Objective Our previous studies have demonstrated that neuropeptide S(NPS) respectively facilitates olfactory function and ameliorates olfactory spatial memory impairment through selective activation of the neurons bearing NPS receptor(NPSR) in the olfactory cortex and the subiculum complex. High level expression of NPSR m RNA in the posterior medial amygdala suggests that NPS-NPSR system might be involved in the regulation of defensive and risk assessment behaviors induced by alarm pheromones(APs). The present study was undertaken to investigate that the effect of intracerebroventricular(i.c.v.) injection of NPS or co-injection of NPSR antagonist on APs-induced defensive and risk assessment behaviors, and the NPS-induced activated neurons in the posterodorsal medial amygdaloid nuclei(MePD) and posteroventral medial amygdaloid nuclei(Me PV) in mice, respectively. Methods APs was harvested by electrical stimulation of the donor mouse’s perianal region, and control was collected by electrical stimulation of the neck skin. A modified open field chamber was used to detect the defensive and risk assessment behaviors. The potential neuronal targets of NPS in the Me PV and MePD were identified with c-Fos immunohistochemistry, and the coexpression of NPSR in the NPS-induced activated neurons was examined with dualimmunofluorescence. Results(1) APs significantly reduced the outside duration(p < 0.01) and walking distance(p < 0.01), meanwhile increased the conceal duration(p < 0.01) and headout duration(p < 0.01) compared to control.(2) Central administration of a range dose(0.1, 0.5 and 1 nmol) of NPS attenuated the APs-induced defensive and risk assessment behaviors. The dose of 0.5 nmol NPS showed the most effective.(3) [D-Val] NPS, a selective antagonist of NPSR, dosed at 20 and 40 nmol(i.c.v.) significantly dosedependently antagonized the inhibitory effects of NPS(0.5 nmol) on APs-induced defensive and risk assessment behaviors.(4) In comparison with vehicle, NPS signific

Abstract:Objective alarm pheromones(APs)are very important communicating substances in species survival,which conducts hazards,***,distress,presence of predators,*** the individual survival,sufficiently sleep is *** order to avoided predators,are the APs able to waking up mice during sleep time until *** Spherical Treadmill Olfaction-Sleep Recording System(STOSRS)was employed in this study,the mouse(C57BL/6J)head was restrained by a head plate,and the animal was able to maneuver on the top surface of an air-supported floating Styrofoam ***(which collected during killing of mice with CO electrical stimulation after anesthesia)directional/quantitative delivered during non-rapid eye movement sleep(NREM)or rapid eye movement sleep(REM)which were determined by EEG/EMG and digital *** waking latency(WL)and EEG power analysis were used to reveal the EEG/EMG characteristics of the arousal induced by APs in ***(l)Animals were able to sleeping(NREM and REM)on the STOSRS after one week *** EEG/EMGcharacteristics of the NREM and REM sleep were similar as unrestrained ***,when the mouse was movement on the Styrofoam ball,the EEG display typical 9 wave(4.5-9.0 Hz)same as REM sleep,meanwhile,the EMG shown high activity.(2)Air delivered into nasal cavity at NREM sleep,the WL was randomly from 5 s to 116 s(n=11).APs delivered display steady WL(9.31±0.71 s,n=8)and the EEG characteristics were changed to movement wave(described above).(3)During the REM sleep,as the same effects of air,the WL was randomly from 7.5 s to 138 s(n=5)after APs delivered.(4)The distribution of Grueneberg ganglion was 3D reconstruction by using OMP-GFP *** APs can awake mice within the NREM sleep,this pivotal effect protects animals to avoided predators and contributed to species survival.

Objective Pheromone was defined as a chemical released by one organism that modulates the behavior or physiology of a second organism of the same species. In mammals, pheromones regulate endocrine status;signal individual identity;and evoke instinctive sexual, nurturing, and fighting behaviors. When encountered dangerous stimuli, or subjected to such stressors as foot-shock, CO asphyxiation or injection of the pain-inducing hypertonic solution, mice often emit alarm pheromones(APs) to alert conspecifics or closely related species of the imminent danger. But the neural circuits to control the APs release had been rarely involved. Methods Neurotropic virus were injected in the bladder wall or the pontine micturition center(PMC) of male C57 BL/6 mice, and the function of circuits responsible for APs release were dissected through optogentics. Results Our preliminary data showed 1) the urine of stressed mice in the condition of a rat present contains APs which were aversive for conspecifics;2) the central regulatory networks of the bladder and preputial gland, both of which are the main source of pheromones in mice, are almost the same by injected the neurotropic pseudorabies virus(PRV152);meanwhile, by the modified rabies virus, mapped the monosynaptic input patterns to the pontine micturition center(PMC), which is proposed to be the integrative center for micturition and pheromone releasing;3) the circuit of vl PAG-PMC would be responsible for APs release. Conclusion The results showed vl PAG-PMC could be responsible for alarm pheromones releasing, which would lay foundation for future studies on the mechanism of fear and aversion, or the perception of APs. Understanding these circuits of micturition seems to offer a route to treatment of conditions, such as urgency incontinence or overactive bladder, in patients without overt neurological disease.
