triterpene acids（TAs） are the major bioactive constituents in the medicinal fungus Ganoderma ***, fermentative production of TAs has not been optimized for commercial use, and whether the TAs isolated from submerged culture G. lingzhi mycelia possess antitumor activity needs to be further proven. In this study, enhanced TA yield and productivity were attained with G. lingzhi using response surface methodology. The interactions of three variables were studied, i.e., initial pH, dissolved oxygen（DO） and fermentation temperature,using the Box-Benhnken design. The optimum conditions were an initial pH of 5.9, 20.0 % DO and 28.6°C. These conditions resulted in a TA yield of 308.1 mg/L in a 5-L stirred bioreactor. Furthermore, the optimized conditions were then successfully scaled up to a production scale of 200 L, and maximum TA production and productivity of295.3 mg/L and 49.2 mg/L per day were achieved, which represented 80.9% and 111.5% increases compared with the non-optimized conditions, respectively. Additionally, the triterpene acid extract（TAE） from G. lingzhi mycelia was first found to be cytotoxic to the SMMC-7721 and SW620 cell lines in vitro, and the TAE exhibited dose-dependent antitumor activity against the solid tumor sarcoma 180 in vivo. Chemical analysis revealed that the key active triterpene compounds, ganoderic acid T and ganoderic acid Me, dominantly existed in the extract.

Akebia trifoliata(Thunb.) Koidz has long been used as a diuretic and an antiphlogistic in traditional Chinese medicine for *** far,phytochemical studies on the stems of this plant have revealed structurally diverse triterpenoids,phenolics and *** pericarp of *** was deduced to be rich in bioactive chemicals which might be ideal candidates that could be developed as effective and safe agents for pharmaceutical ***,those potential bioactive compounds in the pericarps have yet not well been ***,we carried out a phytochemical study on the pericarps of ***,by which a series of oleanane-type triterpenoids were identified and some of them showed antimicrobial *** continuation of this work,six new triterpene acids(1-6) were further *** on extensive spectroscopic analysis,these new structures were elucidated as 2α,3β,23-trihydroxymultiflor-7-en-28-oic acid(1),2α,3β,23-trihydroxy-taraxastan-28,20β-olide(2),2,29-dihydroxy-3-oxo-23-noroleana-1,4,12-trien-28-acid(3),3-hydroxy-2-oxo-23-noroleana-3,12-dien-28-acid(4),2-hydroxy-3-oxo-23,29-dinor oleana-1,4,12-trien-28-acid(5) and 4-oxo-3,23-dinor-2,3-seco-olean-12-ene-2,28-diate(6).In vitro antibacterial assay revealed that compounds 3-5 were active against tested gram-(+) bacteria Staphyloccocus Aureus(CMCC26003),Bacillus Cereus(CMCC63302) and Bacillus Subtilis(CMCC63501) with MIC values ranging from 6.25 to 12.5 μg/***,compounds 3-5 were further revealed to show in vitro growth inhibitory activity against human tumor A549 and HeLa cell lines.

Thin Layer Chromatography and Chemometric Studies of Selected Potentilla Species

Herbal medicinal products have a long-standing therapeutic record. To ensure the quality of herbal products specific identification tests which allow experts to discriminate related species and/or potential adulterants/substitutes are required. The purpose of the research was to recommend an original and simple method for the separation of closely related five triterpenic acids (ursolic, oleanolic, tormentic, euscaphic, pomolic acid) and its application to chemotaxonomy studies. 17 standard samples of Potentilla species and 3 test samples were chromatographed (with or without prechromatographic derivatization) on silica gel plates using the mobile phase: chloroform-diethyl ether-methanol-formic acid (30:10:1:0.2 v/v/v/v);they were subsequently derivatized, and visualized in UV 366 nm light. After images received pre-treatment (montaging stack, removal of noise, background subtract, horizontal equalization, two types of warping) exploratory analysis of the investigated Potentilla species fingerprints was processed. The method without prechromatographic derivatization was used to create differential fingerprints. Good results were obtained by the application of r-project iterative algorithm for subtract background and our simple algorithm for warping to image processing of TLC chromatograms for the first time.

BACKGROUND:triterpenes（especially triterpene acids） is one of the major physiologically active components of Ganoderma ***,submerged fermentation techniques of triterpenes is not optimized for commercial use,as total triterpene yield is still very *** aims are to develop a two-stage p H control strategy integrated with a stimulus,an ether extract（EECM,from the medicinal insect,Catharsius molossus）,for enhanced *** triterpene acid ***:The triterpene acid production of 302.76 mg/L was obtained in the *** culture with EECM,which was higher by 60% compared with control cultures not fed *** on the feeding EECM method,a new two-stage p H control strategy was integrated resulting in triterpene acid yields of 361.47 mg/*** studies optimized interactions between EECM feeding and two-stage p H control strategies,which resulted in an optimal EECM concentration of 185.89 mg/L,optimal p H values of 5.07 in the first 72 h incubation and 4.37 after 72 h with a maximum triterpene acid concentration of 449.37 mg/*** represented a 137% increase compared to that of the control ***,using a 150-L fermenter under these conditions,a triterpene acid concentration of 431.83 mg/L was ***:Enhanced production of triterpene acid in submerged cultures of *** was achieved using an EECM feeding stimulus integrated with a two-stage p H control strategy in a large scale.
