The phenomenon of cytogenetic adaptive and cross-adaptive response induced by low dose irradiation and chemical mutagen in mice is described. We found, firstly, that adaptation can be induced by acute low dose Xirradiation (0-100 mGy). Secondly, a cross-adaptation can occur between X-irradiation and mitomycin C (MMC). And finally, mice pre-exposed to chronic low dose rate￣(60)Co-Gamma irradiation (0-226.0 mGy/day) are less susceptible to chromosome aberration induced by subsequent acute higher Xirradiation. Therefore, our data suggest that radioadaptive response depends on dose, dose rate and time interval. Possible mechanisms are also discussed.

本文对四名正常儿童和五名口服甲酸棉酚的男性的子代外周血淋巴细胞进行染色体畸变和SCE分析。结果表明:(1)5名服药者子代的染色体断裂、间隙和多倍体细胞均没有明显的变化。(2)他们的SCE平均值为5.90±0.40/细胞,SCE范围为1～12/细胞;对照组SCE平均值为5.06±0.13/细胞,SCE范围为2～9/细胞。两者间差别在统计学上无显著性意义(P>0.05)。

Objective It aims to study potential genotoxicity of almond *** A bacterial reverse mutation assay was performed on *** strains TA97,TA98,TA100,TA102,and TA1535 in the absence or presence of S-9 mixture at a dose range of 312.5 to 5 000 μg/plate.A micronucleus test and a mammalian bone marrow chromosome aberration tests were performed in Swiss Albino （CD-1） mice at doses of 625,1 250,and 2 500 mg/kg bw *** Almond skins exerted no mutagenic activity in various bacterial strains of Salmonella typhimurium in either the absence or the presence of metabolic activation at all doses *** doses of almond skins did not affect the proportions of immature to total erythrocytes,the number of micronuclei in the immature erythrocytes,or the number of structural and numerical chromosomal aberrations of Swiss albino *** Almond skins are not genotoxic under the conditions of the in vitro bacterial reverse mutation assay and two in vivo tests-micronucleus test and mammalian bone marrow chromosome aberration test,which supports the safety of almond skins for dietary consumption.

The effects of different concentrations of Zn￣(2+)ion on root growth,cell division,and nucleoli of Allium cepa were studied. The test Zn￣(2+) ion concentration was made up from zinc sulphate (ZnSO4. 7H2O) ranging from 10￣(-7) to 10￣(-2) mol/L. The solutions were prepared in tap water (pH =6. 5).The results indicated that Zn￣(2+) could obviously inhibit root growth at concentrations from 10￣(-4)to 10￣(-2) mol/*** treated with zinc sulphate showed the presence of c-mitosis, anaphase bridges,including sticky and fluidized bridges (at 10￣(-3) to 10￣(-2) mol/L) , chromosome stickiness, irregularly shaped nuclei, broken nuclei and micronuclei. A toxicity effect was also observed on the nucleoli using silver staining technique after 48h of treatment with 10￣(-4)to 10￣(-2) mol/L Zn￣(2+), e. g,the nucleolar particulate material scattered around the nucleoli in the nucleus of root tip cells.

Combination of Cytogenetic Analysis and Molecular Screening in Patients with de novo Acute Myeloid Leukemia

Nowadays the role of genetic findings in determining the diagnosis,therapy and prognosis of acute myeloid leukemia(AML) has become more *** improve and validate the detection of clonal chromosomal aberrations in leukemia,we designed a combined application of karyotyping with multiplex reverse transcription-polymerase chain reaction(RT-PCR) and fluorescence in situ hybridization(FISH),and addressed the expression and distribution of fusion genes among the subtypes of Chinese adult patients with de novo *** RT-PCR assays were performed on 477 samples from newly diagnosed AML patients,and cytogenetic data were obtained from 373 of them by R or G banding techniques and those in some cases were confirmed by *** PCR products in some suspected cases were tested by two-directional *** results showed that except unqualified samples,fusion genes were detected by multiplex RT-PCR in 211 of 474 patients(44.51%),including AML1-ETO,CBFβ-MYH11,PML-RARα,PLZF-RARα,NPM-RARα,MLL rearrangements,BCR-ABL,DEK-CAN,SET-CAN,TEL-PDGFR,TLS-ERG,AML1-MDS1(EVI-1).In 373 patients,who took both multiplex RT-PCR and karyotype analysis,the detection rate of chromosomal aberrations by using multiplex RT-PCR and karyotyping was 160/373(42.89%) and 179/373(47.98%) respectively,and the combination could optimize the detection rate of clonal genetic abnormalities to 216/373(57.90%).The PCR results from 11 cases 'normal' in karyotyping but abnormal in RT-PCR for MLL rearrangements were confirmed by two-directional *** is concluded that karyotype studies remain the cornerstone for genetic testing;conventional cytogenetics and molecular-based methods are complementary tests for the detection of clonal genetic aberrations in AML,especially for the cryptic or submicroscopic *** a genetic marker has been identified by combined analysis,it could be used to monitor residual disease during/after chemotherapy,by quantitative RT-PCR and/or FISH.

Background:Neuroblastoma(NB)is a heterogeneous disease with respect to genomic abnormalities and clinical *** recent advances in our understanding of the association between the genetic aberrations and clinical features,it remains one of the major challenges to predict prognosis and stratify patients for determining personalized therapy in this *** aim of this study was to develop an effective prognosis prediction model for NB ***:We integrated diverse computational analyses to define gene signatures that reflect MYCN activity and chromosomal aberrations including deletion of chromosome 1p(Chr1p_del)and chromosome 11q(Chr11q_del)as well as chromosome 11q whole loss(Chr11q_wls).We evaluated the prognostic and predictive values of these signatures in seven NB gene expression datasets(the number of samples ranges from 94 to 498,with a total of 2120)generated from both RNA sequencing and microarray ***:MYCN signature was a more effective prognostic marker than MYCN amplification status and MYCN ***,the Chr1p_del score was more prognostic than Chr1p *** activity scores of MYCN,Chr1p_del and Chr11q_del were associated with poor prognosis,while the Chr11q_wls score was linked to good *** integrated the activity scores of MYCN,Chr1p_del,Chr11q_del,and Chr11q_wls and clinical variables into an integrative prognostic model,which displayed significant performance over the clinical variables or each genomic aberration ***:Our integrative gene signature model shows a significantly improved forecast performance with prognostic and predictive information,and thereby can be served as a biomarker to stratify NB patients for prognosis evaluation and surveillance programs.

The peripheral blood lymphocyte chromosomes in a case of 60Co γ rays accident were examined at 2.5h after exposure. The frequency of the dicentrics plus centric rings was 89% and exposure dose was estimated to be 4.78 (4.53～4.88) Gy. The examinations of lymphocyte chromosome aberration within follow-up 12α showed that the incidence of Dic+R reduced with lg regression (r = -0.9895). While the number of cells with stable aberration remained unchanged and showed a tendency to increase. During the period of leukemia, bone marrow cell chromosome aberrations were studied by method of G-banding. Of 13 cells observed, 4 cells belonged to normal karyotypes. Among 9 aberration cells, 12 aberrations were detected. The majority of which were classified as translocation, deletion and inversion. Numeric aberrations were-9,-12,-20,-22,-y. This case suggested that acute lymphocytic leukemia was induced after radiation accident.
