Itch（pruritus） is one of the most disabling syndromes in patients suffering from skin, liver, or kidney diseases. Our previous study highlighted a key role of oxidative stress in acute itch. Here, we evaluated the effects of antioxidants in mouse models of acute and chronic itch and explored the potential mechanisms. The effects of systemic administration of the antioxidants N-acetyl-Lcysteine（NAC） and N-tert-butyl-a-phenylnitrone（PBN）were determined by behavioral tests in mouse models of acute itch induced by compound 48/80 or chloroquine, and chronic itch by treatment with a mixture of acetonediethyl-ether-water. We found that systemic administration of NAC or PBN significantly alleviated compound 48/80-and chloroquine-induced acute itch in a dose-dependent manner, attenuated dry skin-induced chronic itch, and suppressed oxidative stress in the affected skin.antioxidants significantly decreased the accumulation of intracellular reactive oxygen species directly induced by compound 48/80 and chloroquine in the cultured dorsal root ganglia-derived cell line ND7-23. Finally, the antioxidants remarkably inhibited the compound 48/80-induced phosphorylation of extracellular signal-regulated kinase in the spinal cord. These results indicated that oxidative stress plays a critical role in acute and chronic itch in the periphery and spinal cord and antioxidant treatment may be a promising strategy for anti-itch therapy.

Reactive oxygen species are the intermediates that are formed during the normal metabolic process which are effectively neutralized by the antioxidant system of the body.Any imbalance in this neutralization process causes oxidative stress which has been implicated as one of the cause in diseases such as Alzheimer's disease, cardiovascular disorders, cancer etc. Research has enabled the use of antioxidants as therapeutic agents in the treatment of various diseases. Literature also puts forth the negative effects of using antioxidants in the treatment of diseases. This review is a compilation of both the beneficial and detrimental effects of use of antioxidants in the treatment of diseases such as cancer, cardiovascular diseases, diabetes and oral diseases.

Single-atom nanozymes(SAzymes)are emerging as promising alternatives to mimic natural enzyme,which is due to high atomic utilization efficiency,well-defined geometric,and unique electronic structure.Herein,Fe single atoms supported on Ti_(3)C_(2)T_(x)(Fe-SA/Ti_(3)C_(2)T_(x))with intrinsic peroxidase activity is developed,further constructing a sensitive Raman sensor array for sensing of five antioxidants.Fe-SA/Ti_(3)C_(2)T_(x)shows excellent peroxidase-like performance in catalyzing the oxidation of 3,3',5,5'-tetramethylbenzidine(TMB)with colorimetric reactions.X-ray adsorption fine structure(XAFS)reveals that the electron transport between the Ti_(3)C_(2)T_(x)and Fe atoms occurs along Fe-O-Ti ligands,meanwhile the density functional theory(DFT)calculations confirm the spontaneous dissociation of H_(2)O_(2)and the formation of OH radicals.Furthermore,the peroxidase-like Fe-SA/Ti_(3)C_(2)T_(x)was used as surface enhanced Raman scattering(SERS)substrate of oxidized TMB(TMB+)and achieved satisfied signal amplification performance.Using the blocking effects of free radical reactions,one-off identification of 5 antioxidants,including ascorbic acid(AA),uric acid(UA),glutathione(GSH),melatonin(Mel),and tea polyphenols(TPP),could be realized with this high identifiable catalytic property.This principle could realize 100%distinguish accuracy combined with linear discriminant analysis(LDA)and heat map data analysis.A wide detection concentration ranges from 10^(-8)to 10^(-3)M for five antioxidants was also achieved.

Parkinson’s disease(PD) is a chronic progressive neurodegenerative disease. It results from the death of dopaminergic neurons. The pathophysiological mechanisms in idiopathic PD include the production of α-synuclein and mitochondrial respiratory function-affecting complex I, caused by reactive oxygen species.Therefore, the use of natural antioxidants in PD may provide an alternative therapy that prevents oxidative stress and reduces disease progression. In this review, the effects of hydroxytyrosol, Ginkgo biloba,Withania somnifera, curcumin, green tea, and Hypericum perforatum in PD animal and cell line models are compared and discussed. The reviewed antioxidants show evidence of protecting neural cells from oxidative stress in animal and cell models of PD. However, the clinical efficacy of these phytochemicals needs to be optimised and further investigated.

Effects of Plasticizers,antioxidants and Burning Rate Modifiers on Aging Performance of the HTPB/HMDI Composite Solid Propellant

The effects of plasticizers,antioxidants and burning rate modifiers on the aging performance of the composite solid propellant based on hydroxyl-terminated polybutadiene(HTPB)/hexamethylene diisocyanate(HMDI)were explored by apply-ing an accelerated aging program for 90 day at 70 ℃. The HTPB propellant matrix with the diisooctyl sebacate(DOS)as plasti-cizers and diisooctyl azelate(DOZ), antioxidants as N,N ′-Diphenyl-p-phenylenediamine(AO) and 2,2′-methylenebis(4-methyl-6-tert-butylphenol)(cyanox 2246)and burning rate modifiers as barium ferrite(BF),copper chromites(CC)and fer-ric oxide(FO)were varied. Results show that sample(S1)which based on DOS decreases the stress value and increases the strain value which considered to be an excellent start for aging program. Sample(S3)containing AO presents the higher resis-tance to oxidation showing the better performance that reflects on increasing the shelf life of the composite solid propellant mo-tor. Sample(S5)which based on BF enhances the ballistic performance among over the other tested two samples. The accelerat-ed aging program allowed us to estimate the motor in-service lifetime.

Role of mitochondria, oxidative stress and the response to antioxidants in myalgic encephalomyelitis/chronic fatigue syndrome: A possible approach to SARS-CoV-2 ’long-haulers’?

A significant number of SARS-CoV-2 (COVID-19) pandemic patients have developed chronic symptoms lasting weeks or months which are very similar to those described for myalgic encephalomyelitis/chronic fatigue syndrome. This study reviews the current literature and understanding of the role that mitochondria, oxidative stress and antioxidants may play in the understanding of the pathophysiology and treatment of chronic fatigue. It describes what is known about the dysfunctional pathways which can develop in mitochondria and their relationship to chronic fatigue. It also reviews what is known about oxidative stress and how this can be related to the pathophysiology of fatigue, as well as examining the potential for specific therapy directed at mitochondria for the treatment of chronic fatigue in the form of antioxidants. This study identifies areas which require urgent, further research in order to fully elucidate the clinical and therapeutic potential of these approaches.

Lanthanum chloride improves maize grain yield by promoting photosynthetic characteristics, antioxidants enzymes and endogenous hormone at reproductive stages

Despite an increase in application spectrum of rare earth elements in agriculture, all studies show that the suitable accumulation of rare earth elements can improve the crop seedling growth, but there is little research about REEs on physiological mechanisms of crops at reproductive stages. Therefore, this study was conducted to examine the possible potential benefits of lanthanum chloride(LaCl3) on the senescence and grain yield responses of maize. In this study, maize seeds were pre-treated by soaking with LaCl3 at the concentrations of 0(CK), 400(LC1), 800(LC2) and 1200(LC3) μmol/L, to evaluate its effect on the green leaf area, chlorophyll contents, photosynthesis, antioxidants, endogenous hormones in the later crop growth stages. The results show that LC1 and LC2 treatments evidently increase green leaf area, above ground dry biomass, accompanied by a distinct increase in the chlorophyll contents, and photo synthetic capacity, which promote the ear characteristics and grain yield of maize. In addition, LC1 and LC2 treatments simultaneously increase the activities of antioxidants, including superoxide dismutases, catalases, peroxidases, soluble protein, and enhanced levels of auxin, gibberellin and zeatin,following a dose-response tendency. Themalondialdehyde and abscisic acid levels transiently increase with the progression in the growth stage of the crop but are markedly decreased at LC1 and LC2 treatments, while LC3 treatment has no significant effect on malondialdehyde and even accelerates the accumulation of abscisic acid in maize leaves. Our data suggest that seed priming with LaCl3 at a suitable concentration range(400-800 μmol/L) can prolong the functional periods of leaves, increase photosynthetic capacity, enhance antioxidant activity, and alter endogenous hormone levels at reproductive stages, resulting in delaying leaf senescence rate and increasing yield. However, the moderate concentration of LaCl3 for maize is LC2(800 μmol/L), and can be effectively used to improve grain yield of maize.

Dear Editor, Mitochondrial complex I is important for cellular ATP producti on by tran sporti ng electro ns and gen erati ng proton gradient across the mitochondrial inner membrane (Hirst, 2013). It is also a major cellular locus where electron leakage to oxygen produces superoxide, an ROS (reactive oxygen species), particularly under oxidative stress conditions. Dysfunctional complex I contributes to the most comm on oxidative phosphorylation disorder in humans, with many iden tified gen etic mutati ons in complex I sub units causing a variety of human disorders including Leigh syndrome, encephalomyopathy, cardiomyopathy, parkinsonism and hereditary optic neuropathy (Hirst, 2013;Guo et al., 2017). In addition, complex I is the major target of many parkinsonismcausing neurotoxins including rotenone and MPTP. Past biochemical, cell biological and structural studies have elucidated how complex I functions normally in mitochondrial respiration (Hirst, 2013;Guo et al., 2017;Letts and Sazanov, 2017). Nonetheless, our understanding of mechanisms how complex I dysfunction leads to human diseases is far from completion;therapeutic targets and strategies are urgently needed.

Natural variability of essential oil and antioxidants in the medicinal plant Turnera diffusa

Objective:To evaluate differences in yield and composition of the essential oil and antioxidant contents in T.diffusa plants from localities in central region of Tamaulipas.Methods:Samples were collected in Tamaulipas,Mexico in the arid zone.Essential oil was obtained through steam distillation and analyzed using GC-MS.Polyphenol contents,antioxidant activities using ABTS and ferric reducing antioxidant power(FRAP) methods also were evaluated.Results:A total of 21 compunds were identified in the essential oils;nevertheless,only Eucalyptol,1,4-Methanocycloocta[d]pyridazine,1,4,4a,5,6,9,10,10a-octahydro-11,11-dimethyl-,(1à,4à,4aà,10aà) y Ethanone,1-(1,3-dimethyl-3-cyclohexen-1-yl) were detected in the three sites.Highest contents were registered in the sample from Padrón y Juárez with phenolic content of 33.85 mg GAE/g of dry material and antioxidant activities with ABTS 72.32% and with FRAP 21.33 mg GAE/g of dry material.Statistical differences were observed in essential oil,phenolics and antioxidants contents between populations.Conclusions:Results suggest that climatic differences and origin influence the phytochemicals in the medicinal plant T.diffusa,and thus,it is worth to consider such effects for industrial and medicinal purposes.
