baculovirus Surface Display of Zika Virus Envelope Protein Protects against Virus Challenge in Mouse Model

Zika virus(ZIKV)is emerging as a significant pathogen worldwide and may cause severe neurological disorders such as fetal microcephaly and Guillain-Barre syndrome.No drug or listed vaccines are currently available for preventing ZIKV infection.As a major target of neutralizing,ZIKV envelop(E)protein usually used for vaccine development.Nevertheless,the immunogenicity of ZIKV envelop(E)protein expressed by baculovirus display system has never been assessed.In this study,we reported a new strategy for surface display of ZIKV E protein by a recombinant baculovirus vector derived from Autographa californica multiple nuclear polyhedrosis virus(AcMNPV)and assessed its immunogenicity in mice.We produced recombinant fusion ZIKV E protein linked with signal peptide(SP)and transmembrane domain(TM)of AcMNPV GP64.The results showed that the recombinant protein was easy to produce by baculovirus display system.BALB/c mice immunized with this recombinant E protein developed ZIKV specific serum antibodies.The anti-E protein sera from the mice were able to effectively neutralize ZIKV in vitro.More importantly,AG6(IFN-a/b and IFN-c receptor deficient)mice immunized with recombinant E protein were protected against lethal ZIKV challenge.Together,thesefindings demonstrated that the recombinant E protein displayed by baculovirus can be conveniently prepared and displayed good immunogenicity in immunized mice.It is a promising practical approach for prompting the development of vaccine and related immunology research.

baculovirus vector-mediated transfer of NIS gene into colon tumor cells for radionuclide therapy

AIM:To investigate the feasibility of radionuclide therapy of colon tumor cells by baculovirus vector-mediated transfer of the sodium/iodide symporter(NIS) gene.METHODS:A recombinant baculovirus plasmid carrying the NIS gene was constructed,and the viruses(BacNIS) were prepared using the Bac-to-Bac system.The infection efficiency in the colon cancer cell line SW1116 of a green fluorescent protein(GFP) expressing baculovirus(Bac-GFP) at different multiplicities of infection(MOI) with various concentrations of sodium butyrate was determined by flow cytometry.An in vitro cytotoxicity assay was also conducted after infection of SW1116 cells with Bac-NIS.Iodine uptake of Bac-NIS infected SW1116 cells and inhibition of this uptake by sodium perchlorate was examined,and the effect of Bac-NISmediated 131 I in killing tumor cells was evaluated by cell colony formation tests.RESULTS:Infection and transgene expression in SW1116with Bac-GFP were significantly enhanced by sodium butyrate,as up to 72% of SW1116 cells were infected with the virus at MOI of 400 and sodium butyrate at 0.5 mmol/L.No obvious cytotoxicity was observed under these conditions.Infection of SW1116 with Bac-NIS allowed uptake of 131 I in these tumor cells,which could be inhibited by sodium perchlorate.The viability of SW1116 cells infected with Bac-NIS was significantly lower than with Bac-GFP,suggesting that NIS gene-mediated 131 I uptake could specifically kill tumor cells.CONCLUSION:baculovirus vector-mediated NIS gene therapy is a potential approach for treatment of colon cancer.

baculovirus has been widely used for the production of recombinant proteins ininsect cells.Since the finding that baculovirus can efficiently transduce mamma-lian cells,the applications of baculovirus have been greatly expanded.The pros-pects and drawbacks of baculovirus-mediated gene expression,either in insect orin mammalian cells,are reviewed.Recent progresses in expanding the applicationsto studies of gene regulation,viral vector preparation,in vivo and ex vivo genetherapy studies,generation of vaccine vectors,etc are discussed and the effortsdirected towards overcoming the existing bottlenecks are particularly emphasized.

baculovirus ETL promoter acts as a shuttle promoter between insect cells and mammalian cells

Aim:To identify a shuttle promoter that can mediate gene expression in bothinsect cells and mammalian cells to facilitate the development of a baculovirusvector-based mammalian cell gene delivery vehicle.Methods:Recombinantbaculoviruses carrying the β-galactosidase reporter gene under the control of anearly to late(ETL)promoter of the Autographa californica multiple nuclear poly-hedrosis virus(AcMNPV)or a cytomegalovirus immediate early promoter(CMVpromoter)were constructed.COS1,HeLa,CHO-K1,hFobl.19,and MCF-7 mam-malian cells were tested for the expression of β-galactosidase.Results:ETLpromoter activity was higher in bone-derived hFob 1.19 than in COS1,HeLa,CHO-K1,or MCF-7 mammalian cells.The transient plasmid transfection assay indi-cated that ETL promoter activity in mammalian cells was dependent on baculovirusgene expression.Conclusion:ETL promoter activity in mammalian cells isbaculovirus gene expression-dependent,and the shuttle promoter will facilitatethe application of baculovirus expression vectors in mammalian cell expressionsystems and for gene therapy.

Human lactoferrin (hLf) is a multifunctional iron-binding glycoprotein.In this study,weamplified hLfcDNA by reverse transcription-polymerase chain reaction from normal human mammary gland.The nucleotide sequence of the hLf was identical to the known hLf.We constructed a recombinant virus,vBm-hLf,harboring the hLf gene and exploited the BmN cells as host to produce recombinant human lactoferrin(rhLf).It was found that a recombinant protein with a molecular mass of approximately 78 kDa was expressed.Approximately 13.5μg rhLf was purified from 1-2×10~5 BmN cells infected by vBm-hLf and the rhLfproved to be biologically active.This method established in our study will pave the way for efficient productionof rhLf for further application of this protein in the future.

baculovirus-expressed FAdV-4 penton base protein protects chicken against hepatitis-hydropericardium syndrome

Hepatitis-hydropericardium syndrome(HHS)is an infectious disease caused by fowl adenovirus serotype 4(FAdV-4).Several structural and non-structural proteins of FAdV-4 have been expressed in Escherichia coli and baculovirus expression system to develop candidate subunit vaccines.However,the protective efficiency of baculovirus-expressed penton base protein has not been assessed.In this study,two recombinant capsid proteins,penton base and fiber-2,were constructed.And then,penton base and fiber-2 were administrated alone or together to specific pathogen-free(SPF)chickens at 14 days of life and boosted at 28 days of life.At 42 days of life,the immunized groups and the control group were challenged with FAdV-4 virulent strain.Results show that inoculating penton base or penton base+fiber-2 provided 100%protection to the chickens.All groups vaccinated with the recombinant protein produced detectable antibodies and showed no apparent lesions.Thus,baculovirus-expressed penton base protein is a promising candidate subunit vaccine.

Microbiota modulates gut immunity and promotes baculovirus infection in Helicoverpa armigera

baculoviruses are natural enemies of agricultural and forest insect pests and play an important role in biological pest control.Oral infection by baculovirus in the insect midgut is necessary for establishing systemic infection and eventually killing the insect.Since the insect midgut continuously encounters microbiota,the gut microbiota could affect baculovirus infection.Here,we demonstrated that gut microbiota modulates immune responses and promotes baculovirus infection in the cotton bollworm,Helicoverpa armigera.After oral infection,numerous host immunity-related genes including genes encoding Toll and immune deficiency(IMD)pathway components were upregulated in the midgut.Elimination of the gut microbiota significantly increased the resistance to viral infection in H.armigera.Quantitative real-time reverse transcription polymerase chain reaction and proteomic analysis showed that downregulation of the antiviral factor prophenoloxidase(PPO)could be mediated by microbiota during infection.It implied that midgut microbiota diminishes the expression of PPO to facilitate viral infection in H.armigera.Our findings revealed that the microbiota plays an important role in modulating the resistance of H.armigera to baculovirus infection,providing new insights in applying biopesticide.

Introduction of temperature-sensitive helper and donor plasmids into Bac-to-Bac baculovirus expression systems

In the baculovirus shuttle vector(bacmid) system, a helper plasmid and a donor plasmid are employed to insert heterologous genes into a cloned baculovirus genome via Tn7 transposition in Escherichia coli. The helper and donor plasmids are usually cotransfected with constructed bacmids into insect cells, which will lead to integration of these plasmids into the viral genome,and hence to the production of defective virions. In this study, to facilitate the preparation of plasmid-free recombinant bacmids, we modified a set of helper and donor plasmids by replacing their replication origins with that of a temperature-sensitive(ts) plasmid, p SIM6. Using the resulting ts helper plasmid p MON7124 ts and the ts donor plasmid p FB1ts-PH-GFP, a recombinant bacmid,b Ac WT-PG(-), was constructed, and the transposition efficiency was found to be 33.1%. The plasmids were then removed by culturing at 37 °C. For b Ac WT-PG(-), the infectious progeny virus titer and the protein expression level under the control of the polyhedrin promoter were similar to those of a bacmid constructed with unmodified helper and donor plasmids. These ts plasmids will be useful for obtaining plasmid-free bacmids for both heterologous protein production and fundamental studies of baculovirus biology.

Construction and Characterization of a Novel Bacmid AcBac-Syn Based on a Synthesized baculovirus Genome

baculoviruses are large DNA viruses which have been widely used as expression vectors and biological insecticides.Homologous recombination and Bac-to-Bac system have been the main methods for manipulating the baculovirus genome.Recently, we generated a synthetic baculovirus Ac MNPV-WIV-Syn1 which fully resembled its parental virus Autographa californica multiple nucleopolyhedrovirus(Ac MNPV). Here, we report the modification of Ac MNPV-WIV-Syn1 into a novel bacmid, Ac Bac-Syn, which can be used as a backbone for Bac-to-Bac system. To achieve this, a vector contained a Lac Z:att Tn7 and egfp cassette was constructed, and recombined with a linearized Ac MNPV-WIV-Syn1 genome by transformation-associated recombination in yeast to generate bacmid Ac Bac-Syn. The bacmid was then transfected to insect cells and the rescued virus showed similar biological characteristics to the wild-type virus in terms of the kinetics of budded virus production, the morphology of occlusion bodies, and the oral infectivity in insect larvae. For demonstration, a red fluorescent protein gene Dsred was transposed into the att Tn7 site by conventional Bac-to-Bac method, and the transfection and infection assays showed that Ac Bac-Syn can be readily used for foreign gene insertion and expression.Ac Bac-Syn has several advantages over the conventional Ac MNPV bacmids, such as it contains an egfp reporter gene which facilitates visualization of virus propagation and titration;its DNA copy numbers could be induced to a higher level in E. coli;and the retaining of the native polyhedrin gene in the genome making it an attractive system for studying the functions of gene related to occlusion body assembly and oral infection.
