Evoking immune system to potentiate nanocatalytic therapy through activation of cgas-sting pathway

Nanocatalytic therapy shows great potential for therapeutic interventions. However, therapeutic efficiency is often limited by unsatisfactory enzyme activity and lack of the coordination of immune system. Therefore, engineering nanozymes activity enhancement while activating immune system will be an effective strategy to achieve efficient tumor therapy. Herein, we synthesize a DSPE-PEG-FA modified manganese dioxide-based dual-atom nanozyme（MDF）, on which iridium and platinum atoms are anchored. The obtained MDF can simultaneously mimic four enzyme activities of catalase, oxidase, peroxidase, and glutathione oxidase, set off a reactive oxygen species（ROS） storm, cause tumor cell death. The enzyme activity of MDF can be enhanced by its own photothermal effect. Meanwhile, MDF can consume intracellular glutathione and release Mn2+, which can prevent generated ROS from consumption and further activate cyclic guanosine monophosphate-adenosine monophosphate synthase-stimulator of interferon genes（cgas-sting） pathway and promote the secretion of type I interferon, which will help promote dendritic cells maturation, present antigens to T lymphocytes to help kill tumor cells. Ultimately, MDF shows excellent tumor suppressive effects. This work provides a new paradigm for the field of nanozymes and offers a new reference for involvement of cgas-sting pathway activation in tumor catalytic therapy.

In the realm of autoimmune and inflammatory diseases,the cyclic GMP-AMP synthase(cgas)stimulator of interferon genes(sting)signaling pathway has been thoroughly investigated and *** this,the clinical approval of drugs targeting the cgas-sting pathway has been *** Total glucosides of paeony(TGP)is highly anti-inflammatory and is commonly used in the treatment of rheumatoid arthritis(RA),emerged as a subject of our *** found that the TGP markedly reduced the activation of the cgas-sting signaling pathway,triggered by various cgas-sting agonists,in mouse bone marrow-derived macrophages(BMDMs)and Tohoku Hospital Pediatrics-1(THP-1)*** inhibition was noted alongside the suppression of interferon regulatory factor 3(IRF3)phosphorylation and the expression of interferon-beta(IFN-β),C-X-C motif chemokine ligand 10(CXCL10),and inflammatory mediators such as tumor necrosis factor-alpha(TNF-α)and interleukin-6(IL-6).The mechanism of action appeared to involve the TGP’s attenuation of the sting-IRF3 interaction,without affecting sting oligomerization,thereby inhibiting the activation of downstream signaling *** vivo,the TGP hindered the initiation of the cgas-sting pathway by the sting agonist dimethylxanthenone-4-acetic acid(DMXAA)and exhibited promising therapeutic effects in a model of acute liver injury induced by lipopolysaccharide(LPS)and D-galactosamine(D-GalN).Our findings underscore the potential of the TGP as an effective inhibitor of the cgas-sting pathway,offering a new treatment avenue for inflammatory and autoimmune diseases mediated by this pathway.

Biomolecular condensates formed by phase separation are widespread and play critical roles in many physiological and pathological ***-sting signaling functions to detect aberrant DNA signals to initiate anti-infection defense and antitumor *** the same time,cgas-sting signaling must be carefully regulated to maintain immune ***,exciting recent studies have reported that biomolecular phase separation exists and plays important roles in different steps of cgas-sting signaling,including cgas condensates,sting condensates,and IRF3 *** addition,several intracellular and extracellular factors have been proposed to modulate the condensates in cgas-sting *** studies reveal novel activation and regulation mechanisms of cgas-sting signaling and provide new opportunities for drug ***,we summarize recent advances in the phase separation of cgas-sting signaling and the development of potential drugs targeting these innate immune condensates.

DNA-induced liquid-liquid phase separation of cyclic GMP-AMP synthase（cgas）triggers a switchlike response to detect pathogen infection and promote innate immune *** and how pathogens manipulate cgas-DNA condensation to mediate immune evasion is *** report the identification of a structurally related viral tegument protein family,represented by ORF52 and VP22 from gammaand alpha-herpesvirinae respectively,that employs a conserved mechanism to restrict cgas-DNA phase ***52/VP22 proteins accumulate into,and effectively disrupt,the preformed cgas-DNA condensation both in vitro and in *** inhibition process is dependent on DNA-induced liquid-liquid phase separation of the viral protein,rather than a direct interaction with ***,highly abundant ORF52 proteins carried within viral particles are able to target cgas-DNA phase separation in early infection *** results define ORF52/VP22-type tegument proteins as a family of inhibitors targeting cgas-DNA phase separation and demonstrate a mechanism for how viruses overcome innate immunity.
