Hepatocellular carcinoma(HCC),a common malignancy worldwide,still lacks effective clinical *** study aimed to investigate the oncogenes that affect the progression of HCC and their possible *** our study,we initially confirmed a higher level of PRDX2 in the bile of HCC patients compared to those with choledocholithiasis by 2-DE,LC-MS,and ***,we demonstrated the high expression of peroxiredoxin 2(PRDX2)in HCC based on the TCGA database and clinical sample ***,PRDX2 overexpression enhanced the viability of HCC *** PRDX2 silencing induced senescence of HCC *** vivo,knockdown of PRDX2 significantly reduced the weight of xenograft ***2 also was found to activate the wnt/β-catenin pathway by inducingβ-catenin nuclear ***,we proved that silencing PRDX2 could inhibit proliferation and wnt/β-catenin pathway while promoting senescence in HCC cells.

BACKGROUND Calculus bovis(CB),used in traditional Chinese medicine,exhibits anti-tumor effects in various cancer *** also constitutes an integral component of a compound formulation known as Pien Tze Huang,which is indicated for the treatment of liver ***,its impact on the liver cancer tumor microenvironment,particularly on tumor-associated macrophages(TAMs),is not well *** To elucidate the anti-liver cancer effect of CB by inhibiting M2-TAM polarization via wnt/β-catenin pathway *** This study identified the active components of CB using UPLC-Q-TOF-MS,evaluated its anti-neoplastic effects in a nude mouse model,and elucidated the underlying mechanisms via network pharmacology,transcriptomics,and molecular *** vitro assays were used to investigate the effects of CB-containing serum on HepG2 cells and M2-TAMs,and wnt pathway modulation was validated by real-time reverse transcriptase-polymerase chain reaction and Western blot *** This study identified 22 active components in CB,11 of which were detected in the *** investigations have demonstrated the ability of CB to effectively inhibit liver tumor *** integrated approach employing network pharmacology,transcriptomics,and molecular docking implicated the wnt signaling pathway as a target of the antineoplastic activity of CB by suppressing M2-TAM *** vitro and in vivo experiments further confirmed that CB significantly hinders M2-TAM polarization and suppresses wnt/β-catenin pathway *** inhibitory effect of CB on M2-TAMs was reversed when treated with the wnt agonist SKL2001,confirming its pathway *** This study demonstrated that CB mediates inhibition of M2-TAM polarization through the wnt/β-catenin pathway,contributing to the suppression of liver cancer growth.

Increasing the osteogenic differentiation ability and decreasing the adipogenic differentiation ability of bone marrow mesenchymal stem cells(BMSCs)is a potential strategy for the treatment of osteoporosis(OP).Naturally derived oligosaccharides have shown significant anti-osteoporotic ***(NST),an oligosaccharide,was isolated from the roots of Morinda officinalis How.(MO).The aim of the present study was to investigate the effects of NST on bone loss in ovariectomized mice,and explore the underlying mechanism of NST in promoting differentiation of BMSCs to *** of NST(40,80 and 160 mg/kg)and the positive control of estradiol valerate(0.2 mg/kg)for 8 weeks significantly prevented bone loss induced by ovariectomy(OVX),increased the bone mass density(BMD),improved the bone microarchitecture and reduced urine calcium and deoxypyridinoline(DPD)in ovariectomized mice,while inhibited the increase of body weight without significantly affecting the uterus ***,we found that NST increased osteogenic differentiation,inhibited adipogenic differentiation of BMSCs in vitro,and upregulated the expression of the key proteins of BMP and wnt/β-catenin *** addition,Noggin and Dickkopf-related protein-1(DKK-1)reversed the effect of NST on osteogenic differentiation and expression of the key proteins in BMP and wnt/β-catenin *** luciferase activities and the molecular docking analysis further supported the mechanism of *** conclusion,these results indicating that NST can be clinically used as a potential alternative medicine for the prevention and treatment of postmenopausal osteoporosis.

Radiotherapy for head and neck cancer can cause serious side effects, including severe damage to the salivary glands, resulting in symptoms such as xerostomia, dental caries, and oral infection. Because of the lack of long-term treatment for the symptoms of xerostomia, current research has focused on finding endogenous stem cells that can differentiate into various cell lineages to replace lost tissues and restore functions. Here, we report that Sox9^(+)cells can differentiate into various salivary epithelial cell lineages under homeostatic conditions. After ablating Sox9^(+) cells, the salivary glands of irradiated mice showed more severe phenotypes and the reduced proliferative capacity. Analysis of online single-cell RNAsequencing data reveals the enrichment of the wnt/β-catenin pathway in the Sox9^(+) cell ***, treatment with a wnt/β-catenin inhibitor in irradiated mice inhibits the regenerative capability of Sox9^(+) cells. Finally, we show that Sox9^(+) cells are capable of forming organoids in vitro and that transplanting these organoids into salivary glands after radiation partially restored salivary gland *** results suggest that regenerative therapy targeting Sox9^(+) cells is a promising approach to treat radiation-induced salivary gland injury.
