt b)gene,and nuclear microsatellite(simple sequence repeat,SSR),we analyzed the genetic diversity of this isolated population.The results show that sequence genetic diversity was low(Hd:0.290 and P i:0.000 77 for cyt b gene),while the SSR genetic diversity was high(Ho:0.824 4±0.147 2,He:0.823 5±0.145 1).Analysis indicated that this population had experienced a bottleneck,with inbreeding and small effective population size(around 50).Based on SSR data,relatedness analyzing revealed that the 120 samples were grouped into 10 completely independent clusters.It was inferred that the mating system of M.pellegrini was polygamy.We suggested that the low genetic diversity could be induced by the overfishing and inbreeding depression.Therefore,we suggested that the urgent conservation measures should be taken to control the overfi shing and give better conditions for the fish to grow and spawn,then to restore population size.

Effects of Various Degrees of Antennal Ablation on mating and Oviposition Preferences of the Diamondback Moth,Plutella xylostella L.

Using scanning electron microscopy, we investigated the distribution of the trichoid, basiconic, and coeloconic sensilla on the antennae of the diamondback moth （DBM;Plutella xylostella）. The trichoid sensilla were the most abundant sensory organ, and the male moth antennae host signiifcantly more trichoid sensilla than female moth antennae. Conversely, basiconic and coeloconic sensilla were found more frequently on female than on male antennae. We performed experiments with various degrees of antennal ablation and demonstrated that DBM antennae played a key role in the control of mating and oviposition. We found that neither oviposition preference nor mating behaviors changed signiifcantly when less than 1/4 of both antennae were removed. However, there was a signiifcant behavioral change when the antennae were ablated by more than half. As the length of the antenna was shortened, the successful mating rate decreased and mating peak was delayed. An otherwise consistent host preference for oviposition was eliminated when both antennae were completely removed. Furthermore, we found that the number of trichoid sensilla was positively correlated with mating rate and oviposition preference. However, the numbers of basiconic and coeloconic sensilla were not correlated with mating rate and mating peak, but highly correlated with oviposition preference. Taken together, our results indicate that antennal sensory information plays a critical role in the mating and oviposition behaviors of this economically important pest.

Population structure and mating system of the Red-crested Korhaan(Lophotis ruficrista) in South Africa

Background: The mating system of the korhaans and bustards in southern Africa is either based on polygyny or monogamy. The Red-crested Korhaan(Lophotis ruficrista) has been described as polygynous but otherwise very little is known about its breeding biology. The aims were to describe the population structure and male behavior during breeding.Methods: The data collected for this paper was mainly based on field transect surveys carried out in 2009, 2010 and 2011. The population structure of the Red-crested Korhaan in the temperate Borakalalo Game Reserve(BGR)was compared with that in the arid Molopo Nature Reserve(MNR)(both situated in the North West province of South Africa). The study was mainly conducted in the BGR but additional work was conducted in the MNR for comparison.Results: The difference in mean group sizes between MNR(1.03) and BGR(1.07) was statistically not significant.Group sizes were not affected by climate despite the fact that the BGR received about 650 mm precipitation per annum and the MNR about 200 mm. In both reserves the apparent sex ratios were skewed in favor of males(1:0.29 in the BGR and 1:0.1 in the MNR). The population was dispersed in a clumped manner which is ascribed to the formation of leks. Leks were positioned in open habitat while females invariably concealed themselves under cover.Conclusions: The leks formed the pivot of the mating system of the Red-crested Korhaan and are identifiable social structures that field ecologists can use to monitor population stability.

Despite the importance of reproduction in insects,its relation with multi-stop flight remains poorly understood in migratory species.To clarify whether reproductive maturation commences during the multi-stop flight or after the completion of migration,we conducted physiological and behavioral assays in the rice leaf roller Cnaphalocrocis medinalis with laboratory-simulated conditions and field-captured populations.We found that the ovarian development was significantly promoted by tethered flight treatment for 1-2 nights when compared to the unflown group,while the flight muscle development was not impaired.There was no significant difference in flight duration,flight distance and flight velocity between mated and virgin female moths,indicating that mated moths remained competent for the subsequent flights as did the virgins.Using an integrated field assay,we identified that over 60%of the female moths in the migrating populations cap-tured by high-altitude searchlights in the Immigration period of a season had completed the ovarian development and mating.Sexually mature and mated moths collected in the rice field in the Emigration period were found capable of engaging in migratory take-off,as observed using an indoor monitoring platform.Overall,our findings point out that C.med-inalis managed to complete reproductive maturation to a large extent during the multi-stop migratory flight without compromising the migration performance.Such a cost-effective strategy ensures a successful migration for the moths.These findings advance our under-standing of the relationship between reproduction and migration,thus shedding light on the development of novel control measures for the outbreak of migratory insect pests.

Behavior of Brown Bears (Ursus arctos) in Captivity during mating Season

From 1991 to 1992, we observed behavior of 7 brown bears (5 females, 2 males) in mating season. Re-Sults showed tkat the oertrus of brown bears was mainly from May to July. The oeStrus ofmost individuals occwred at theend of May and the beginning of June. There were some differences in the oestrus among individuals. Average duration ofthe oestrus 30.8 days (16-50 days). The mating behavior was mainly observed during 8.00-9.00 a.m., and 15.00-16.00p.m.. Rest and sleep were the next behavior after mating for most bears, especially for male bears. All bears had poor appe-the. The Pattern and process of a successful mating was that a male bear approached a female, smelled the pudendum of thefemale, climbed on female hip, mated, parted and rested. Oestrus females approached males sometimes.
