Crosstalk between PC 12 cells and endothelial cells in an artificial neurovascular niche constructed by a dual-functionalized selfassembling peptide nanofiber hydrogel

The coordination between neurogenesis and angiogenesis plays an important role in nerve tissue development and regeneration.Recently,using bioactive materials to drive neurogenic and angiogenic responses has gained increasing attention.Understanding the neurovascular link between regulatory cues offers valuable insight into the mechanisms underlying nerve regeneration and the design of new bioactive materials.In this study,we utilized a dual-functionalized peptide nanofiber hydrogel presenting the brain-derived neurotrophic factor and vascular endothelial growth factor mimetic peptides RGIDKRHWNSQ(RGI)and KLTWQELYQLKYKGI(KLT)to construct an artificial neurovascular microenvironment.The dual-functionalized peptide nanofiber hydrogel enhanced the neurite outgrowth of pheochromocytoma(PC12)cells and tube-like structures formation of human umbilical vein endothelial cells(HUVECs)in vitro,and promoted rapid lesion infiltration of neural and vascular cells in a rat brain injury model.Using indirect co-culture models,we found that the dual-functionalized peptide hydrogel effectively mediated neurovascular crosstalk by regulating secretion of paracrine factors from PC12 cells and HUVECs.When the two cells types were directly co-cultured on the dua卜functionalized peptide hydrogel,the efficiency of cell-cell communication was enhanced,which further accelerated the differentiation and maturation of PC12 cells with an increased number of pseudopodia and spread morphology,and HUVECs tube-like structure formation.In summary,the dual-functionalized peptide nanofiber hydrogel successfully formed an artificial neurovascular niche to directly regulate the behaviors of neural and vascular cells and promote their neurovascular crosstalk through paracrine signaling and direct cell-cell contact.

Eletromagnetic Detection of Mild brain injury: A Novel Imaging Approach to Post Concussive Syndrome

Introduction: Mild traumatic brain injury (mTBI) is a common injury, with nearly 3 - 4 million cases annually in the United States alone. Neuroimaging in patients with mTBI provides little benefit, and is usually not indicated as the diagnosis is primarily clinical. It is theorized that microvascular trauma to the brain may be present in mTBI, that may not be captured by routine MRI and CT scans. Electromagnetic (EM) waves may provide a more sensitive medical imaging modality to provide objective data in the diagnosis of mTBI. Methods: COMSOL simulation software was utilized to mimic the anatomy of the human skull including skin, cranium, cerebrospinal fluid (CSF), gray-matter tissue of the brain, and microvasculature within the neural tissue. The effects of penetrating EM waves were simulated using the finite element analysis software and results were generated to identify feasibility and efficacy. Frequency ranges from 7 GHz to 15 GHz were considered, with 0.6 and 1 W power applied. Results: Variations between the differing frequency levels generated different energy levels within the neural tissue—particularly when comparing normal microvasculature versus hemorrhage from microvasculature. This difference within the neural tissue was subsequently identified, via simulation, serving as a potential imaging modality for future work. Conclusion: The use of electromagnetic imaging of the brain after concussive events may play a role in future mTBI diagnosis. Utilizing the proper depth frequency and wavelength, neural tissue and microvascular trauma may be identified utilizing finite element analysis.

Sport level and sex differences in sport-related concussion among Japanese collegiate athletes:Epidemiology,knowledge,reporting behaviors,and reported symptoms

This study investigated the incidence of sport-related concussion(SRC)in sports,effect of athlete knowledge on reporting behavior differences between collegiate and non-collegiate athletes,and differences in SRC symptoms between sexes and level of participation.In this cross-sectional survey,1344 Japanese collegiate and non-collegiate athletes from a single institute were analyzed.Using a web-based survey,demographics,general SRC,knowledge of SRC,the most recent SRC reporting behaviors,and symptom presentation were examined.The prevalence of SRC during the academic year 2016-2017 was 2.68(95%confidence interval[CI]:1.88-3.69)across all sports.The prevalence of SRC was 33.3(95%CI:17.96-51.83)in rugby union and 8.33(95%CI:1.03-27.00)in women's soccer.The prevalence of SRC in males(3.47[95%CI:2.38-4.86]was 3.65 times higher than that in females(0.95[95%CI:0.26-2.41]).In total,the mean total score of knowledge was 5.30(4.2)across 25 questions;dizziness was the most well-known symptom(867/1344,64.5%),followed by headache(59.3%).Being more emotional(44/1345,3.3%)was the least frequently known symptom.Level of participation did not affect scores(5.16[3.96]vs.5.52[4.54];p=0.131).All 87 disclosing participants experienced drowsiness and irritability and felt more emotional.In terms of sex and participant level,no significant differences were found in any symptoms.This study found very low rates of concussion education in Japan.Dissemination of concussion education is essential in the future to recognize concussion earlier and prevent severe concussive injury.

Cerebral autoregulation(CA)dysfunction is a strong predictor of clinical outcome in patients with acute brain injury(ABI).CA dysfunction is a potential pathologic defect that may lead to secondary injury and worse functional outcomes.Early therapeutic hypothermia(TH)in patients with ABI is controversial.Many factors,including patient selection,timing,treatment depth,duration,and rewarming strategy,impact its clinical efficacy.Therefore,optimizing the benefit of TH is an important issue.This paper reviews the state of current research on the impact of TH on CA function,which may provide the basis and direction for CA-oriented target temperature management.

Computerized Training of Working Memory for Patients with Acquired brain injury

Background: Patients with acquired brain injury often experience impaired working memory (WM), a condition that can make everyday life activities and work difficult. Objectives: This study investigates the effects of computerized WM training on WM skills, cognitive tests, activity performance and estimated health and whether the effects of computerized WM training can be attributed to sex or time since injury. Methods: Forty-eight patients with acquired brain injury underwent computerized WM training. Patients were tested by a neuropsychologist and interviewed by an occupational therapist just prior and 20 weeks after completion of training. Results: Patients who participated in computerized WM training significantly improved their WM skills shown in WM index, their neuropsychological test scores, and their self-estimated health scores. They also significantly improved their performance of individually defined WM-related everyday activities and their satisfaction with the performance of these activities. There was a significant difference in terms of WM index, WM-related daily activity performance, and satisfaction with respect to time since injury. Conclusion: Computerized WM training can improve cognitive and everyday performance for patients with acquired brain injury. Patients can improve their cognitive functions a long time after suffering a brain injury or disease. This effect is greater if WM training is used early in the rehabilitation.

Biomarkers are biological molecules that provide information about the physiological condition of an organism such as early signals of health deterioration.The aim of this study was to compare the relative gene expression of four molecular biomarkers related to oxidative stress,and neuronal damage in Piaractus brachypomus under three different conditions(exposure to chlorpyrifos(CPF),menthol and eugenol anesthesia,and brain injury).Bioinformatic analyses were conducted to design primers for the molecular biomarkers,then,the relative gene expression of COX1,GPX1,BDNF and CALB1 genes was measured by qPCR using brain tissue cDNA from the treated animals as template and 2-ΔΔCT method was performed.EF1a gene was used as reference gene.The results showed a response to chemical stress in the CPF treatment samples where GPX1,BDNF and CALB1 genes were up-regulated.Finally,in menthol/eugenol and brain injury treatments,no alterations in the relative gene expression were observed.

Standardization of apple cancellation test for neglect patients in Korea:An observational study

BACKGROUND Neglect can be divided into two types using apple cancellation test(apple test):Egocentric neglect(EN)and allocentric neglect(AN).However,in South Korea,apple test results and decision criteria are still largely dependent on tests by foreign countries.AIM To establish a new South Korea standard and improve the accuracy of neglect assessment,the apple experiment was standardized in this study.METHODS This study was conducted on 223 healthy subjects for a total of 7 mo from August 2021 to February 2022.Standardization was carried out using the original apple test developed by Bickerton in 2011.In scoring for the apple test,total omission error refers to the number of missed targets(full apple)in the entire test sheet(left,middle,and right area).The score for EN is the difference between the correct number of right area and the correct number of left area(excluding the middle area).For AN,the score is difference between the number of left opening apples and number of right opening apples(including the middle area).Linear regression analysis was used for standardization using the general characteristics of subjects and the results of the apple test.RESULTS The cut-off score,which is the standard value indicating the pathological condition by combining the results of all subjects,is as follows:Total omission error(5),error for EN(2),and error for AN(2).Also,differences in cut-off score according to age were found.CONCLUSION This study will be helpful in facilitating a more accurate differential diagnosis of neglect.

Differences in brain structure in patients with distinct sites of chronic pain:A voxel-based morphometric analysis

A reduction in gray matter volume is common in patients with chronic back pain, and different types of pain are associated with gray matter abnormalities in distinct brain regions. To examine differ- ences in brain morphology in patients with low back pain or neck and upper back pain, we investi- gated changes in gray matter volume in chronic back pain patients having different sites of pain using voxel-based morphometry. A reduction in cortical gray matter volume was found primarily in the left postcentral gyrus and in the left precuneus and bilateral cuneal cortex of patients with low back pain. In these patients, there was an increase in subcortical gray matter volume in the bilateral putamen and accumbens, right pallidum, right caudate nucleus, and left amygdala. In upper back pain patients, reduced cortical gray matter volume was found in the left precentral and left postcen- tral cortices. Our findings suggest that regional gray matter volume abnormalities in low back pain patients are more extensive than in upper back pain patients. Subcortical gray matter volume in- creases are found only in patients with low back pain.

Luteolin attenuates neuronal apoptosis in the hippocampi of diabetic encephalopathy rats

Luteolin （3＇,4＇,5,7-tetrahydroxyflavone） has powerful anti-apoptotic and antioxidant properties. This study aimed to investigate the effects of luteolin on hyperglycemia-mediated apoptosis in the hippocampi of rats with streptozotocin-induced diabetic encephalopathy after injection into the tail veins, and the molecular mechanisms involved. Biochemistry and terminal deoxynucleotidyl transferase mediated dUTP nick end labelling detection results showed that luteolin treatment （given twice daily for 15 days） significantly inhibited hyperglycemia-mediated apoptosis, decreased malondialdehyde levels and increased glutathione levels in the hippocampi of streptozotocin- induced diabetic rats. Western blot analysis revealed that luteolin also inhibited the expression of apoptosis-related factors and cytochrome c release from mitochondria. Luteolin also improved the learning and memory abiJities of rats with diabetic encephalopathy in a water maze test. Further western blot analysis revealed that luteolin treatment facilitated neuronal cell survival through activation of the phosphatidylinositol 3-kinase/Akt signaling pathway, an extracellular signal pathway involved in the suppression of cell apoptosis and promotion of cell survival. These experimental findings indicate that luteolin can inhibit apoptosis of hippocampal nerve cells in rats with diabetic encephalopathy, and that this effect is mediated by an indirect antioxidative effect, the inhibition of activation of apoptosis-related factors and the activation of phosphatidylinositol 3-kinase/Akt signal pathway.

Acupuncture at Waiguan(SJ5) and sham points influences activation of functional brain areas of ischemic stroke patients: a functional magnetic resonance imaging study

Most studies addressing the specificity of meridians and acupuncture points have focused mainly on the different neural effects of acupuncture at different points in healthy individuals. This study examined the effects of acupuncture on brain function in a pathological context. Sixteen patients with ischemic stroke were randomly assigned to true point group （true acupuncture at right Waiguan （SJ5）） and sham point group （sham acupuncture）. Results of functional magnetic resonance imaging revealed activation in right parietal lobe （Brodmann areas 7 and 19）, the right temporal lobe （Brodmann area 39）, the right limbic lobe （Brodmann area 23） and bilateral oc-cipital lobes （Brodmann area 18）. Furthermore, inhibition of bilateral frontal lobes （Brodmann area 4, 6, and 45）, right parietal lobe （Brodmann areas 1 and 5） and left temporal lobe （Brodmann area 21 ） were observed in the true point group. Activation in the precuneus of right parietal lobe （Brodmann area 7） and inhibition of the left superior frontal gyrus （Brodmann area 10） was observed in the sham group. Compared with sham acupuncture, acupuncture at Waiguan in stroke patients inhibited Brodmann area 5 on the healthy side. Results indicated that the altered specificity of sensation-associated cortex （Brodmann area 5） is possibly associated with a central mechanism of acupuncture at Waiguan for stroke patients.
