glycogen serves as the principal energy reserve for metabolic processes in aquatic shellfish and substantially contributes to the flavor and quality of oysters.The Jinjiang oyster(Crassostrea ariakensis)is an economically and ecologically important species in China.In the present study,RNA sequencing(RNA-seq)and assay for transposase-accessible chromatin using sequencing(ATAC-seq)were performed to investigate gene expression and chromatin accessibility variations in oysters with different glycogen contents.Analysis identified 9483 differentially expressed genes(DEGs)and 7215 genes with significantly differential chromatin accessibility(DCAGs)were obtained,with an overlap of 2600 genes between them.Notably,a significant proportion of these genes were enriched in pathways related to glycogen metabolism,including“glycogen metabolic process”and“Starch and sucrose metabolism”.In addition,genome-wide association study(GWAS)identified 526 single nucleotide polymorphism(SNP)loci associated with glycogen content.These loci corresponded to 241 genes,63 of which were categorized as both DEGs and DCAGs.This study enriches basic research data and provides insights into the molecular mechanisms underlying the regulation of glycogen metabolism in C.ariakensis.

glycogen hepatopathy in type-1 diabetes mellitus: A case report

BACKGROUND It has been studied that fluctuating glucose levels may superimpose glycated hemoglobin in determining the risk for diabetes mellitus(DM)complications.While non-alcoholic steatohepatitis(NASH)remains a predominant cause of elevated transaminases in Type 2 DM due to a strong underplay of metabolic syndrome,Type 1 DM can contrastingly affect the liver in a direct,benign,and reversible manner,causing glycogen hepatopathy(GH)-with a good prognosis.CASE SUMMARY A 50-year-old female with history of poorly controlled type 1 DM,status post cholecystectomy several years ago,and obesity presented with nausea,vomiting,and abdominal pain.Her vitals at the time of admission were stable.On physical examination,she had diffuse abdominal tenderness.Her finger-stick glucose was 612 mg/dL with elevated ketones and low bicarbonate.Her labs revealed abnormal liver studies:AST 1460 U/L,ALP:682 U/L,ALP:569 U/L,total bilirubin:0.3mg/dL,normal total protein,albumin,and prothrombin time/international normalized ratio(PT/INR).A magnetic resonance cholangiopancreatography(MRCP)demonstrated mild intra and extra-hepatic biliary ductal dilation without evidence of choledocholithiasis.She subsequently underwent a diagnostic ERCP which showed a moderately dilated CBD,for which a stent was placed.Studies for viral hepatitis,Wilson’s Disease,alpha-1-antitrypsin,and iron panel came back normal.Due to waxing and waning transaminases during the hospital course,a liver biopsy was eventually done,revealing slightly enlarged hepatocytes that were PAS-positive,suggestive of glycogenic hepatopathy.With treatment of hyperglycemia and ensuing strict glycemic control,her transaminases improved,and she was discharged.CONCLUSION With a negative hepatocellular and cholestatic work-up,our patient likely had GH,a close differential for NASH but a poorly recognized entity.GH,first described in 1930 as a component of Mauriac syndrome,is believed to be due to glucose and insulin levels fluctuation.Dual echo magnetic resonance imaging sequencing and compu

glycogen plays essential roles in glucose metabolism.Imaging glycogen in the liver,the major glycogen reservoir in the body,may shed new light on many metabolic disorders.^(13)C magnetic resonance spectroscopy(MRS)has become the mainstream method for monitoring glycogen in the body.However,the equipment of special hard-ware to standard clinical magnetic resonance imaging(MRI)scanners limits its clinical applications.Herein,we utilized endogenous glycogen as a T_(2)-based relaxation contrast agent for imaging glycogen metabolism in the liver in vivo.The in vitro results demonstrated that the transverse relaxation rate of glycogen strongly correlates with the concentration,pH,and field strength.Based on the Swift-Connick theory,we characterized the exchange property of glycogen and measured the exchange rate of glycogen as 31,847 Hz at 37°C.Besides,the viscosity and echo spacing showed no apparent effect on the transverse relaxation rate.This unique feature enables vi-sualization of glycogen signaling in vivo through T_(2)-weighted MRI.Two hours-post intraperitoneal injection of glucagon,a clinical drug to promote glycogenolysis and gluconeogenesis,the signal intensity of the mice’s liver increased by 1.8 times from the T_(2)-weighted imaging experiment due to the decomposition of glycogen.This study provides a convenient imaging strategy to non-invasively investigate glycogen metabolism in the liver,which may find clinical applications in metabolic diseases.

The role of glycogen phosphorylase in glycogen biogenesis in skeletal muscle after exercise

Initially it was believed that phosphorylase was responsible for both glycogen breakdown and synthesis in the living cell.The discovery of glycogen synthase and McArdle's disease(lack of phosphorylase activity),together with the high Pi/glucose 1-P ratio in skeletal muscle,demonstrated that glycogen synthesis could not be attributed to reversal of the phosphorylase reaction.Rather,glycogen synthesis was attributable solely to the activity of glycogen synthase,subsequent to the transport of glucose into the cell.However,the well-established observation that phosphorylase was inactivated(i.e.,dephosphorylated)during the initial recovery period after prior exercise,when the rate of glycogen accumulation is highest and independent of insulin,suggested that phosphorylase could play an active role in glycogen accumulation.But the quantitative contribution of phosphorylase inactivation was not established until recently,when studying isolated murine muscle preparations during recovery from repeated contractions at temperatures ranging from 25 to 35C.Thus,in both slow-twitch,oxidative and fast-twitch,glycolytic muscles,inactivation of phosphorylase accounted for 45%–75%of glycogen accumulation during the initial hours of recovery following repeated contractions.Such data indicate that phosphorylase inactivation may be the most important mechanism for glycogen accumulation under defined conditions.These results support the initial belief that phosphorylase plays a quantitative role in glycogen formation in the living cell.However,the mechanism is not via activation of phosphorylase,but rather via inactivation of the enzyme.

Redox Regulation of glycogen Biosynthesis in the Cyanobacterium Synechocystis sp. PCC 6803： Analysis of the AGP and glycogen Synthases

glycogen constitutes the major carbon storage source in cyanobacteria, as starch in algae and higher plants. glycogen and starch synthesis is linked to active photosynthesis and both of them are degraded to glucose in the dark to maintain cell metabolism. Control of glycogen biosynthesis in cyanobacteria could be mediated by the regulation of the enzymes involved in this process, ADP-glucose pyrophosphorylase （AGP） and glycogen synthase, which were identified as putative thioredoxin targets. We have analyzed whether both enzymes were subjected to redox modification using purified recombinant enzymes or cell extracts in the model cyanobacterium Synechocystis sp. PCC 6803. Our results indicate that both AGP and glycogen synthases are sensitive to copper oxidation. However, only AGP exhibits a decrease in its enzymatic activity, which is recovered after reduction by DTT or reduced thioredoxin （TrxA）, suggesting a redox control of AGP. In order to elucidate the role in redox control of the cysteine residues present on the AGP sequence （C45, C185, C320, and C337）, they were replaced with serine. All AGP mutant proteins remained active when expressed in Synechocystis, although they showed different electrophoretic mobility profiles after copper oxidation, reflecting a complex pattern of cysteines interaction.

Spleen-kidney supplementing formula alleviates insulin resistance via regulating AKT/glycogen synthase kinase 3β pathway in rats with type 2 diabetic induced by high-fat diet

OBJECTIVE: To explore the molecular mechanism underpinning the action by investigating its effect on glycogen content and AKT(also known as protein kinase B)/glycogen synthase kinase 3β(GSK-3β) pathway in the liver of rats with type 2 diabetic induced by high-fat diet.METHODS: The rat model of type 2 diabetes was induced by high-fat diet and multiple low-dose streptozotocin injection. Diabetic rats were divided into five groups: the model control group, the Metformin group, spleen-kidney supplementing formula groups of low, medium and high doses. Fasting blood glucose(FBG) levels were measured before treatment and every two weeks during treatment.After the treatment, oral glucose tolerance test was performed, and hemoglobin A1c (HbA1c) and C-peptide were measured to assess the formula's effect on glucose metabolism and insulin resistance. The protein expression levels of AKT, GSK-3βand their phosphorylated forms in the liver were also measured to study the formula's role in insulin signaling pathway.RESULTS: Spleen-kidney supplementing formula significantly relieved the symptoms of polydipsia,polyuria and weight loss in type 2 diabetic rats, reduced FBG and HbA1c levels, increased glycogen content, and improved insulin sensitivity. The anti-diabetic effects of spleen-kidney supplementing formula are dose dependent. It also increased the total AKT protein level and the GSK-3β phosphorylation in the liver of type 2 diabetic rats.CONCLUSION: Spleen-kidney supplementing formula has hypoglycemic effect and relieves insulin resistance by enhancing AKT/GSK-3β signaling pathway in the liver of type 2 diabetic rats.

Relationship Between Single Nucleotide Polymorphism of glycogen Synthase Gene of Pacific Oyster Crassostrea gigas and Its glycogen Content

glycogen is important not only for the energy supplementary of oysters, but also for human consumption. High glycogen content can improve the stress survival of oyster. A key enzyme in glycogenesis is glycogen synthase that is encoded by glycogen synthase gene GYS. In this study, the relationship between single nucleotide polymorphisms(SNPs) in coding regions of Crassostrea gigas GYS(Cg-GYS) and individual glycogen content was investigated with 321 individuals from five full-sib families. Single-strand conformation polymorphism(SSCP) procedure was combined with sequencing to confirm individual SNP genotypes of Cg-GYS. Least-square analysis of variance was performed to assess the relationship of variation in glycogen content of C. gigas with single SNP genotype and SNP haplotype. As a consequence, six SNPs were found in coding regions to be significantly associated with glycogen content(P glycogen content(P glycogen content and provided molecular biological information for the selective breeding of good quality traits of C. gigas.

Research on polyhydroxyalkanoates and glycogen transformations: Key aspects to biologic nitrogen and phosphorus removal in low dissolved oxygen systems

In this paper,a study was conducted on the effect of polyhydroxyalkanoates(PHA)and glycogen transformations on biologic nitrogen and phosphorus removal in low dissolved oxygen(DO)systems.Two laboratory-scale sequencing batch reactors(SBR1 and SBR2)were operating with anaerobic/aerobic(low DO,0.15-0.45 mg·L^(-1))configurations,which cultured a propionic to acetic acid ratio(molar carbon ratio)of 1.0 and 2.0,respectively.Fewer poly-3-hydroxybutyrate(PHB),total PHA,and glycogen transformations were observed with the increase of propionic/acetic acid,along with more poly-3-hydroxyvalerate(PHV)and poly-3-hydroxy-2-methyvalerate(PH2MV)shifts.The total nitrogen(TN)removal efficiency was 68%and 82%in SBR1 and SBR2,respectively.In the two SBRs,the soluble ortho-phosphate(SOP)removal efficiency was 94%and 99%,and the average sludge polyphosphate(poly-P)content(g·g-MLVSS^(-1))was 8.3%and 10.2%,respectively.Thus,the propionic to acetic acid ratio of the influent greatly influenced the PHA form and quantity,glycogen transformation,and poly-P contained in activated sludge and further determined TN and SOP removal efficiency.Moreover,significant correlations between the SOP removal rate and the(PHV+PH2MV)/PHA ratio were observed(R^(2)>0.99).Accordingly,PHA and glycogen transformations should be taken into account as key components for optimizing anaerobic/aerobic(low DO)biologic nitrogen and phosphorus removal systems.

3D-QSAR studies on glycogen phosphorylase inhibitors by flexible comparative molecular field analysis

Canceling grids accommodating probes in comparative molecular field analysis (CoMFA), the idea of flexibleness is introduced into the CoMFA, and in combination with swarm intelligent algorithm which attempts to optimize distributions of diverse probes around drug molecules, a new 3D-QSAR method is proposed in this context as flexible comparative molecular field analysis (FCoMFA). In preliminary at-tempts to performing QSAR studies on 47 glycogen phosphorylase inhibitors, FCoMFA is employed and confirmed to be potent to exploring ligand-receptor interaction manners at active positions and thus to generating stable and predictable models. Simultaneously by an intuitive graphics regarding probe distribution patterns, impacts of different substituted groups on activities is also given an insight into.

Multiple endocrine factors regulate nutrient mobilization and storage in Aedes aegypti during a gonadotrophic cycle

Anautogenous mosquitoes must blood feed on a vertebrate host to produce eggs.Each gonadotrophic cycle is subdivided into a sugar-feeding previtellogenic phase that produces primary follicles and a blood meal-activated vitellogenic phase in which large numbers of eggs synchronously mature and are laid.Multiple endocrine factors including juvenile hormone(JH),insulin-like peptides(ILPs),ovary ecdysteroidogenic hormone(OEH),and 20-hydroxyecdysone(20E)coordinate each gonadotrophic cycle.Egg formation also requires nutrients from feeding that are stored in the fat body.Regulation of egg formation is best understood in Aedes aegypti but the role different endocrine factors play in regulating nutrient mobilization and storage remains unclear.In this study,we report that adult female Ae.aegypti maintained triacylglycerol(TAG)stores during the previtellogenic phase of the first gonadotrophic cycle while glycogen stores declined.In contrast,TAG and glycogen stores were rapidly mobilized during the vitellogenic phase and then replenishment.Several genes encoding enzymes with functions in TAG and glycogen metabolism were differentially expressed in the fat body,which suggested regulation was mediated in part at the transcriptional level.Gain of function assays indicated that stored nutrients were primarily mobilized by adipokinetic hormone(AKH)while juvenoids and OEH regulated replenishment.ILP3 further showed evidence of negatively regulating certain lipolytic enzymes.Loss of function assays indicated AKH depends on the AKH receptor(AKHR)for function.Altogether,our results indicate that the opposing activities of different hormones regulate nutrient stores during a gonadotrophic cycle in Ae.aegypti.
