Expression of Syk in non-small cell lung cancer and its relationship with clinicopathological parameters

This study aims to research the expression of spleen tyrosine kinase(Syk)in non-small cell lung cancer(NSCLC)and the relationship between Syk and clinico-pathologic factors and *** was applied to detect the expression of Syk and p53 protein in 39 cases of NSCLC(23 cases of lung squamous cell can-cer,16 cases of lung adenocarcinoma)and tumor-sur-rounding normal lung *** positive rate of Syk was 46.15%(18/39)and 100%(39/39)in NSCLC and tumor-surrounding normal lung tissues,*** expres-sion level of Syk in NSCLC was signiﬁcantly lower than that in tumor-surrounding normal lung tissues(P=0.000).The Syk expression was positively correlated with the p53 expression in NSCLC specimens(P=0.025).There was no signiﬁcant association between Syk expression and lymph node metastasis,differentiation degree,tumor size and tumor node metastasis(TNM).The present study demonstrated that Syk was aberrantly expressed in the NSCLC and might have a signiﬁcant impact on tumor growth and progression.

OBJECTIVE:To observe the effects of the Yiqi Qingdu prescription(益气清毒方)on intermediate-stage and advanced non-small-cell lung cancer(NSCLC).METHODS:In total,300 patients with intermediate-stage or advanced NSCLC were randomly and equally divided into three groups using computer-generated random numbers as follows:Western medicine(WM),Chinese medicine(CM),and integrated Traditional Chinese and Western Medicine(IM).After 3 months of treatment,the overall response rate(ORR);disease control rate(DCR);symptom score(SS);Karnofsky performance status(KPS);adverse event score;counts of CD3^(+),CD4^(+),and CD8^(+)cells;CD4^(+)/CD8^(+)ratio;and carcinoembryonic antigen(CEA)level were compared among the ***:The ORRs were 30.36%,20.24%,and 7.87%in the IM,CM,and WM groups,respectively,whereas the DCRs were 85%,75%,and 73%,*** to the CM group,the ORR was significantly higher in the WM and IM groups,whereas the DCR was significantly higher in the IM group(all Pcell counts were significantly decreased in the WM group(P=0.031).In the IM group,CD8^(+)cell counts were increased after treatment,whereas the CD4^(+)/CD8^(+)ratio was decreased(both Pcell counts were significantly higher in the IM group,whereas the CD4^(+)/CD8^(+)ratio was significantly lower(P=0.011).Relative to the CM group,CD8^(+)cell counts were significantly higher(P=0.001)and the CD4^(+)/CD8^(+)ratio was significant ly lower in the IM group(P=0.001).CEA levels were significantly increased in the CM group(P=0.023).CONCLUSION:The Yiqi Qingdu prescription can improve the outcomes of WM in patients with NSCLC

Herbal formula Renshenwuweizi decoction induces p53-mediated cell cycle arrest and apoptosis in A549 cells

OBJECTIVE:To investigate the effect of Renshenwuweizi decoction(RSWWZ decoction)on the growth of non-small cell lung cancer cells in ***:A549 non-small cell lung cancer cells were divided into two groups:control and RSWWZ decoction treatment *** Counting Kit-8 was used to measure the inhibitory effect of RSWWZ decoction on the growth of A549 cells.4’,6-diamidino-2-phenylindole staining and Annexin V-fluorescein isothiocyanate/propidium iodide double staining were used to investigate apoptosis in A549 cells following RSWWZ decoction treatment,and the mitochondrial membrane potential of treated cells was detected with Rhodamine *** cycle progression was analyzed by flow *** m RNA levels of p53,Bax,B-cell lymphoma-2(Bcl-2)and p21 were measured by quantitative real-time reverse transcription polymerase chain *** protein expressions of p53,Bax,Bcl-2,p21,cyclin-dependent kinases 2(CDK2),and cyclin A were detected by Western ***:RSWWZ decoction reduced the viability of A549 cells in a dose-dependent manner by inducing apoptosis and decreased mitochondrial membrane *** decoction increased p53 and Bax expression and decreased Bcl-2 expression in a dose-dependent *** decoction also induced an S-phase cell cycle arrest by increasing p21 and decreasing cyclin A and CDK2 ***:In vitro experiments revealed that the Renshenwuweizi decoction-induced decrease in A549 cell proliferation was achieved by inducing apoptosis and S-phase cell cycle arrest via the p53 *** findings provide the experimental basis for Renshenwuweizi decoction treatment of lung cancer.

Improved local control without elective nodal radiotherapy in patients with unresectable NSCLC treated by 3D-CRT

To investigate the influence of prophylactic elec-tive nodal irradiation on the therapeutic results of definitive radiotherapy for patients with stage IIIA or stage IIIB unre-sectable non-small-cell lung cancer,55 patients with clini-cally inoperable advanced non-small-cell lung cancer were *** four cycles of induction chemotherapy,the patients were divided into two groups at *** one group,the elective nodal irradiation was included in clinical tumor volume(CTV)of definitive radiotherapy(ENI group);and in the other group,elective nodal irradiation was not included in CTV(non-ENI group).For the patients in the ENI group,the mean prescription dose for gross tumor volumes was 58.4 Gy,while for the patients in the non-ENI group,it was 65.8 Gy(P<0.05).The responsive rates were 45.8%and 74.0%(P<0.05),and the rate of the elective nodal failure(ENF)was 4.2%and 11.1%,***-Meier analysis showed that the mean local-progression-free survival time was 11.0 and 15.0 months,and one-year local-failure rates were 51.9%and 24.5%(P<0.05).The median overall survival time was 13.0 and 15.0 months,respectively(P=0.084).The one-year survival rates were 55.7%and 72.5%,and two-year survival rates were 0%and 19.9%.There was no significant difference in the occurrences of radiation-associated complications between the two *** results showed that omitting elective nodal irradiation did not result in a high incidence of elective nodal *** the contrary,it decreased local failure by increasing prescrip-tion doses to the primary diseases and lymphadenopaphy,and thereby it may further prolong the patients’survival.
