Construction, Expression, and Characterization of a Recombinant Annexin B1-Low Molecular Weight Urokinase chimera in Escherichia coli

To produce a thrombi-targeting plasminogen activator, low molecular weight single-chain urokinase gene (scuPA32k) was spliced with the full-length cDNA of annexin B1 gene (anxB1) by overlap extension method. The fused gene anxB1scuPA was ligated into pET28a vector, transformed into E. coli BL21-RIL, and then induced to express under the control of T7 promoter. The AnxB1ScuPA protein ex- pressed amounted to 22% of the total bacterial proteins. The product was refolded, and then purified by using DEAE Sepharose fast flow ion-exchange column and Superdex S-200 gel-filtration column. HPLC analysis revealed that the final purity is about 95%. The specific activity of AnxB1ScuPA, measured as amidolytic activity, reached 100,000 IU/mg. It had a similar S2444 catalytic efficiency (kcat/Km) to ScuPA32k, and also showed high activated-platelet membrane-binding activity and anticoagulant activity, indicating that the chi- mera fully retained the components of enzymatic and membrane-binding activities of the parent molecules. In vivo test revealed that, the dogs administered with AnxB1ScuPA had less reperfusion time, higher reperfu- sion ratio, and less bleeding effects than those with urokinase. These findings indicated that AnxB1ScuPA might have advantages over current available thrombolytic agents.

chimera dynamics in nonlocally coupled moving phase oscillators

chimera states,a symmetry-breaking spatiotemporal pattern in nonlocally coupled dynamical units,prevail in a variety of systems.However,the interaction structures among oscillators are static in most of studies on chimera state.In this work,we consider a population of agents.Each agent carries a phase oscillator.We assume that agents perform Brownian motions on a ring and interact with each other with a kernel function dependent on the distance between them.When agents are motionless,the model allows for several dynamical states including two different chimera states (the type-Ⅰ and the type-Ⅱ chimeras).The movement of agents changes the relative positions among them and produces perpetual noise to impact on the model dynamics.We find that the response of the coupled phase oscillators to the movement of agents depends on both the phase lag α,determining the stabilities of chimera states,and the agent mobility D.For low mobility,the synchronous state transits to the type-Ⅰ chimera state for α close to π/2 and attracts other initial states otherwise.For intermediate mobility,the coupled oscillators randomly jump among different dynamical states and the jump dynamics depends on α.We investigate the statistical properties in these different dynamical regimes and present the scaling laws between the transient time and the mobility for low mobility and relations between the mean lifetimes of different dynamical states and the mobility for intermediate mobility.

Utilization of central disk of blastoderm and germinal crescent region of interspecific germline chimera between chicken and quail

OBJECTIVE:The production of interspecific germline chimeras between chicken and quail were attempted employing the dissociated cells derived from the blastodermal central disk (stage X) and the germinal crescent region of embryo (stage 7-8).;METHODS:The central disk (CD) of the area pellucida in chicken blastoderm (stage X) and the germinal crescent region (GCR) of embryo (stage 7-8) were dispersed and injected into the subgerminal cavity of quail blastoderm (stage X). Injected eggs were incubated for 7 days or to hatching. The donor chicken DNA was detected by the polymerase chain reaction.;RESULTS:In day-7 embryos, chicken DNA was detected in 5 gonads and 9 brains from 53 survived embryos received chicken CD cells, and 1 gonads and 6 brains from 27 survived embryos received chicken GCR. Chicken DNA was also detected from the semen of one adult male hatched from eggs received chicken GCR cells.;CONCLUSIONS:CD and GCR cells as the donors showed the possibility to produce the interspecific germline chimera, but further studies are needed to make necessary improvement.

Noise and delay sustained chimera state in small world neuronal network

chimera state in neuronal network means the coexistence of synchronized and desynchronized firing patterns. It attracts much attention recently due to its possible relevance to the phenomenon of unihemispheric sleep in mammals. In this paper, we search for chimera state in a noisy small-world neuronal network, in which the neurons are delayed coupled. We found both transient and permanent chimera state when time delay is close to a critical value. The chimera state occurs due to the competition between the synchronized and desynchronized patterns in the neuronal network. On the other hand, intermediate intensity of noise facilitates the occurrence of delay-sustained chimera states. Comparison between noise and delay shows that time delay is the key factor determining the chimera state, whereas noise is a subordinate one.

Giant teratoma with isolated intestinal duplication in adult: A case report and review of literature

BACKGROUND A combination of diseases is a rare phenomenon.Their clinical manifestations can vary,and the diagnosis can be challenging.Intestinal duplication is a rare congenital malformation,whereas retroperitoneal teratoma is a tumor in the retroperitoneal space,derived from the remaining embryonic tissue.There are relatively few clinical findings on adult retroperitoneal benign tumors.It is hard to believe that these two rare diseases can happen to the same person.CASE SUMMARY A 19-year-old woman complaining of abdominal pain with nausea and vomiting was admitted.Abdominal computed tomography angiography was suggested for invasive teratoma.Intraoperative exploration revealed that the giant teratoma was connected to an isolated intestinal tract in the retroperitoneum.The postoperative pathological examination revealed that mature giant teratoma was present with intestinal duplication.This was a rare intraoperative finding that was successfully treated surgically.CONCLUSION The clinical manifestations of intestinal duplication malformation are various,and difficult to diagnose before the operation.The possibility of intestinal replication should be considered when intraperitoneal cystic lesions are present.

At present, in order to improve the security capabilities of the civil navigation signal system, both Galileo and GPS have proposed a navigation signal authentication scheme. However, there is no civil official navigation signal authentication scheme for Beidou Navigation System, so it is necessary to propose a navigation signal authentication scheme suitable for Beidou signal system. This paper is mainly to prepare for the subsequent research on the navigation signal authentication scheme adapted to Beidou B1 C signal. chimera is applied to Beidou B1 C signal to study the impact of the dispersion of markers. In this paper, by setting other parameters constant, define the markers’ dispersion. Under circumstances of different dispersion, research the collision probability of the segment and the correlation performance of the encrypted ranging code. Draw the following conclusion that the dispersion of markers rarely affects the correlation performance. While the dispersion is higher, the collision probability of the segment will be greater. In the design of spreading code authentication scheme, it is sufficient to determine the dispersion of markers from the perspective of the collision probability of the segment.

Redesigning transcription factor Cre1 for alleviating carbon catabolite repression in Trichoderma reesei

Carbon catabolite repression(CCR),which is mainly mediated by Cre1 and triggered by glucose,leads to a decrease in cellulase production in Trichoderma reesei.Many studies have focused on modifying Cre1 for alleviating CCR.Based on the homologous alignment of CreA from wild-type Penicillium oxalicum 114–2(Po-0)and cellulase hyperproducer JUA10-1(Po-1),we constructed a C-terminus substitution strain—Po-2—with decreased transcriptional levels of cellulase and enhanced CCR.Results revealed that the C-terminal domain of CreAPo−1 plays an important role in alleviating CCR.Furthermore,we replaced the C-terminus of Cre1 with that of CreAPo−1 in T.reesei(Tr-0)and generated Tr-1.As a control,the C-terminus of Cre1 was truncated and Tr-2 was generated.The transcriptional profiles of these transformants revealed that the C-terminal chimera greatly improves cellulase transcription in the presence of glucose and thus upregulates cellulase in the presence of glucose and weakens CCR,consistent with truncating the C-terminus of Cre1 in Tr-0.Therefore,we propose constructing a C-terminal chimera as a new strategy to improve cellulase production and alleviate CCR in the presence of glucose.

Blastocyst complementation by pluripotent stem cell(PSC)injection is believed to be the most promising method to generate xenogeneic organs.However,ethical issues prevent the study of human chimeras in the late embryonic stage of development.Primate embryonic stem cells(ESCs),which have similar pluripotency to human ESCs,are a good model for studying interspecies chimerism and organ generation.However,whether primate ESCs can be used in xenogenous grafts remains unclear.In this study,we evaluated the chimeric ability of cynomolgus monkey(Macaca fascicularis)ESCs(cmESCs)in pigs,which are excellent hosts because of their many similarities to humans.We report an optimized culture medium that enhanced the anti-apoptotic ability of cmESCs and improved the development of chimeric embryos,in which domesticated cmESCs(D-ESCs)injected into pig blastocysts differentiated into cells of all three germ layers.In addition,we obtained two neonatal interspecies chimeras,in which we observed tissue-specific D-ESC differentiation.Taken together,the results demonstrate the capability of D-ESCs to integrate and differentiate into functional cells in a porcine model,with a chimeric ratio of 0.001-0.0001 in different neonate tissues.We believe this work will facilitate future developments in xenogeneic organogenesis,bringing us one step closer to producing tissue-specific functional cells and organs in a large animal model through interspecies blastocyst complementation.
