Experimental autoimmune neuritis(EAN) is the animal model of human acute inflammatory demyelinating polyradiculoneuropathies(AIDP), an auto-immune inflammatory demyelination disease of the peripheral nervous system(PNS) and the world’s leading cause of acute autoimmune neuromuscular paralysis. EAN and AIDP are characterized by self-limitation with spontaneous recovery;however, endogenous pathways that regulate inflammation resolution in EAN and AIDP remain elusive. Here, we demonstrated that resolvin D1(Rv D1)-enahnced macrophage phagocytosis of apoptotic T cells induced transforming growth factor-β(TGF-β) to promote inflammation resolution and disease recovery in EAN. We determined that Rv D1, its synthetic enzyme and its receptor were greatly increased in PNS during the recovery stage of EAN. Both endogenous and exogenous Rv D1 increased regulatory T(Treg) cell and anti-inflammatory macrophage counts in PNS, enhanced inflammation resolution and promoted disease recovery in EAN rats. Moreover, Rv D1 up-regulated TGF-β level and pharmacologic inhibition of TGF-β signaling suppressed Rv D1-induced Treg cell count, but not anti-inflammatory macrophage count, and Rv D1-improved inflammation resolution and disease recovery in EAN rats. Mechanistically, Rv D1 promoted macrophage phagocytosis of apoptotic T cells in PNS, leading to reduced apoptotic T cell accumulation in PNS, up-regulated TGF-β by macrophages, increased Treg cell count in PNS and inflammation resolution in EAN rats. Thus, these data highlight the crucial role of Rv D1 as an important proresolving molecule in EAN and suggest that it has potential as a therapeutic target in human neuritis.

Nanomedicines for the treatment of rheumatoid arthritis:State of art and potential therapeutic strategies

Increasing understanding of the pathogenesis of rheumatoid arthritis(RA)has remarkably promoted the development of effective therapeutic regimens of ***,the inadequate response to current therapies in a proportion of patients,the systemic toxicity accompanied by longterm administration or distribution in non-targeted sites and the comprised efficacy caused by undesirable bioavailability,are still unsettled problems lying across the full remission of *** far,these existing limitations have inspired comprehensive academic researches on nanomedicines for RA treatment.A variety of versatile nanocarriers with controllable physicochemical properties,tailorable drug release pattern or active targeting ability were fabricated to enhance the drug delivery efficiency in RA *** review aims to provide an up-to-date progress regarding to RA treatment using nanomedicines in the last 5 years and concisely discuss the potential application of several newly emerged therapeutic strategies such as inducing the antigen-specific tolerance,pro-resolving therapy or regulating the immunometabolism for RA treatments.

Maresin1(MaR1),a new anti-inflammatory and pro-resolving lipid mediator,has been proven to exert organprotective effects in septic animal ***,the potential mechanisms are still not fully *** this study,we sought to explore the impact of MaR1 on metabolic dysfunction in cecal ligation and puncture(CLP) induced septic *** found that MaR1 significantly increased overall survival rate and attenuated lung and liver injuries in septic *** addition,MaR1 markedly reduced the levels of pro-inflammatory cytokines(TNF-α and IL-6) and alleviated mitochondrial *** on a 1 H NMR-based metabolomics analysis,CLP-induced septic mice had increased levels of acetate,pyruvate and lactate in serum and decreased levels of alanine,aspartate,glutamate and fumarate in ***,these metabolic disorders,mainly involving energy and amino acid metabolism,can be recovered by MaR1 ***,our results suggest that the protective effects of MaR1 on sepsis could be related to the recovery of metabolic dysfunction and the alleviation of inflammation and mitochondrial damage.

Efferocytosis refers to the process that phagocytes recognize and remove the apoptotic cells, which is essential for maintaining tissue homeostasis both in physiological and pathological conditions. Numerous studies have demonstrated that efferocytosis can prevent secondary necrosis and proinflammatory factor release, leading to the resolution of inflammation and tissue immunological tolerance in numerous diseases such as stroke. Stroke is a leading cause of death and morbidity for adults worldwide. Persistent inflammation triggered by the dead cells or cell debris is a major contributor to post-stroke brain damage. Effective efferocytosis might be an efficient strategy to minimize inflammation and restore brain homeostasis for neuronal regeneration and function recovery. In this review, we will discuss the phagocytes in the brain, the molecular mechanisms underlying efferocytosis, the role of efferocytosis in inflammation resolution, and the potential therapeutic applications targeting efferocytosis in stroke.
