Severe burn injury alters intestinal microbiota composition and impairs intestinal barrier in mice

Background:The intestinal barrier integrity is crucial for maintaining intestinal homeostasis,and the mechanisms of intestinal barrier disruption induced by burn injury remain *** study was aimed to investigate the changes of intestinal microbiota and barrier function in burned mice to further comprehend the mechanisms of burn-induced intestinal barrier ***:Samples were from mice inflicted with 30%total body surface area(TBSA)full-thickness *** intestinal permeability,tight junction proteins expressions,zonula occludens-1(ZO-1)localization,inflammatory cytokines expressions,and short-chain fatty acids(SCFAs)contents were *** microbial community was assessed via 16S rDNA Illumina ***:The intestinal permeability was increased after severe burn injury,peaking at 6 h post-burn,with approximately 20-folds of the control(pinflammatory cytokines(interleukin(IL)-1βand IL-6)in ileal mucosa were increased,whereas the expressions of anti-inflammatory cytokines(IL-4 and IL-13)were decreased following burn injury(p<0.05).In addition,burned mice showed an alteration of intestinal microbial community,such as decreased diversity,reduced Bacteroidetes abundance,and increased Firmicutes ***:The severe burn-induced intestinal barrier dysfunction is along with the alterations of microbial community.

Can Helicobacter pylori infection influence human reproduction?

Helicobacter pylori (H. pylori) infection could be associated with extra-digestive diseases. Here, we report the evidences concerning the decrease in reproductive potential occurring in individuals infected by H. pylori, especially by strains expressing CagA. This infection is more prevalent in individuals with fertility disorders. Infected women have anti-H. pylori antibodies in cervical mucus and follicular fluid that may decrease sperm motility and cross react immunologically with spermatozoa, conceivably hampering the oocyte/sperm fusion. Infection by CagA positive organisms enhances the risk of preeclampsia, which is a main cause of foetus death. These findings are supported by the results of experimental infections of pregnant mice, which may cause reabsorption of a high number of foetuses and alter the balance between Th1 and Th2 cell response. Infected men have decreased sperm motility, viability and numbers of normally shaped sperm and augmented systemic levels of inflammatory cytokines, such as tumor necrosis factor-α, which may damage spermatozoa. In countries where parasitic infestation is endemic, detrimental effects of infection upon spermatozoa may not occur, because the immune response to parasites could determine a switch from a predominant Th1 type to Th2 type lymphocytes, with production of anti-inflammatory cytokines. In conclusion, the evidences gathered until now should be taken into consideration for future studies aiming to explore the possible role of H. pylori infection on human reproduction.

Green tea polyphenols ameliorate non-alcoholic fatty liver disease through upregulating AMPK activation in high fat fed Zucker fatty rats

AIM To investigate protective effects and molecular mechanisms of green tea polyphenols(GTP) on nonalcoholic fatty liver disease(NAFLD) in Zucker fatty(ZF) *** Male ZF rats were fed a high-fat diet(HFD) for 2 wk then treated with GTP(200 mg/kg) or saline(5 m L/kg) for 8 wk, with Zucker lean rat as their control. At the end of experiment, serum and liver tissue were collected for measurement of metabolic parameters, alanine aminotransferase(ALT) and aspartate aminotransferase(AST), inflammatory cytokines and hepatic triglyceride and liver histology. Immunoblotting was used to detect phosphorylation of AMP-activated protein kinase(AMPK) acetyl-Co A carboxylase(ACC), and sterol regulatory element-binding protein 1c(SREBP1c). RESULTS Genetically obese ZF rats on a HFD presented with metabolic features of hepatic pathological changes comparable to human with NAFLD. GTP intervention decreased weight gain(10.1%, P = 0.052) and significantly lowered visceral fat(31.0%, P < 0.01). Compared with ZF-controls, GTP treatment significantly reduced fasting serum insulin, glucose and lipids levels. Reduction in serum ALT and AST levels(both P < 0.01) were observed in GTP-treated ZF rats. GTP treatment also attenuated the elevated TNFα and IL-6 in the circulation. The increased hepatic TG accumulation and cytoplasmic lipid droplet were attenuated by GTP treatment, associated with significantly increased expression of AMPK-Thr172(P < 0.05) and phosphorylated ACC and SREBP1c(both P < 0.05), indicating diminished hepatic lipogenesis and triglycerides out flux from liver in GTP treated rats. CONCLUSION The protective effects of GTP against HFD-induced NAFLD in genetically obese ZF rats are positively correlated to reduction in hepatic lipogenesis through upregulating the AMPK pathway.

Changes and Significance of cytokines in Serum of Rabbits with Acute Pulmonary Embolism

Steroidal alkaloids ameliorate cell proliferation,oxidative stress,inflammation and histology outcome in vitro and in vivo

Objective: To investigate the effects of steroidal alkaloids and sarcovagine D isolated from Sarcococca hookeriana var. digyna on cell-proliferation and secretion of inflammatory cytokines in TNF-a-induced human RA-FLS MH7 A in vitro, and against complete Freund's adjuvant(CFA)-induced arthritis rats in vivo. Methods: CCK-8 assay was utilized to evaluate the anti-proliferation activity in vitro. In in-vivo study, rats were randomly divided into control group, model group(CFA), steroidal alkaloids(STA) groups(5.0, 2.5 and 1.25 g/kg BW),and sarcovagine D(SD) groups(50, 100 and 200 mg/kg BW), 10 rats for each group. To evaluate the anti-inflammation effect, the histology, biochemical parameters and expression of inflammatory cytokines were detected. Results: Steroidal alkaloids and sarcovagine D showed strong anti-proliferative activity during MH7 A cell culture proliferation and downregulated NO levels, and inflammatory cytokines(IL-1β, IL-6 and PGE_2) in vitro. The rats treated with CFA induced ankle inflammation and lymphocyte infiltration, upregulated IL-1β, TNF-α,IL-6, PGE_2, MDA and NO levels, and downregulated IL-10 and SOD. On the contrary,steroidal alkaloids ameliorated all the changes induced by CFA in rats, suggesting the antiinflammatory effect of steroidal alkaloids and sarcovagine D. Conclusions: The results suggest that Sarcococca hookeriana var. digyna has anti-inflammatory effect. Steroidal alkaloids and sarcovagine D has the potential to cure RA.

Clinical study of warm needling moxibustion plus intra-articular injection of sodium hyaluronate for chondromalacia patellae

Chronic rhinitis and rhinosinusitis are among the most common conditions world-wide with significant morbidity and decreased quality of life. Although the pathogenesis of these conditions is multifactorial, there has been increasing evidence for the role of environmental factors such as aeroallergens and air pollutants as initiating or exacerbating factors. This review will outline the current literature focusing on the role of aeroallergens and air pollution in the pathogenesis of chronic sinonasal inflammatory conditions.

Guidelines for treatment of severe acute pancreatitis

Severe acute pancreatitis is a disease of rap- id onset and progression with high mortality and morbidity. A new concept suggests that it is multifac- torially induced and involves multi-links and that therapy must be directed to several more important links at the same time. The regimen of integrated traditional Chinese and western medicine acts on the same or different sites and links to inhibit or block the inflammatory cascades. Concomitantly, preven- tive measures were taken at the start of the treatment for preventing the progression of the disease, protec- ting the function of vital organs, and avoiding the oc- currence of complications. No mortality or serious complications occurred in a series of 38 patients with Balthazar's CT grading of D and E. The treatment regimen and the mechanisms of actions of these drugs were delineated in detail in the tables and figure; the regimen is highly recommended for clinical use.

Sepsis, as one of the leading causes of death globally, is characterized by a systemic inflammatory disorder combined with organ dysfunction as the presence of infection. According to the emergence of antibiotic-resistant bacteria, e.g., carbapenem-resistant Klebsiella pneumoniae(CRKP), the treatment for sepsis or septic shock in clinic becomes more intractable. Ivangustin is derived from Inula japonica and could inhibit the secretion of nitric oxide(NO) induced by LPS in Raw264.7 cells significantly. Secoeudesma sesquiterpenes lactones A(SESLA), the synthetic analog of ivangustin, was shown to have higher anti-inflammatory activity than ivangustin. Here, we investigated the anti-inflammatory role and the underlying molecular mechanism of SESLA in LPS or CRKP-evoked inflammation. The results indicated that SESLA inhibited the expression of inflammatory mediators such as IL-6, TNF-α, IL-1β, MCP-1/CCL2, IL-10, IL-12 p70 and NO in Raw264.7 cells, mouse peritoneal macrophages, immortalized bone marrow-derived macrophages, dendritic cells and human monocyte-like THP-1 upon stimulation with LPS or heat-killed CRKP(HK-CRKP). SESLA was shown to improve the sensitiveness of invalid antibiotics to some extent and alleviate mice from lethal dose of CRKP infection. PI3 K/Akt, NF-ΚB and ERK pathways accounted for the anti-inflammatory role of SESLA after LPS stimulation. According to the notable anti-inflammatory role in vitro and protective role in sepsis mouse model, SESLA in combination with antibiotics may offer great promise for the treatment of inflammatory diseases and sepsis, even caused by antibiotic-resistant bacteria.
