PP2A interacts with katanin to promote microtubule organization and conical cell morphogenesis

The organization of the microtubule cytoskeleton is critical for cell and organ *** evolutionarily conserved microtubule-severing enzyme katanin plays critical roles in microtubule organization in the plant and animal *** previously used conical cell of Arabidopsis thaliana petals as a model system to investigate cortical microtubule organization and cell morphogenesis and determined that katanin promotes the formation of circumferential cortical microtubule arrays in conical ***,we demonstrate that the conserved protein phosphatase PP2A interacts with and dephosphorylates katanin to promote the formation of circumferential cortical microtubule arrays in conical *** undergoes cycles of phosphorylation and *** co-immunoprecipitation coupled with mass spectrometry,we identified PP2A subunits as katanin-interacting *** biochemical studies showed that PP2 A interacts with and dephosphorylates katanin to stabilize its cellular *** to the katanin mutant,mutants for genes encoding PP2A subunits showed disordered cortical microtubule arrays and defective conical cell *** together,these findings identify PP2A as a regulator of conical cell shape and suggest that PP2A mediates katanin phospho-regulation during plant cell morphogenesis.

The katanin Microtubule Severing Protein in Plants

katanin is a heterodimeric microtubule （MT） severing protein that uses energy from ATP hydrolysis to generate internal breaks along MTs. katanin p60, one of the two subunits, possesses ATPase and MT-binding/severing activities, and the p 80 subunit is responsible for targeting of katanin to certain subcellular locations. In animals, katanin plays an important role in the release of MTs from their nucleation sites in the centrosome. It is also involved in severing MTs into smaller fragments which can serve as templates for further polymerization to increase MT number during meiotic and mitotic spindle assembly. katanin homologs are present in a wide variety of plant species. The Arabidopsis katanin homolog has been shown to possess ATP-dependent MT severing activity in vitro and exhibit a punctate localization pattern at the cell cortex and the perinuclear region. Disruption of katanin functions by genetic mutations causes a delay in the disappearance of the perinuclear MT array and results in an aberrant organization of cortical MTs in elongating cells. Consequently, katanin mutations lead to defects in cell elongation, cellulose microfibril deposition, and hormonal responses. Studies of katanin in plants provide new insights into our understanding of its roles in cellular functions.

Microtubule（MT） defects lead to various neurological diseases such as hereditary spastic *** many MT regulators,katanin is the first identified MT-severing protein,but its neuronal functions have not been characterized in a multi-cellular *** this study,we generated multiple mutations in Drosophila katanin 60 and manipulated its expression in a tissue-specific manner to dissect the in vivo functions of *** katanin 60is essential for adult viability as its null mutants are pupal *** 60 nulls showed an increased MT network formation,whereas overexpression of katanin 60 resulted in reduced MT network with fragmented MTs at neuronal and non-neuronal *** the neuromuscular junctions,katanin 60 mutants showed excess satellite boutons,reduced neurotransmission efficacy,and more enlarged *** sensory neurons,katanin 60 negatively regulates dendritic *** interaction analyses supported that increased level of acetylation of MTs renders them more susceptible to katanin *** our knowledge,this study represents the first in vivo characterization of katanin 60’s role in synapse and dendritic development,and offers new insight into the neuronal roles of MT-severing proteins.

微管剪切蛋白katanin是ATPase家族的一员,由katanin60和katanin80两个亚基组成。其中katanin80亚基没有酶切功能但可使katanin定位于中心体。katanin60亚基具有ATP酶活,行使微管剪切功能。目前对katanin体内功能的研究只限于模式动物线虫,尚没有在其他模式动物如果蝇和小鼠中研究,因此,体外实验的结果还没有得到体内试验的验证。其次,虽然体外的研究阐明了katanin突变可以导致其细胞中微管网络的破坏,但是由于这些研究是基于体外培养细胞的研究,因此不能完全解释katanin在个体生长发育特别是神经肌肉系统发育过程中的作用。我们从VDRC(维也纳果蝇中心)购得katanin60RNAi果蝇工具株;并利用p因子定点跳出的方法获得了katanin60的突变体果蝇;通过扩增katanin60CDS连入pUAST载体,显微注射到果蝇胚胎后进行筛选和定位,得到katanin60过表达果蝇工具株。并从国外合作者处得到抗体,随后利用免疫染色和生化分子等方法检测katanin60过表达和敲减果蝇以及突变体的蛋白表达量和表达位置。在鉴定和验证以上各种工具后,我们用微管特异性抗体对果蝇三龄幼虫肌肉细胞的微管结构进行了观察,同时还用神经系统也以表达的蛋白观察了果蝇神经突触结构的发育。我们在果蝇体内进行了研究。免疫染色结果显示,在肌肉细胞中,过表达katanin60可以提高微管剪切的程度,且优先剪切细胞核周围的微管,造成细胞核周围微管密度减小,而敲减果蝇katanin60的表达可以造成细胞核周围微管密度增加。katanin60突变体果蝇的肌肉细胞微管成簇且延伸至细胞边缘。在冷冻处理恢复微管的实验中我们看到,katanin功能的行使需要一定的微管密度。katanin不仅影响果蝇肌肉微管的发育,同时影响神经系统的发育。在katanin60过表达和突变体果蝇中,神经肌肉接头卫星扣结数目较野生型明显增多。

微管是细胞骨架的主要成份,参与细胞内物质的运输与细胞形态的维持,还与有丝分裂和减数分裂等生命活动密切相关。大多数微管都表现出动力学的不稳定性,处于动态的聚合和解聚及之间的随机转换状态。Kinesin-13、Stathmin和katanin是三类能够解聚微管的蛋白质,在纺锤体组装、染色体分离和神经元发育过程中起重要作用。本文主要对这三类微管解聚相关蛋白质的结构、功能、解聚机制进行了简要介绍,并对它们的解聚机制进行了比较。

种子大小形状是农作物重要的农艺性状,粒大饱满的种子是农业育种和生产的努力方向。从拟南芥这一模式生物入手,探索种子大小形状的调控机理,进行深入的研究具有重要的意义。本课题采用正向遗传学方法,从两个圆种子突变体材料er5(EMS诱变材料)、lip1(T-DNA插入材料,但T-DNA丢失)出发,利用图位克隆技术结合高通量测序的方法克隆突变基因。另外从整体水平、细胞学水平以及分子水平分析突变体表型,并结合表型对突变基因进行一些基本的功能分析,试图探究种子形状调控相关机理和植物细胞中的极性伸长问题。主要取得如下结果:1.两个纯合突变体都具有比较一致的表型。纯合突变体植物体各组织器官都显示出横向变宽,纵向变短的表型,具体为叶圆,叶柄短且粗,茎粗且脆,果荚短且粗等。er5杂合突变体植株体现类似但较弱的表型,为半显性遗传,而lip1杂合植株与野生型植株类似,为典型的隐性遗传。***5,lip1突变基因克隆。通过图位克隆结合下代测序,两个突变基因都被克隆,突变位点都位于AT1G80350基因上,该基因编码微管切割蛋白katanin p60。不同的是er5为点突变,位于最后一个外显子倒数第5个碱基(C to T),造成p60蛋白C末端倒数第二个氨基酸由丝氨酸突变为苯丙氨酸(S to F)。lip1可能是因为T-DNA插入引起重组的原因造成p60基因在第六个外显子与第六个内含子交界处缺失43个碱基。3.突变体细胞学表型及机理探索。er5和lip1纯合突变体各器官表型经细胞生物学分析表明都是由体细胞极性伸长受阻引起。自然条件下,育性非常差,产生极少量种子,人工授粉可部分恢复育性产生较多种子。突变体种子为圆形,且体现母性效应。通过DIC显微镜观察,纯合圆种子的胚和胚柄细胞的极性伸长也受阻,发育异常,并且存在具有多个胚的种子。为进一步探究多胚产生的时期和原因,已将大孢子母细胞(MMC)、卵细胞、助细胞、中心细胞的特异性标记和不同种激素报告系统引入突变体。这些材料将为进一步观察突变体雌配子体发育的过程,各细胞形态、位置、数量以及细胞命运的变化,从而为回答雌配子体发育过程中细胞极性或激素的极性分布对其发育的影响,植物胚胎发生等生物学问题提供了材料基础。
