Clinicopathological and immunological features of new onset kidney disease: a rare event after SARS-CoV-2 Vaccination

The onset of various kidney diseases has been reported after severe acute respiratory syndrome coronavirus2(SARS-CoV-2) Vaccination.However,detailed clinical and pathological features are lacking.We screened and analyzed patients with newly diagnosed kidney diseases after inactivated SARS-CoV-2 Vaccination in Peking University First Hospital from January 2021 to August 2021,and compared them with the reported cases in the literature.We obtained samples of blood,urine and renal biopsy tissues.Clinical and laboratory information,as well as light microscopy,immunostaining and ultrastructural observations,were described.The SARS-CoV-2 spike protein and nucle oprotein were stained using the immunofluorescence technique in the kidney biopsy samples.SARS-CoV-2 specific antibodies were tested using magnetic particle chemiluminescence immunoassay.The study group included 17 patients with a range of conditions including immune-complex-mediated kidney diseases(IgA nephropathy,membranous nephropathy and lupus nephritis),podocytopathy(minimal change disease and focal segmental glomerulosclerosis) and others(antineutrophil-cytoplasmic-antibody-associated vasculitis,anti-glomerular basement membrane nephritis,acute tubulointerstitial nephritis and thrombotic micro angiopathy).Seven patients(41.18%)developed renal disease after the first dose and ten(58.82%) after the second dose.The kidney disease spectrum as well as clinicopathological features are similar across different types of SARS-CoV-2 vaccines.We found no definitive evidence of SARS-CoV-2 spike protein or nucleoprotein deposition in the kidney biopsy samples.Seropositive markers implicated abnormal immune responses in predisposed individuals.Treatment and follow-up(median=86 days) showed that biopsy diagnosis informed treatment and prognosis in all patients.In conclusion,we observed various kidney diseases following SARS-CoV-2 vaccine administration,which show a high consistency across different types of SARS-CoV-2 vaccines.Our findings provide evidence against direct

Amoeba-inspired magnetic microgel assembly assisted by engineered dextran-binding protein for Vaccination against lifethreatening systemic infection

Vaccination is critical for population protection from pathogenic infections.However,its efficiency is frequently compromised by a failure of antigen retention and presentation.Herein,we designed a dextran-binding protein DexBP,which is composed of the carbohydrate-binding domains of Trichoderma reesei cellobiohydrolases Cel6A and Cel7A,together with the sequence of the fluorescent protein mCherry.DexBP was further prepared by engineered Escherichia coli cells and grafted to magnetic nanoparticles.The magnetic nanoparticles were integrated with a dextran/poly(vinyl alcohol)framework and a reactive oxygen species-responsive linker,obtaining magnetic polymeric microgels for carrying pathogen antigen.Similar to amoeba aggregation,the microgels self-assembled to form aggregates and further induced dendritic cell aggregation.This step-by-step assembly retained antigens at lymph nodes,promoted antigen presentation,stimulated humoral immunity,and protected the mice from lifethreatening systemic infections.This study developed a magnetic microgel-assembling platform for dynamically regulating immune response during protection of the body from dangerous infections.

Background:Coronavirus disease 2019(COVID-19)has potential risks for both clinically worsening pulmonary hypertension(PH)and increasing mortality.However,the data regarding the protective role of Vaccination in this population are still lacking.This study aimed to assess the safety of approved Vaccination for patients with PH.Methods:In this national prospective cohort study,patients diagnosed with PH(World Health Organization[WHO]groups 1 and 4)were enrolled from October 2021 to April 2022.The primary outcome was the composite of PH-related major adverse events.We used an inverse probability weighting(IPW)approach to control for possible confounding factors in the baseline characteristics of patients.Results:In total,706 patients with PH participated in this study(mean age,40.3 years;mean duration after diagnosis of PH,8.2 years).All patients received standardized treatment for PH in accordance with guidelines for the diagnosis and treatment of PH in China.Among them,278 patients did not receive Vaccination,whereas 428 patients completed the Vaccination series.None of the participants were infected with COVID-19 during our study period.Overall,398 patients received inactivated severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)vaccine,whereas 30 received recombinant protein subunit vaccine.After adjusting for baseline covariates using the IPW approach,the odds of any adverse events due to PH in the vaccinated group did not statistically significantly increase(27/428[6.3%]vs.24/278[8.6%],odds ratio=0.72,P=0.302).Approximately half of the vaccinated patients reported at least one post-Vaccination side effects,most of which were mild,including pain at the injection site(159/428,37.1%),fever(11/428,2.6%),and fatigue(26/428,6.1%).Conclusions:COVID-19 Vaccination did not significantly augment the PH-related major adverse events for patients with WHO groups 1 and 4 PH,although there were some tolerable side effects.A large-scale randomized controlled trial is warranted to confirm this finding.The final approval

During the coronavirus disease 2019(COVID-19)pandemic,vaccines help control the spread of infection.To date,47 vaccines have been approved,with another 227 candidates in various stages of development.In the short period of time since the beginning of their use,evidence has begun to emerge of complications following Vaccination in the form of the development or exacerbation of a number of pathological conditions(Block et al.,2022;Haseeb et al.,2022).For example,a population-based study in France identified 1612 cases of myocarditis and 1613 cases of pericarditis requiring hospital treatment within five months of Vaccination(le Vu et al.,2022).

Summary What is already known about this topic?Herpes zoster(shingles)is a common skin condition in older adults,which usually presents as a painful rash with blisters.Vaccination is the most effective method to prevent shingles.However,there is not sufficient population-based epidemiological data in China to optimize the timing of zoster Vaccination.What is added by this report?Clustering analyses of population-wide epidemiological data from the Healthcare Big Data Platform in Yichang,China showed that the average annual zoster incidence is the highest among people 55 years or older,at 10 cases per thousand persons per year,making this age group the optimal target population for Vaccination.Incidence was lower but increased with age among younger adults,28–54 years old.What are the implications for public health practice?With limited Vaccination resources,zoster Vaccinations should be targeted at adults 55 years or older who are at the greatest risk for shingles.Research should be conducted to understand the risk of shingles among young and middle-aged adults and identify triggers of shingles:potentially leading to preventive measures.

Impact of Vaccination against COVID-19 on the outcomes of in vitro fertilization–embryo transfer:a retrospective cohort study

Background: Vaccination against coronavirus disease 2019 (COVID-19) has become the primary approach in the fight against the spread of COVID-19. Studies have shown that Vaccination against COVID-19 has adverse effects, particularly on human reproductive health, despite the fact that Vaccination rates are still on the rise. However, few studies have reported whether Vaccination affects the outcome of in vitro fertilization-embryo transfer (IVF-ET) or not. In this study, we compared the outcome of IVF-ET and the development of follicles and embryos between vaccinated and unvaccinated groups. Methods: A single-center retrospective cohort study of 10,541 in vitro fertilization (IVF) cycles was conducted from June 2020 to August 2021. 835 IVF cycles with a history of Vaccination against COVID-19 and 1670 IVF cycles that served as negative controls were selected and analyzed utilizing the Matchlt package of R software (http://***/) and the nearest neighbor matching algorithm for propensity-matched analysis at a 1:2 ratio. Results: The number of oocytes collected in the vaccinated group and the unvaccinated group were 8.00 (0, 40.00) and 9.00 (0, 77.00) ( P = 0.073) and the good-quality embryo rates of the two groups were 0.56±0.32 and 0.56±0.31 averagely ( P = 0.964). Clinical pregnancy rates for the vaccinated group and unvaccinated group were 42.4% (155/366) and 40.2% (328/816) ( P = 0.486) and biochemical pregnancy rates were 7.1% (26/366) and 8.7% (71/816) ( P = 0.355). Two other factors were analyzed in this study;Vaccination among different genders and different types (inactivated vaccine or recombinant adenovirus vaccine) showed no statistically significant effect on the above outcomes. Conclusions: In our findings, Vaccination against COVID-19 showed no statistically significant effect on the outcomes of IVF-ET and the development of follicles and embryos, nor did the gender of the vaccinated person or the formulation of vaccines show significant effects.

Although the development of covID-19 vaccines has been a remarkable success,the heterogeneous individual antibody generation and decline over time are unknown and still hard to predict.In this study,blood samples were collected from 163 participants who next received two doses of an inactivated COvVID-19 vaccine(CoronaVac)at a 28-day interval.Using TMT-based proteomics,we identified 1,715 serum and 7,342 peripheral blood mononuclear cells(PBMCs)proteins.We proposed two sets of potential biomarkers(seven from serum,five from PBMCs)at baseline using machine learning,and predicted the individual seropositivity 57 days after Vaccination(AUC=0.87).Based on the four PBMC's potential biomarkers,we predicted the antibody persistence until 180 days after Vaccination(AUC=0.79).Our data highlighted characteristic hematological host responses,including altered lymphocyte migration regulation,neutrophil degranulation,and humoral immune response.This study proposed potential blood-derived protein biomarkers before Vaccination for predicting heterogeneous antibody generation and decline after coVID-19 Vaccination,shedding light on immunization mechanisms and individual booster shot planning.

Mpox (monkeypox) knowledge, concern, willingness to change behaviour, and seek Vaccination: Results of a national cross-sectional survey

Background: In mid-2022, a global mpox (formerly 'monkeypox') outbreak affecting predominantly gay and bisexual men emerged in non-endemic countries. Australia had never previously recorded mpox cases and there was no prior research on knowledge or attitudes to mpox among gay and bisexual men across Australia. Methods: We conducted a national, online cross-sectional survey between August 2022 and September 2022. Participants were recruited through community organisation promotions, online advertising, and direct email invitations. Eligible participants were gay, bisexual or queer; identified as male (cisgender or transgender) or non-binary; aged 16 years or older; and lived in Australia. The main outcome measures were: knowledge and concern about mpox; recognition of mpox symptoms and transmission routes; Vaccination history; acceptability of behavioural changes to reduce mpox risk, and willingness to be vaccinated. Results: Of 2287 participants, most participants were male (2189/2287; 95.7%) and gay (1894/2287; 82.8%). Nearly all had heard about mpox (2255/2287; 98.6%), and the majority were concerned about acquiring it (1461/2287; 64.4%). Most of the 2268 participants not previously diagnosed with mpox correctly identified skin lesions (2087; 92%), rash (1977; 87.2%), and fever (1647; 72.6%) as potential symptoms, and prolonged and brief skin-To-skin contact as potential ways to acquire mpox (2124, 93.7%; and 1860, 82%, respectively). The most acceptable behavioural changes were reducing or avoiding attendance at sex parties (1494; 65.9%) and sex-on-premises venues (1503; 66.4%), and having fewer sexual partners (1466; 64.6%). Most unvaccinated and undiagnosed participants were willing to be vaccinated (1457/1733; 84.1%). Conclusions: People at risk of mpox should be supported to adopt acceptable risk reduction strategies during outbreaks and to seek Vaccination. © 2023 CSIRO. All rights reserved.
