Objective To better understand the pathological causes of bone loss in a space environment, including microgravity, ionizing radiation, and ultradian *** Sprague Dawley （SD） rats were randomly divided into a baseline group, a control group, a hindlimb suspension group, a radiation group, a ultradian rhythms group and a combined-three-factor group. After four weeks of hindlimb suspension followed by X-ray exposure and/or ultradian rhythms, biomechanical properties, bone mineral density, histological analysis, microstructure parameters, and bone turnover markers were detected to evaluate bone loss in hindlimbs of *** Simulated microgravity or combined-three factors treatment led to a significant decrease in the biomechanical properties of bones, reduction in bone mineral density, and deterioration of trabecular parameters. Ionizing radiation exposure also showed adverse impact while ultradian rhythms had no significant effect on these outcomes. Decrease in the concentration of the turnover markers bone alkaline phosphatase （bALP）, osteocalcin （OCN）, and tartrate-resistant acid phosphatase-5b （TRAP-Sb） in serum was in line with the changes in trabecular *** Simulated microgravity is the main contributor of bone loss. Radiation also results in deleterious effects but ultradian rhythms has no significant effect. Combined-three factors treatment do not exacerbate bone loss when compared to simulated microgravity treatment alone.

Type 2 diabetes mellitus(T2DM)exacerbates irreversible bone loss in periodontitis,but the mechanism of impaired bone regeneration caused by the abnormal metabolic process of T2DM remains *** are regarded as the critical mediator in diabetic impairment of regeneration via organ or tissue ***,we find that abnormally elevated exosomes derived from metabolically impaired liver in T2DM are significantly enriched in the periodontal region and induced pyroptosis of periodontal ligament cells(PDLCs).Mechanistically,fatty acid synthase(Fasn),the main differentially expressed molecule in diabetic exosomes results in ectopic fatty acid synthesis in PDLCs and activates the cleavage of gasdermin *** of liver Fasn effectively mitigates pyroptosis of PDLCs and alleviates bone *** findings elucidate the mechanism of exacerbated bone loss in diabetic periodontitis and reveal the exosome-mediated organ communication in the“liver-bone”axis,which shed light on the prevention and treatment of diabetic bone disorders in the future.

Background:Escherichia coli(***)infection in humans and animals usually comes with gut dysbiosis,which is potential culprit to skeletal health,it is still unclear to whether diet interfered gut microbiome changes can be a protective strategy to bone loss ***,the effects of resistant starch from raw potato starch(RPS),a type of prebiotic,on ***-induced bone loss and gut microbial composition in meat ducks were ***:The results showed that dietary 12%RPS treatment improved bone quality,depressed bone resorption,and attenuated the pro-inflammatory reaction in both ileum and bone ***,the 12%RPS diet also increased the abundance of Firmicutes in ***-treated birds,along with higher production of short-chain fatty acids(SCFAs)especially propionate and *** addition ofβ-acid,an inhibitor of bacterial SCFAs production,to the drinking water of ducks fed 12%RPS diet significantly decreased SCFAs level in cecum content and eliminated RPS-induced tibial mass ***,treatment with MI-2 to abrogate mucosa-associated lymphoid tissue lymphoma translocation protein 1(Malt1)activity replicated the protective role of dietary 12%RPS in ***-induced bone loss including reduced the inhibition on nuclear factorκB(NF-κB)inflammasome activation,decreased bone resorption,and improved bone quality,which were correlated with comparable and higher regulatory T cells(Treg)frequency in MI-2 and 12%RPS group,***:These findings suggested that the diet with 12%RPS could alleviate ***-induced bone loss in meat ducks by changing the gut microbial composition and promoting concomitant SCFAs production,and consequently inhibiting Malt1/NF-κB inflammasome activation and Treg cells expansion.

Computational Model for the Underlying Mechanisms Regulating bone loss by Mechanical Unloading and Estrogen Deficiency

The objective of this paper is to investigate the different effects of disuse and estrogen deficiency on bone loss and the underlying mechanisms.A mechanical-biological factors coupled computational model was built to simulate different patterns of bone loss induced in female rats by hind limb unloading,ovariectomy,or both in an animal study.A remodeling analysis was performed on a representative cross section of 6 mm2 of cancellous bone in the distal femoral metaphysis of the *** BMU activation frequency,the refilling rate,and the principal compressive strain in the state of mechanical unloading and estrogen deficiency were simulated to interpret the underlying *** bone loss patterns due to mechanical unloading,estrogen deficiency,or both all corresponded with the experimental *** results show that mechanical unloading and estrogen deficiency cause different bone loss patterns;moreover,mechanical unloading induces a greater degree of bone loss than estrogen deficiency,which can lead to improved treatment and prevention strategies for osteoporosis.

Low bone mineral density(BMD)or low bone mass is common in patients with inflammatory bowel disease(IBD).Studies have shown that low BMD is also common in patients with ulcerative colitis(UC)even after colectomy and ileal pouch-anal anastomosis(IPAA).The reported frequency of osteopenia ranged from 26-55%and that of osteoporosis ranged from 13-32%in patients with *** age,low body mass index,lack of calcium supplementation and high inflammatory activity with villous atrophy in the ileo-anal pouch are risk factors for low bone mass in pouch *** loss is also common in patients with IBD and *** professional society guidelines do not specifically address the need for surveillance in patients with ileal pouches or ostomy.A growing body of evidence suggests that patients with ileal pouch or ostomy are at an increased risk of bone *** prospective studies,screening and surveillance using dual energy X-ray absorptiometry(DEXA)along with calcium/vitamin D supplementation may be beneficial in those patients.
