Zbtb1 is a member of the zinc finger and BTB domain containing protein family of transcriptional factors,which is critical for T cell *** previous study,we found that Zbtb1 can regulate the expression of IL-7 Rα during T cell development of ***-7 signaling is transduced by the IL-7 receptor,which is essential for T cell maturation and *** developmental defects of T cells induced by Zbtb1 mutation,the present study aimed to investigate the regulation of IL-7 Rα gene expression by Zbtb1 ***’re going to overexpress Zbtb1 gene in vitro and in vivo,which can confirm the inhibitory effect of Zbtb1 on IL-7 Rα*** binding of Zbtb1 to cis-element related to IL-7 Raexpression will be identified by Biotin ChIP-seq *** key sites of cis-element for Zbtb1 binding will be mutated by CRISPR-Cas9 technique in a mouse DN thymocyte cell line(Dl).The differential regulation of IL-7 Rαwill be tested in wild type and mutant D1 cell line by overexpression of Zbtb1,which can illustrate the molecular mechanism for IL-7 Rαregulation by *** results for this study will shed light on the function and mechanism of Zbtb1 on T cell development.

Single-cell transcriptomic analysis reveals the developmental trajectory and transcriptional regulatory networks of pigment glands in Gossypium bicki

Comprehensive utilization of cottonseeds is limited by the presence of pigment glands and its inclusion *** ideal cotton has glandless seeds but a glanded plant,a trait found in only a few Australian wild cotton species,including Gossypium *** of this trait into cultivated species has proved to be *** the biological processes toward pigment gland morphogenesis and the associated underlying molecular mechanisms will facilitate breeding of cultivated cotton varieties with the trait of glandless seeds and glanded *** this study,single-cell RNA sequencing(scRNA-seg)was performed on 12222 protoplasts isolated from cotyledons of germinating *** seeds 48 h after *** into 14 distinct clusters unsupervisedly,these cells could be grouped into eight cell populations with the assistance of known cell marker *** pigment gland cells were well separated from others and could be separated into pigment gland parenchyma cells,secretory cells,and apoptotic *** integrating the pigment gland cell developmental trajectory,transcription factor regulatory networks,and core transcription factor functional validation,we established a model for pigment gland *** this model,light and gibberellin were verified to promote the formation of pigment *** addition,three novel genes,GbiERF114(ETHYLENE RESPONSE FACTOR 114),GbiZAT11(ZINC FINGER OF ARABIDOPSIS THALIANA 11),and GbiNTL9(NAC TRANSCRIPTION FACTOR-LIKE 9),Were found to affect pigment gland ***,these findings provide new insights into pigment gland morphogenesis and lay the cornerstone for future cotton scRNA-seq investigations.

Deletion of IFT20 in early stage T lymphocyte differentiation inhibits the development of col agen-induced arthritis

IFT20 is the smallest member of the intraflagellar transport protein（IFT） complex B. It is involved in cilia formation. Studies of IFT20 have been confined to ciliated cells. Recently, IFT20 was found to be also expressed in non-ciliated T cells and have functions in immune synapse formation and signaling in vitro. However, how IFT20 regulates T-cell development and activation in vivo is still unknown. We deleted the IFT20 gene in early and later stages of T-cell development by crossing IFT20flox/flox（IFT20f/f） mice with Lck-Cre and CD4-Cre transgenic mice, and investigated the role of IFT20 in T-cell maturation and in the development of T cell-mediated collagen-induced arthritis（CIA）. We found that both Lck-Cre/IFT20f/f and CD4-Cre/IFT20f/f mice were indistinguishable from their wild-type littermates in body size, as well as in the morphology and weight of the spleen and thymus. However, the number of CD4- and CD8-positive cells was significantly lower in thymus and spleen in Lck-Cre/IFT20f/f mice. Meanwhile, the incidence and severity of CIA symptoms were significantly decreased, and inflammation in the paw was significantly inhibited in Lck-Cre/IFT20f/f mice compared to Lck-Cre/IFT201/1littermates. Deletion IFT20 in more mature T cells of CD4-Cre/IFT20f/f mice had only mild effects on the development of T cells and CIA. The expression of IL-1b, IL-6 and TGF-b1 were significantly downregulated in the paw of Lck-Cre/IFT20f/f mice, but just slight decreased in CD4-Cre/IFT20f/f mice. These results demonstrate that deletion of IFT20 in the early stage of T-cell development inhibited CIA development through regulating T-cell development and the expression of critical cytokines.
