Seropharmacological Study on Osteogenic Effects of Post-Absorption Ingredients of An Osteoprotective herbal formula

Objective： To further investigate the in vitro effects of an osteoprotective herbal formula ＂ELP＂（Herba Epimedii, Fructus Ligustri Lucidi and Fructus Psora/eae） using seropharmacological approach. Methods： Rats were fed with ELP or its individual component herbs for 2 days. The serum containing the post-absorbed ingredients of the herbal items were collected for cell culture using UMR106 cell, RAW264.7 cell and mesenchymal stem cell （MSC） isolated from the bone marrow of the rats. The effects of the herbal-containing serum on cell toxicity were detected by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay; bromodeoxyuridine assay was conducted to measure the cell proliferation of UMR106 cell and MSC; cell activity was measured using colorimetric method, and mRNA expression of runt-related transcription factor 2 （Runx2）, alkaline phosphatase （ALP） and osteopontin （OPN） of UMR106 and MSC as well as matrix metalloproteinase 9 （MMP-9）, tartrate-resistant acid phosphatase （TRAP） and cathepsin K of RAW264.7 were analyzed using real-time reverse-transcription polymerase chain reaction. Results： ELP and its component serum exhibited no cytotoxic effects on the cells. The ELP-containing serum increased the proliferation of UMR106 cell and MSC by 25.7% and 14.4%, respectively and the alkaline phosphatase activity of MSC was increased by 42.6%. On the contrary, it inhibited the RAW264.7 cell differentiation by 29.2%. ELP serum upregulated the Runx2 expression of UMR and MSC by 1.18 fold and 1.27 fold, respectively. It also upregulated ALP and OPN expression in MSC by 1.69- and 2.12-fold, respectively. On the other hand, ELP serum down-regulated MMP-9 and cathepsin K expression of RAW264.7 cell by 0.46- and 0.36-fold, respectively. Conclusions： The serum of the animals fed with ELP contains active ingredients which are effective in promoting osteogenesis and inhibiting osteoclastogenesis.

Polyherbal formula SC-E3 inhibits rheumatoid arthritis activity in a mouse model of type-Ⅱcollagen-induced arthritis

Objective:SC-E3 is a polyherbal formula that contains five medicinal herbs used frequently in traditional herbal *** our previous study,we demonstrated the antioxidant and anti-inflammatory effects of *** present study examined the effects of SC-E3 in a mouse model of type-II collagen-induced arthritis(CIA).Methods:In vivo,male DBA/1 J mice were immunized by intradermal injection of bovine type-II collagen and complete or incomplete Freund’s adjuvant,to induce ***-E3 was orally administered daily for 23 *** vitro,bone marrow-derived macrophages(BMMs)were treated with macrophage colony-stimulating factor(M-CSF)and receptor activator of nuclear factor-j B ligand(RANKL)in the absence or presence of ***:Administrations of SC-E3 were found to have anti-arthritic effects in the joints of CIA mice,as evidenced by reduced paw swelling,bone erosion and deformation,inflammatory cell infiltration,and inflammation in synovial ***-E3 also reduced serum levels of tumor necrosis factor-a,interleukin-1 b,aspartate aminotransferase and alanine ***,tartrateresistant acid phosphatase-positive osteoclast numbers in the joints were significantly lower in SC-E3-treated CIA mice than in CIA *** addition,the differentiations of BMMs to multinucleated osteoclasts induced by M-CSF and RANKL stimulation were dose-dependently reduced by ***:These results suggest that SC-E3 possesses substantial anti-arthritic activity because it inhibits pro-inflammatory cytokines and osteoclastogenesis,and that SC-E3 has potential therapeutic use for the treatment of rheumatoid arthritis.

Antioxidant and antiaging effect of traditional Thai rejuvenation medicines in Caenorhabditis elegans

Objective: This study explored the rejuvenation mechanisms of Thai polyherbal medicines using different approaches, including in vitro methods, as well as a well-defined nematode model, Caenorhabditis ***: THP-R-SR012 decoction was selected from 23 polyherbal medicines, based on metal-chelating and chain-breaking antioxidant capacities. The influences of this extract on the survival and some stress biomarkers of C. elegans under paraquat-induced oxidative stress were evaluated. Furthermore, lifespan analysis and levels of lipofuscin accumulation were examined in senescent nematodes. The phytochemical profile of THP-R-SR012 was ***: Supplementation with THP-R-SR012 decoction significantly increased the mean lifespan and reduced the oxidative damage to C. elegans under oxidative stress conditions. Further, THP-R-SR012 supplementation slightly influenced the lifespan and the level of lipofuscin accumulation during ***-related phytochemical constituents of THP-R-SR012 decoction were rutin, naringenin, 3,4-dihydroxybenzoic acid, gallic acid, glycyrrhizic acid, demethoxycurcumin and 18 a-glycyrrhetinic ***: The antioxidant potential of THP-R-SR012 was due to its scavenging properties, its enhancement of antioxidant-related enzyme activities, and the presence of the antioxidant-related *** results support the traditional use of THP-R-SR012 decoction as a tonic for nourishing and strengthening the whole body.

Changes of Anti-tuberculosis Herbs formula during Past Three Decades in Contrast to Ancient Ones

Objective:To investigate the evolution of herbal medicine in treating tuberculosis(TB)and encourage anti-TB drug discovery and ***:In this study,477 ancient traditional Chinese medicine formulae were collected from the Dictionary of Traditional Chinese Medicine Prescriptions and 172 modern Chinese medicine formulae(from 1986 to 2016)were collected by searching 4 databases:Wan Fang Data Knowledge Service Platform,China National Knowledge Infrastructure Database(CNKI),China Science and Technology Journal Database(VIP)and Chinese Bio-medical Literature and Retrieval System(Sino Med)in *** restricted the search to publications in *** data analysis was done using the Traditional Chinese Medicine Inheritance Support System version 2 ***:There were 425 herbs in the 477 ancient formulae and 257 herbs in the 172 modern *** of the top 30 herbs were shared by both modern and ancient *** are Radix Ophiopogonis,Astragalus membranaceus,Fritillaria cirrhosa,Dried rehmannia glutinosa,Poria cocos,Angelica sinensis,Prepared rehmannia glutinosa,Platycodon Root,Radix paeoniae alba,Schisandra chinensis,Bighead atractylodes rhizome,Rhizoma anemarrhenae,Cortex lycii radicis and Radix *** two groups of herbs with a high correlation coefficient were found in both modern and ancient prescriptions,the Dried rehmannia glutinosa with Radix ophiopogonis,and Radix ophiopogonis with Prepared rehmannia *** were 9 and 15 core herb combinations in modern and ancient prescriptions,respectively,but no one was found simutaniously in both modern and ancient ***:Although there were wide variations in the herb groups and herb combinations in the formulae,half of the top 30 herbs were found in both modern and ancient *** core herb combinations in modern and ancient prescriptions could help us to improve the priscription for treatment of TB.

Introduction: Bu-Shen-Ning-Xin decoction(BSNXD) is a traditional Chinese medicinal composition that has been used as a remedy for postmenopausal osteoporosis, but the mechanisms affecting bone metabolism are not fully ***: We investigated the molecular mechanism and signaling pathway underlying the effect of BSNXD on *** and Methods: A postmenopausal osteoporosis animal model generated by ovariectomywas administered BSNXD and drug-derived serum was prepared. An enzyme immunoassay was conducted to measure the 17-β-estradiol(E2) concentration in the drug-derived serum. Bone marrow-derived monocyte/macrophage precursor cells were treated with drug-derived serum, and tartrate-resistance acid phosphatase staining was conducted to observe osteoclastogenesis. A bone resorption assay was performed to analyze the effect on osteoclastic resorptive ***-time PCR, flow cytometry, Western blotting, transfection, and luciferase assays were conducted to explore the related ***: E2 was not elevated in BSNXD-derived serum. BSNXD-derived serum suppressed receptor activation of nuclear factor κB ligand(RANKL)-activated osteoclastogenesis in a dose-dependent manner;this effect could be reversed by estrogen receptor α antagonist methyl-piperidino-pyrazole. The serum suppressed RANKL-induced NF-κB transcription and inhibited the accumulation of nuclear factor of activated T-cells, cytoplasmic 1 in osteoclast precursor cells;the inhibitory effect was abolished by methyl-piperidino-pyrazole but not the estrogen receptor β antagonist or androgen receptor ***: These results collectively suggest that administration of BSNXD presents inhibitoryeffects on osteoclast differentiation by abrogating the RANKL-induced nuclear factor of activated T-cells, cytoplasmic 1 and NF-κB signaling pathways downstream of estrogen receptor α,thereby contributing to the inhibitory effect on bone resorption.

通过长期毒性试验和急性毒性试验,观察抗抑郁中药复方开心散对Wistar大鼠和昆明种小鼠所发生的各种毒性反应,评价其安全性。方法:昆明小鼠急性毒性试验:给药途径为灌胃,两次灌胃间隔8小时。试验分成60.04、48.03、38.42、30.74、24.59、19.67g/kg 6个剂量组,每组20只动物,雌雄各半,试验观察7天。长期毒性试验中,Wistar大鼠分为4组,每组动物30只,雌雄各半,试验分为对照组、低剂量组1g/kg、中剂量组3g/kg、高剂量组9g/kg。大鼠给药途径为灌胃,每周给药6次,给药期为90天,停药后观察30天,分别检测体重、血象、血清生化指标、脏器系数等。结果:开心散在9g/kg剂量组对大鼠有一定的长期毒性反应,但是在停药30天后各项指标恢复正常,说明对机体的损伤是可逆的。急性毒性试验表明小鼠的LD为32.59g/kg,NOAEL值为19.67g/kg/day。结论:开心散口服给药对正常大鼠的一般行为无明显影响。长期高剂量口服给药对大鼠有轻度的可逆性损伤,长期低剂量口服给药对大鼠无损伤.本实验表明开心散停药后对大鼠无延迟毒性作用,对正常大鼠总体是基本安全的。
