Fenton metal nanomedicines for imaging-guided combinatorial chemodynamic therapy against cancer

Chemodynamic therapy(CDT)is considered as a promising modality for selective cancer therapy,which is realized via Fenton reaction-mediated decomposition of endogenous H_(2)O_(2) to produce toxic hydroxyl radical(•OH)for tumor ablation.While extensive efforts have been made to develop CDT-based therapeutics,their in vivo efficacy is usually unsatisfactory due to poor catalytic activity limited by tumor microenvironment,such as anti-oxidative systems,insufficient H_(2)O_(2),and mild acidity.To mitigate these issues,we have witnessed a surge in the development of CDT-based combinatorial nanomedicines with complementary or synergistic mechanisms for enhanced tumor therapy.By virtue of their bio-imaging capabilities,Fenton metal nanomedicines(FMNs)are equipped with intrinsic properties of imaging-guided tumor therapies.In this critical review,we summarize recent progress of this field,focusing on FMNs for imaging-guided combinatorial tumor therapy.First,various Fenton metals with inherent catalytic performances and imaging properties,including Fe,Cu and Mn,were introduced to illustrate their possible applications for tumor theranostics.Then,CDT-based combinatorial systems were reviewed by incorporating many other treatment means,including chemotherapy,photodynamic therapy(PDT),sonodynamic therapy(SDT),photothermal therapy(PTT),starvation therapy and immunotherapy.Next,various imaging approaches based on Fenton metals were presented in detail.Finally,challenges are discussed,and future prospects are speculated in the field to pave way for future developments.

Taxol is a 'blockbuster' antitumor drug produced by Taxus species with extremely low amount, while its analogue 7-β-xylosyl-10-deacetyltaxol is generally much higher in the plants. Both the fungal enzymes LXYL-P1à1 and LXYL-P1à2 can convert 7-β-xylosyl-10-deacetyltaxol into10-deacetyltaxol for Taxol semi-synthesis. Of them, LXYL-P1à2 is twice more active than LXYLP1à1, but there are only 11 significantly different amino acids in terms of the polarity and acidic-basic properties between them. In this study, single and multiple site-directed mutations at the 11 sites from LXYL-P1à1 to LXYL-P1à2 were performed to define the amino acids with upward bias in activities and to acquire variants with improved catalytic properties. Among all the 17 mutants, E12(A72 T/V91 S) was the most active and even displayed 2.8-and 3-fold higher than LXYL-P1à2 on β-xylosidase andβ-glucosidase activities. The possible mechanism for such improvement was proposed by homology modeling and molecular docking between E12 and 7-β-xylosyl-10-deacetyltaxol. The recombinant yeast GS115-P1 E12-7 was constructed by introducing variant E12, the molecular chaperone gene pdi and the bacterial hemoglobin gene vhb. This engineered yeast rendered 4 times higher biomass enzyme activity than GS115-3.5 K-P1à2 that had been used for demo-scale fermentation. Thus, GS115-P1 E12-7 becomes a promising candidate to replace GS115-3.5 K-P1à2 for industrial purpose.

To solve the problem of how to determine the non-scaled interval when processing radar clutter using fractal Brownian motion (FBM) model, a concept of combinatorial FBM model is presented. Since the earth (or sea) surface varies diversely with space, a radar clutter contains several fractal structures, which coexist on all scales. Taking the combination of two FBMs into account, via theoretical derivation we establish a combinatorial FBM model and present a method to estimate its fractal parameters. The correctness of the model and the method is proved by simulation experiments and computation of practial data. Furthermore, we obtain the relationship between fractal parameters when processing combinatorial model with a single FBM model. Meanwhile, by theoretical analysis it is concluded that when combinatorial model is observed on different scales, one of the fractal structures is more obvious.

Since the first successful story was reported in the middle of 1990s,combinatorial materials science has attracted more and more attentions in the materials community.In the past two decades,a great amount of effort has been made to develop combinatorial high-throughput approaches for materials research.However,few high-throughput mechanical characterization methods and tools were reported.To date,a number of micro-scale mechanical characterization tools have been developed,which provided a basis for combinatorial high-throughput mechanical characterization.Many existing micro-mechanical testing apparatuses can be pertinently modified for high-throughput characterization.For example,automated scanning nanoindentation is used for measuring the hardness and elastic modulus of diffusion multiple alloy samples,and cantilever beam arrays are used to parallelly characterize the thermal mechanical behavior of thin films with wide composition gradients.The interpretation of micro-mechanical testing data from thin films and micro-scale samples is most critical and challenging,as the mechanical properties of their bulk counterparts cannot be intuitively extrapolated due to the well-known size and microstructure dependence.Nevertheless,high-throughput mechanical characterization data from combinatorial micro-scale samples still reflect the dependence trend of the mechanical properties on compositions and microstructure,which facilitates the understanding of intrinsic materials behavior and the fast screening of bulk mechanical properties.After the promising compositions and microstructure are pinned down,bulk samples can be prepared to measure the accurate properties and verify the combinatorial high-throughput characterization results.By developing combinatorial high-throughput mechanical characterization methods and tools,in combination with high-throughput synthesis,the structural materials research would be promoted by accelerating the discovery,development,and deployment of high performance structural materials,and by

This paper presents a novel regular Quasi-Cyclic (QC)Low Density Parity Check (LDPC)codes with columnweight three and girth at least eight.These are designed on the basis of combinatorial design in which subsets applied for the construction of circulant matrices are determined by a particular subset.Considering the nonexistence of cycles four and six in the structure of the parity check matrix,a bound for their minimum weight is proposed.The simtdations conducted confirm that without applying a masking technique,the newly implemented codes have a performance similar to or better than other well-known codes.This is evident in the waterfall region, while their error floor at very low Bit Error Rate (BER)is expected.

Objective:To evaluate simultaneous detection and differentiates of Brucella abortus(B.abortus) and Brucella melitensis(B.melitensis) through the combinatorial POR method.Methods:This study was designed using three primers that could simultaneously identify and differentiate two major species of pathogenic Brucella in humans and animals.Identification and differentiation of each species using the size of the PCR product were determined.To determine the specifieity of the method,bacteria close to the genus Brucella were used.Finally,to confirm PCR products.In addition to the products sequence,RFLP was performed on PCR products using restriction enzymes.Results:The method of optimized combinatorial PCR in this study could simultaneously detect and differentiate B.abortus and B.melitensis with high specificity and sensitivity in clinical samples.Differentiation of species is based on the resulting bands: therefore,the band 494 bp for B.abortus and 733 bp for B.melitensis were obtained.RFLP and sequencing results confirmed PCR results.Conclusions:The results of this study shows that without routine diagnostic methods such as culture and serology tests,using the molecular method of combinatorial PCR,important species of Brucella can be simultaneously identified and differentiated in clinical samples.

Based on the binding mode for hydrouracil in photosystem II (PS II) D1 protein, a general method for the solid phase combinatorial synthesis of hydrouracil library was developed. The acryloyl chloride or acid were coupled to Wang resin 1 to afford the acryloyl ester 2. Resin bound ester 2 reacted with various primary amines to give secondary amines 3, which were converted to the resin bound ureas 4 by treatment with isocyanates. Preparation of the hydrouracils 5 was achieved by the acidic cyclization_cleavage from the resin. For characterization purposes, 8 hydrouracil analogues were synthesized in parallel, following which a small library of 9 hydrouracil derivatives was prepared after cleavage from Wang resin. All the 9 bydrouracil derivatives desired were identified by GC_MS.

The option of organizing E-auctions to purchase electricity required for anticipated peak load period is a new one for utility companies.To meet the extra demand load,we develop electricity combinatorial reverse auction(CRA)for the purpose of procuring power from diverse energy sources.In this new,smart electricity market,suppliers of different scales can participate,and homeowners may even take an active role.In our CRA,an item,which is subject to several trading constraints,denotes a time slot that has two conflicting attributes,electricity quantity and price.To secure electricity,we design our auction with two bidding rounds:round one is exclusively for variable energy,and round two allows storage and nonintermittent renewable energy to bid on the remaining items.Our electricity auction leads to a complex winner determination(WD)task that we represent as a resource procurement optimization problem.We solve this problem using multi-objective genetic algorithms in order to find the trade-off solution that best lowers the price and increases the quantity.This solution consists of multiple winning suppliers,their prices,quantities and schedules.We validate our WD approach based on large-scale simulated datasets.We first assess the time-efficiency of our WD method,and we then compare it to well-known heuristic and exact WD techniques.In order to gain an exact idea about the accuracy of WD,we implement two famous exact algorithms for our constrained combinatorial procurement problem.
