Neuroanatomical evidences have revealed direct histaminergic projections from the tuberomamillary nucleus of the hypothalamus to almost whole brain including the ***,it has been proposed that the central histaminergic nervous system holds a key position in the regulation of many basic body functions,our previous studies have further demonstrated a postsynaptic excitatory effect of histamine on the cerebellar Purkinje and nuclear neurons via histamine H2 receptors,suggesting a modulatory effect of histamine on motor *** the present study,using rat brain slice preparations and whole-cell patch recordings,the effects of histamine on two types of the cerebellar nuclear neurons were observed and the underlying ionic mechanisms were further *** slices with 30,100,300μM histamine produced an excitatory response in typeⅠneurons(projection neurons,n = 17) instead of typeⅡneurons (interneuron/local circuit neurons,n = 11) within all three cerebellar nuclei(i.e.,fastigial,interpositus and dentate nucleus). The histamine-induced excitation on typeⅠneurons were found to be evoked by the depolarization of membrane potential (n = 8) under superfusing the slices with *** addition,coupled the depolarization,the input resistance of these neurons excited by histamine slightly decreased,suggesting one or more types of ion channels in the membrane of type I neurons may be ***,2 mM Cs,which can prevent the hyperpolarization-activated cation current(I current), blocked the histamine-induced excitation(n = 6),suggesting the hyperpolarization-activated cation channels may be responsible for the depolarization of typeⅠneurons elicited by *** these results demonstrate that histamine excites cerebellar nuclear typeⅠneurons rather than typeⅡneurons through activation of the hyperpolarization-activated cation *** typeⅠneurons in the cerebellar nuclei are projection neurons whereas typeⅡones are intemeurons and

Signal intensity changes of dentate nucleus on plain MR T1WI innasopharyngeal carcinoma patients after radiotherapy andmultiple injections of gadolinium-base contrast agent

Objective To observe changes of plain MR T1WI signal intensity of dentate nucleus in nasopharyngeal carcinoma patients after radiotherapy and multiple times of intravenous injection of gadolinium-based contrast agent(GBCA).Methods Fifty patients with pathologically confirmed nasopharyngeal carcinoma and received intensity-modulated radiotherapy were retrospectively enrolled as the nasopharyngeal carcinoma group,and 50 patients with other malignant tumors and without history of brain radiotherapy were retrospectively enrolled as the control *** patients received yearly GBCA enhanced MR examinations for the nasopharynx or the head.T1WI signal intensities of the dentate nucleus and the pons on same plane were measured based on images in the year of confirmed diagnosis(recorded as the first year)and in the second to the fifth years.T1WI signal intensity ratio of year i(ranging from 1 to 5)was calculated with values of dentate nucleus divided by values of the pons(ΔSI i),while the percentage of relative changes of year j(ranging from 2 to 5)was calculated withΔSI j compared toΔSI 1(Rchange j).The values of these two parameters were compared,and the correlation ofΔSI and GBCA injection year-time was evaluated within each *** No significant difference of gender,age norΔSI 1 was found between groups(all P>0.05).The second to the fifth yearΔSI and Rchange in nasopharyngeal carcinoma group were all higher than those in control group(all P<0.05).Within both groups,ΔSI was positively correlated with GBCA injection year-time(both P<0.05).Conclusion Patients with nasopharyngeal carcinoma who underwent radiotherapy and multiple times of intravenous injection of GBCA tended to be found with gradually worsening GBCA deposition in dentate nucleus,for which radiotherapy might be a risk factor.

Effect of extracranial electric stimulation at cerebellar fastigial nucleus on serum C-reactive protein of patients with acute cerebral infarction

BACKGROUND： Some reports indicate that electric and/or chemical stimulation at various brain sites of experimental animals can raise regional cerebral blood flow and improve cerebral circulation; however, its mechanism is still unclear.OBJECTIVE： To observe the effects of electric stimulation at cerebellar fastigial nucleus on serum C-reactive protein of patients with acute cerebral infarction.DESIGN： Non-randomized synchronized contrast study.SETTING： The Second People＇s Hospital of Xinxiang City.PARTICIPANTS： A total of 54 patients with acute cerebral infarction were selected from the Department of Neurology, the Second People＇s Hospital of Xinxiang from December 2005 to December 2006. There were 31 males and 23 females, and their ages ranged from 56 to 80 years. All patients met the diagnostic criteria of the Fourth National Cerebrovascular Academic Meeting, were finally diagnosed by using CT examination,and provided the confirmed consent. Based on therapeutic demands, patients were divided into electric stimulation group and routine treatment group with 27 cases in each group. In addition, 21 healthy subjects,including 11 males and 10 females and aging 53 - 78 years, were selected as the control group. All the subjects in the control group did not have any histories of cerebrovascular diseases and severe body diseases.METHODS： Based on routine drug therapy, patients in the electric stimulation group were also treated by using CVFT-010M cerebral circulation function therapeutic device （made in Shanghai）. Electrode was fixed at bilateral mastoid in the first group and at extensible sides of upper limbs in the second group. Electric stimulation was given twice a day and lasted for 30 minutes each time. Ten days were regarded as a *** of device： mode Ⅲ, frequency 198%, and intensity 90% - 110% （bionic current）. Patients in the routine treatment group received the routine drug treatment. Role of orexin in cerebellar motor-emotional integration

Orexin deficiency results in cataplexy, which is characterized by a sudden loss of muscle tone often triggered by strong emotions. On the other hand, physical exercise helps for mood improvement. Yet little is known about the neural circuits and mechanisms underlying the motor-emotional integration. Here, we reveal a direct projection from hypothalamic orexinergic neurons to the cerebellum, a classical subcortical motor structure regulating not only motor coordination but also emotional and cognitive functions. However, the orexinergic neurons projecting to the cerebellum are distinct from the subpopulations projecting to the basolateral amygdala（BLA）, indicating a parallel innervation of the central orexinergic system on motor and emotional circuits. Furthermore, orexinergic afferent inputs in the cerebellum rather than those in the BLA exert an indispensable role in motor control when an animal is facing motor challenge. Interestingly, challenging movements induce a more significant anxiolytic effect than routine ones. Selective excitation of orexin on the glutamatergic projection neurons in the cerebellar nuclei, and the consequent activation of the direct glutamatergic projections from the cerebellar nuclei to the BLA may mediate the emotional improvement induced by challenging movements. These results demonstrate a novel mechanism underlying the motor-emotional integration via the cerebellar-amygdala circuit driven by the central orexinergic system. The findings may contribute to understanding the pathogenesis of cataplexy and account for why exercise improves mood and emotional feelings.
