Dual role of lipids for genome stability and pluripotency facilitates full potency of mouse embryonic stem cells

While Mek1/2 and Gsk3βinhibition("2i")supports the maintenance of murine embryonic stem cells(EsCs)in a homogenous naive state,prolonged culture in 2i results in aneuploidy and DNA hypomethylation that impairs developmental potential.Additionally,2i fails to support derivation and culture of fully potent female ESCs.Here we find that mouse ESCs cultured in 2i/LIF supplemented with lipid-rich albumin(AlbuMAx)undergo pluripotency transition yet maintain genomic stability and full potency over long-term culture.Mechanisticaily,lipids in AlbuMAx impact intracellular metabolism including nucleotide biosynthesis,lipid biogenesis,and TCA cycle intermediates,with enhanced expression of DNMT3s that prevent DNA hypomethylation.lipids induce a formative-like pluripotent state through direct stimulation of Erk2 phosphorylation,which also alleviates X chromosome loss in female ESCs.Importantly,both male and female"all-ESc"mice can be generated from de novo derived ESCs using AlbuMAXbased media.Our findings underscore the importance of lipids to pluripotency and link nutrient cues to genome integrity in early development.

lipids have been found to modulate tumor biology,including proliferation,survival,and metastasis.With the new understanding of tumor immune escape that has developed in recent years,the influence of lipids on the cancer—immunity cycle has also been gradually discovered.First,regarding antigen presentation,cholesterol prevents tumor antigens from being identified by antigen presenting cells.Fatty acids reduce the expression of major histocompatibility complex class I and costimulatory factors in dendritic cells,impairing antigen presentation to T cells.Prostaglandin E2(PGE2)reduce the accumulation of tumor-infiltrating dendritic cells.Regarding T-cell priming and activation,cholesterol destroys the structure of the T-cell receptor and reduces immunodetection.In contrast,cholesterol also promotes T-cell receptor clustering and relative signal transduction.PGE2 represses T-cell proliferation.Finally,regarding T-cell killing of cancer cells,PGE2 and cholesterol weaken granule-dependent cytotoxicity.Moreover,fatty acids,cholesterol,and PGE2 can improve the activity of immunosuppressive cells,increase the expression of immune checkpoints and promote the secretion of immunosuppressive cytokines.Given the regulatory role of lipids in the cancer—immunity cycle,drugs that modulate fatty acids,cholesterol and PGE2 have been envisioned as effective way in restoring antitumor immunity and synergizing with immunotherapy.These strategies have been studied in both preclinical and clinical studies.

Distribution characteristics of lipids from salt sediments in Qaidam Basin and their astrobiological significance

On Earth,salt environments are the most relevant analogues to Martian salt deposits with regard to the search for preserved biomolecules.In this study,we analyzed the distribution of lipids in salt samples from Dalangtan and Qarhan in the Qaidam Basin,which is the most important area to carry out analog research of Mars in China.Furthermore,we compared the lipids distribution results with that from other typical Martian analog sites such as the Atacama Desert.Salt samples in the Qaidam Basin are enriched with fatty acid compounds,including normal fatty acids,unsaturated fatty acids,branched fatty acids,and acyclic isoprene acids.In addtion,glycerol dialkyl glycerol tetraethers(GDGTs)and archaeol compounds were also detected in these salt samples.Compared with the hypersaline samples,the clay samples not only have higher abuandance of fatty acids,GDGTs and archaeol compounds,but also a more diversed lipids composition.Only a few lipids were detected in the pure-saline samples,such as archaeol compounds.These clues suggest that content of salt minerals is the major factor affecting the distribution of lipids.This study provides an analogy for detecting and evaluating the traces of life that may exist on Mars.

Exploiting styrene-maleic acid copolymer grafting chromatographic stationary phase materials for separation of membrane lipids

High performance liquid chromatography-mass spectrometry is one of the most commonly used strategies for lipid analysis.The development of versatile chromatographic stationary phases to meet the increasing demands for separation of complex lipids is very important.Styrene-maleic acid(SMA)copolymer is an amphiphilic polymer,which has been proven to have the ability to solubilize lipid molecules of various structures.In this study,styrene-maleic anhydride copolymer coated silica was first prepared by the thiol-ene click reaction.With L-cysteine hydrochloride or dodecanol as the post-modification reagents,Sil-SMA-amino acid and Sil-SMA-dodecanol stationary phase materials were further successfully fabricated via nucleophilic ring-opening reaction.The Fourier-transform infrared,thermogravimetric analysis,and elemental analysis results confirmed the two stationary phase materials were successfully prepared.Furthermore,both the Sil-SMA-dodecanol column and the Sil-SMA-amino acid column possessed reversed-phase/hydrophilic interaction/ion exchange mixed-mode retention mechanisms.The column efficiency of the Sil-SMA-derivatives columns reached 77,300 N/m.Based on the mixed-mode retention characteristics,the Sil-SMA-derivatives columns achieved both the lipid classes and species separation via a single column.The Sil-SMA-amino acid column was further successfully used to separate lipid extract from gastric cancer cell membrane.All these results demonstrated that the SMA-based stationary phase materials have a good potential for use in lipid separation.

Trends in lipids Level and Dyslipidemia among Chinese Adults, 2002-2015

Objective To investigate the trends of lipid profiles and dyslipidemia among Chinese adults from 2002 to 2015.Methods Data were collected from three nationally representative cross-sectional surveys.Fasting venous blood samples were collected and serum lipids were tested by biochemical analysis and enzymatic determination.Lipid levels and the prevalence of dyslipidemia among adults were analyzed with complex sampling weighting adjustment for age and gender.Results The weighted means of TC, TG, and LDL-c significantly increased linearly from 3.93, 1.12, and 2.12 mmol/L in 2002 to 4.59, 1.41, and 2.78 mmol/L in 2010 and then to 4.63, 1.47, and 2.87 mmol/L in 2015, respectively;by contrast, HDL-c levels decreased significantly from 1.30 mmol/L to 1.26 mmol/L over the same period.Similar trends in mean non-HDL-c and lipid-related ratios were observed.The weighted dyslipidemia prevalence linearly increased;in particular, hypercholesterolemia increased from 1.6% to 5.6% and then to 5.8%, hypertriglyceridemia increased from 5.7% to 13.6% and then to 15.0%, low HDL-c increased from 18.8% to 35.5% and then to 24.9%, and high LDL-c increased from 1.3% to 5.6% and then to 7.2%(P for trend <0.001).Conclusion Dyslipidemia increased among Chinese adults from 2002 to 2015.Development of a comprehensive strategy to decrease lipid levels in this population is urgently required.

Tumours are known to be a heterogeneous group of cells,which is why they are difficult to eradicate.One possible cause for this is the existence of slow-cycling cancer stem cells(CSCs)endowed with stem cell-like properties of self-renewal,which are responsible for resistance to chemotherapy and radiotherapy.In recent years,the role of lipid metabolism has garnered increasing attention in cancer.Specifically,the key roles of enzymes such as stearoyl-CoA desaturase-1 and 3-hydroxy-3-methyl-glutaryl-coenzyme A reductase in CSCs,have gained particular interest.However,despite accumulating evidence on the role of proteins in controlling lipid metabolism,very little is known about the specific role played by lipid products in CSCs.This review highlights recent findings on the role of lipid metabolism in CSCs,focusing on the specific mechanism by which bioactive lipids regulate the fate of CSCs and their involvement in signal transduction pathways.

Distribution characteristics of lipids in hadal sediment in the Yap Trench

In this study,cored sediment samples collected by the Jiaolong Submersible at 6779 m depth from the hadal zone of the Yap Trench in May 2016 were sliced in 1-cm interval from top to bottom,and lipids in each sediment layer were quantitatively and qualitatively analyzed.The vertical distribution profiles of the lipids in the sediment sample,their main existing forms,and their possible sources were investigated.The results show that the concentration of lipid in the surface sediment was the highest with the carbon number from 12 to 27,dominated by medium and short-chain lipids.The total concentration of fatty acids in surface sediment was much higher than those in the offshore and deep-sea areas,being up to 325.77μg/g due to the funnel effect caused by the“V”terrain of the trench.Fatty acid 18:0 was the most abundant lipids in the sediment sample.Abnormal high concentrations of fatty acid 18:1ω7 and alkanes indicated the existence of hydrothermal fluids in the study area.In addition,saturated fatty acids and monounsaturated fatty acids existed mainly in free form,and polyunsaturated fatty acids existed mainly in bound form.Most of the alkanes were in bound form,and their major source was autochthounous input.The carbon number of alcohols in the sediment sample ranged from 12 to 20,mainly existed in bound form.The source of fatty acids was mainly autochthonous input,and the neutral lipids had both marine and terrestrial origin.This is the first study of lipids in hadal sediment of the Yap Trench.The results will promote deeper understanding of organic carbon cycle in marine environment.

Association between dairy intake,lipids and vascular structure and function in diabetes

AIM To determine lipid species that change in response to a change in dairy consumption. In addition, to investigate whether dairy associated lipid species are correlated with changes in measures of vascular structure and function.METHODS A 12-mo randomised controlled trial was conducted to determine the effect of increased consumption of fruit, vegetables and dairy, compared to usual diet, on measures of vascular structure and function in adults with type 1 and type 2 diabetes(n = 108). Thispaper comprises post-hoc analyses investigating the relationship between dairy intake, serum lipid species and vascular health. Central and peripheral blood pressure, carotid femoral pulse wave velocity, augmentation index, serum lipid species and dietary intake were measured at baseline and 3-mo. Common carotid artery intima media thickness was measured at baseline and 12-mo.RESULTS Serum lipid species [lysophosphatidylcholine(LPC) 14:0, LPC 15:0, LPC 16:1, phosphatidylcholine(PC) 29:0 PC 30:0, PC 31:0 and cholesterol ester(CE) 14:0] were associated with the change in full fat dairy consumption(rho 0.19-0.25; P lipids was positively associated with the 3-mo change in central systolic [LPC 14:0(rho 0.30; P = 0.007), PC 30:0(rho 0.28; P = 0.010)] and diastolic blood pressure [LPC 14:0(rho 0.32; P = 0.004), LPC 15:0(rho 0.23; P = 0.04), LPC 16:1(rho 0.23; P = 0.035), PC 29:0(rho 0.28; P = 0.01), PC 30:0(rho 0.36; P = 0.001), PC 31:0(rho 0.30; P = 0.007)] and 12-mo change in common carotid artery intimal medial thickness [CE 14:0(rho 0.22; P = 0.02)]. Pulse wave velocity and augmentation index were unrelated to dairy and lipid species.CONCLUSION An increase in dairy associated lipids appears to be associated with an increase in blood pressure and common carotid intimal medial thickness.

Changes of pigments and lipids composition in Haematococcus pluvialis vegetative cell as affected by monochromatic red light compared with white light

Much attention has been paid on studies of astaxanthin accumulation process in H aematococcus pluvialis industry. However, growth of H. pluvialis in motile vegetative stage is still the most important and problematic part in the whole cultivation process, such as low growth rate and cell yields. Motile vegetative cells are extremely sensitive to various stresses which make it difficult to maintain the cells of this state to grow. Previous reports showed that motile vegetative cells may have higher biomass yields ifapplied monochromatic red light. However, metabolic responses of these cells are not completely understood, which constraints application of this illumination protocol in industry. The aim of this study was to examine how critical biochemical changes of H. pluvialis motile vegetative cells were af fected by red light when compared with white light. Variation of photosynthetic pigments composition and lipids were mainly studied. By comparing growth process of cultures in red light and white light, prominent variation of pigments composition and lipids changes were observed. The results showed that, even though cell proliferation was the same during exponential growth phase, variation of photosynthetic pigment composition and lipids occurred. The final biomass of cell number was higher in red light group than in white light group. The variations were significant different. Increase or decrease of major photosynthetic membrane lipids to some extent did not influence photosynthesis of the vegetative cells during this phase. However, vegetative cells under polychromatic white light other than monochromatic red light need further metabolic process to adjust its pigment composition and lipids, possibly this is energetically and biochemically unfavorable for motile vegetative cells to growth under white light, a light condition normally not considered as a stress.

Tumour growth and dissemination is largely dependent on nutrient availability.It has recently emerged that the tumour microenvironment is rich in a diverse array of lipids that increase in abundance with tumour progression and play a role in promoting tumour growth and metastasis.Here,we describe the pro-tumorigenic roles of lipid uptake,metabolism and synthesis and detail the therapeutic potential of targeting lipid metabolism in cancer.Additionally,we highlight new insights into the distinct immunosuppressive effects of lipids in the tumour microenvironment.lipids threaten an anti-tumour environment whereby metabolic adaptation to lipid metabolism is linked to immune dysfunction.Finally,we describe the differential effects of commondietary lipids on cancer growth which may uncover a role for specific dietary regimens in association with traditional cancer therapies.Understanding the relationship between dietary lipids,tumour,and immune cells is important in the context of obesity which may reveal a possibility to harness the diet in the treatment of cancers.
