WSB1 regulates c-Myc expression through β-catenin signaling and forms a feedforward circuit

The dysregulation of transcription factors is widely associated with tumorigenesis.As the most well-defined transcription factor in multiple types of cancer,c-Myc can transform cells by transactivating various downstream genes.Given that there is no effective way to directly inhibit c-Myc,c-Myc targeting strategies hold great potential for cancer therapy.In this study,we found that WSB1,which has a highly positive correlation with c-Myc in 10 cancer cell lines and clinical samples,is a direct target gene of c-Myc,and can positively regulate c-Myc expression,which forms a feedforward circuit promoting cancer development.RNA sequencing results from Bel-7402 cells confirmed that WSB1 promoted cMyc expression through theβ-catenin pathway.Mechanistically,WSB1 affectedβ-catenin destruction complex-PPP2CA assembly and E3 ubiquitin ligase adaptorβ-TRCP recruitment,which inhibited the ubiquitination ofβ-catenin and transactivated c-Myc.Of interest,the effect of WSB1 on c-Myc was independent of its E3 ligase activity.Moreover,overexpressing WSB1 in the Bel-7402 xenograft model could further strengthen the tumor-driven effect of c-Myc overexpression.Thus,our findings revealed a novel mechanism involved in tumorigenesis in which the WSB1/c-Myc feedforward circuit played an essential role,highlighting a potential c-Myc intervention strategy in cancer treatment.

Upregulation of REL and WSB1 in Patients With Psoriasis and Metabolic Syndrome

Objective:A strong association exists between metabolic syndrome and psoriasis.The current study was performed to explore the gene regulation of metabolic syndrome in patients with psoriasis.Methods:Patients were assessed for psoriasis and metabolic syndrome clinically (Psoriasis Area and Severity Index,height,weight,waist circumference,and blood pressure) and biochemically (lipid profile and fasting blood sugar concentration).Systemic gene regulation was first explored by microarray and analyzed using Transcriptome Analysis Console Software,after which two selected upregulated genes were further validated using polymerase chain reaction and enzyme-linked immunosorbent assay and analyzed using independent samplet test.Results:The analysis showed 7,269 upregulated and 3 downregulated genes at a fold change of 2 andP value of 17 genes were upregulated and none were downregulated at a fold change of 8 andP value of 16 biomarkers aligned with the gene regulation observed in the current study.In particular,theREL transcript was upregulated 12-fold (P = 8.16 × 10-17),while theWSB1 transcript was upregulated 9-fold (P = 9.87 × 10-13).Validation showed thatREL was also upregulated 2-fold in the polymerase chain reaction,and its protein was expressed at 7.140 ng/mLvs.undetectable levels in the cases (P = 0.048).However,WSB1 was upregulated 2-fold in the polymerase chain reaction compared with controls,and unexpectedly,its protein was undetectable in cases but detectable in controls (P = 0.018).Conclusion:The upregulation ofREL andWSB1 was observed in patients with psoriasis and metabolic syndrome,the clinical application ofREL andWSB1 as biomarkers needs further validation for potential future implications in clinical practice.

恶性肿瘤是严重威胁人类生命健康的重大疾病之一。近年来,已发展了多种分子靶向治疗、肿瘤免疫疗法、细胞疗法等针对肿瘤的治疗策略,一定程度上提高了患者生活质量,延长了生存期。然而,恶性肿瘤是一种由多因素共同调控,发病机制极其复杂的疾病,肿瘤的防治仍面临诸多挑战。因此,发现基于新靶标、新机制的肿瘤治疗策略仍是肿瘤治疗的热点,具有重要的研究价值和社会意义。针对神经母细胞瘤和肿瘤转移的干预,本论文一方面采用诱导神经母细胞瘤Neuro2a细胞分化的表型筛选模型,从内部化合物库中发现了具有一定促Neuro2a细胞分化先导分子E22,另一方面基于抗肿瘤转移的表型筛选方法(划痕模型)及机制探索,从内部化合物发现了具有抗肿瘤转移活性的新型WSB1抑制剂先导分子W15,进一步地,本论文开展较为系统的构效关系研究,具体研究工作分述如下:新型促神经母细胞瘤分化的先导分子的发现和优化:神经母细胞瘤的诱导分化治疗被认为是一种有效的治疗策略,但临床上小分子药物只有13-cis RA作为诱导分化剂用于高危NB病人的维持治疗。因此,本论文从课题组内部化合物库出发(22个结构多样、类药化合物),利用Neuro2a作为表型筛选细胞,以细胞神经轴突平均长度作为诱导分化评价指标,发现了结构新颖、作用机制独特的NB细胞诱导分化调节剂E22。首先,与空白组相比,其轴突平均长度较空白组增长了55%,与阳性药13-cis RA(轴突平均长度增长了69%)活性相当。进一步地,本论文通过生物电子等排,骨架跃迁及环合策略,设计、合成了6个系列,共计25个化合物。以Neuro2a细胞的分化比率、细胞轴突平均长度以及最大轴突长度三方面评价目标化合物的促分化活性。结果表明,有10余个化合物促分化活性要优于13-cis RA阳性对照,绝大部分化合物表现出良好的细胞活性,IC在5μM以下。令人感兴趣的是,通过环合策略得到的系列VI化合物,其促分化活性得到了显著提升,尤其是化合物1-95和1-97,分化比率分别为11.73和9.71,分别是阳性药13-cis RA的5.6倍和4.6倍。优选了10个化合物进行细胞周期考察,发现该类化合物能引起细胞G0/G1期阻滞,进一步说明了该类化合物具有诱导细胞分化作用。体外代谢性质研究表明,高活性化合物1-91,1-95和1-97表现出优异的全血稳定性以及中等的肝微粒体稳定性。针对肿瘤微环境的低氧靶标wsb-1的先导分子发现和优化:肿瘤转移往往与肿瘤缺氧微环境关系密切,肿瘤细胞缺氧往往导致肿瘤更具转移性和侵袭性。wsb-1是在缺氧下肿瘤转移的关键蛋白,与肿瘤转移能力密切相关,针对WSB1开发特异性的抑制剂具有重要的研究价值。因此,本论文以前期表型筛选发现的化合物W15作为先导分子,通过生物电子等排,骨架跃迁等合理的药物设计策略,设计、合成了4个系列,共计33个化合物。Wound healing活性测试结果表明,部分化合物在SW1990和H460细胞上都具有良好的抗转移活性,且对细胞毒性较小(IC大于20μM)。在SW1990和H460细胞上抗转移活性优于W15的化合物各有6个。代表性化合物2-18和2-51都能显著的抑制WSB1对下游蛋白RhoGDI的降解和对MORF4L1的泛素化作用,且优于化合物W15。此外,化合物2-18还能显著抑制WSB1过表达的KHOS细胞(KHOS-WSB1)的迁移,抗迁移活性与阳性相当。因此,化合物2-18具有明确的WSB1通路抑制和抗肿瘤转移活性,具有进一步开发价值。综上,本论文针对神经母细胞瘤分化异常和肿瘤微环境低氧靶标wsb-1,通过基于表型筛选、生物电子等排、骨架迁越等合理的药物设计,分别发现了具有高活性的促细胞分化先导分子1-95、1-97和高特异性靶向wsb-1的先导分子2-18,为神经母细胞瘤的促分化治疗和靶向缺氧微环境的抗肿瘤转移治疗奠定了基础。

Intratumoral hypoxia occurs in many solid tumors, where it is associated with the development of metastatic character. However, the connections between these phenomena are not fully understood. In this study, we define an integrative role for the E3 ubiquitin ligase subunit WSB1. In primary osteosarcomas, increased levels of WSB1 correlated with pulmonary metastatic potential. RNAi-mediated attenuation of WSB1 or disruption of its E3 ligase activity potently suppressed tumor metastasis. Quantitative proteomic and functional analyses revealed that WSB1 ubiquitylates the Rho-binding protein RhoGDI2 and promotes its proteasomal degradation, thereby activating Rac1 to stimulate tumor cell motility and invasion. Our findings show how WSB1 regulates key steps of the metastatic cascade in hypoxia-driven osteosarcoma, and they highlight a candidate therapeutic target to potentially improve the survival of patients with metastatic disease.
