Phospholipase D(PLD)and its product phosphatidic acid(PA)function as pleiotropic factors in the regulation of cancer progression,which includes the promotion of cell growth,survival,cell migration,and angiogenesis.1 Chemical inhibition of the enzymatic activity of PLD1 and PLD2,or genetic down-regulation of their expression both induce tumor suppression,1 while the compounds for PLD expression inhibition are barely reported.Here,we astonishingly found that the anti-hyperglycemic agents biguanides decreased PLD1 protein expression in cancer cells and further investigated its underlying regulatory mechanism.

Major depressive disorder(MDD)is a common psychiatric condition characterized by two main symptoms,low mood and anhedonia.About 15–30%of people suffering from MDD do not respond to standard-of-care antidepressants,e.g.,the serotonin re-uptake inhibitors(SSRI),and are considered affected by Treatment-Resistant Depression(TRD).The neurobiology of this condition is presently unknown.Recent attempts of developing novel treatments for TRD have been driven by four major breakthroughs:(1)Increasing dopaminergic neurotransmission improves TRD symptoms;(2)Anhedonia occurs when central dopaminergic neurotransmission is low;(3)Enhanced neuroplasticity is critical for the action of antidepressants;(4)Ketamine shows antidepressant properties in TRD patients and triggers neuroplasticity in preclinical animal models.These breakthroughs are at the basis of a putative human Translational cellular model for antidepressant agents that we are proposing in this article.The rationale is briefly described here.

Protein kinases are a conserved superfamily of proteins that play an essential role in signal transduction by attaching a phosphoryl group to serine,threonine,or tyrosine residues of a protein,hence converting them to phosphoproteins.This process,which can be reversed by protein phosphatases,regulates vital cellular processes in plants,including metabolism,proliferation,and death.It is also key to trigger response mechanisms to drought,increased salinity,and other abiotic stresses(Zhu,2016).

Recent studies have shown that global Translational reprogramming is an early activation event in pattern-triggered immunity,when plants recognize microbe-associated molecular patterns.However,it is not fully known whether Translational regulation also occurs in subsequent immune responses,such as effector-triggered immunity(ETI).In this study,we performed genome-wide ribosome profiling in Arabidopsis upon RPS2-mediated ETI activation and discovered that specific groups of genes were Translationally regulated,mostly in coordination with transcription.These genes encode enzymes involved in aromatic amino acid,phenylpropanoid,camalexin,and sphingolipid metabolism.The functional significance of these components in ETI was confirmed by genetic and biochemical analyses.Our findings provide new insights into diverse Translational regulation of plant immune responses and demonstrate that Translational coordination of metabolic gene expression is an important strategy for ETI.

Neuroimmune system may be involved in the pathological process of bipolar disorder(BD),but the essential association is not fully understood.Accumulating evidence has shown that BD involves the activation of immune cells and the release of inflammatory substances in the central nerve system(CNS).Meanwhile,neuroimmune responses also interact with other hypothesis of the etiology of BD that are widely recognized,such as neurotransmitter systems,neuroendocrine systems,neurotrophic factors,and oxidative stress.Simultaneously,related genes and immune changes in peripheral blood vary with it.Overall,neuroimmunity may play an important role in the pathogenesis of BD,and the inflammatory cytokines,especially interleukin-6 and tumor necrosis factor-alpha,have potential value for the clinical diagnosis and prognosis of BD,as well as predicting the therapeutic effects of drugs.Large-scale studies are needed to extend the evidence on neuroimmunity in BD,and to examine its clinical value for applications such as early prediction and treatment.

Dr.Sonenberg’s group has significantly contributed to the understanding of Translational control in learning and memory.In a recent Nature article,they further reported the cell-typespecific Translational control underlying memory consolidation.1 Long-term potentiation(LTP)has been widely acknowledged as one of the main cellular models of learning and memory.Generally,there are two components of LTP driven by different stimulations and mechanisms.The early phase of LTP(E-LTP)and short-term memory(STM)are evoked for short periods and are temporary,relying primarily on modifications of pre-existing proteins.The late phase of LTP(L-LTP)and consequent longterm memory(LTM)are elicited by strong and repetitive stimulation and are long-lasting,requiring new protein synthesis.

Tumor suppressor genes cooperate with each other in tumors.Three important tumor suppressor proteins,retinoblastoma(Rb),p53,phosphatase,and tensin homolog deleted on chromosome ten(PTEN)are functionally associated and they regulated by postTranslational modification(PTMs)as well.PTMs include phosphorylation,SUMOylation,acetylation,and other novel modifications becoming growing appreciated.Because most of PTMs are reversible,normal cells use them as a switch to control the state of cells being the resting or proliferating,and PTMs also involve in cell survival and cell cycle,which may lead to abnormal proliferation and tumorigenesis.Although a lot of studies focus on the importance of each kind of PTM,further discoveries shows that tumor suppressor genes(TSGs)form a complex“network”by the interaction of modification.Recently,there are several promising strategies for TSGs for they change more frequently than carcinogenic genes in cancers.We here review the necessity,characteristics,and mechanisms of each kind of post-Translational modification on Rb,p53,PTEN,and its influence on the precise and selective function.We also discuss the current antitumoral therapies of Rb,p53 and PTEN as predictive,prognostic,and therapeutic target in cancer.

Protein synthesis involves initiation,elongation,and termination.A number of studies show that translation elongation is highly regulated and is the most energy-intensive process,which controls the efficiency and accuracy of protein synthesis.In the elongation process,eukaryotic elongation factor 2(eEF2)binds to the ribosome to induce its conformation change and promotes the translocation of the tRNAs by hydrolyzing guanosine triphosphate,which then allows the next AA-tRNA to enter the A site for translation elongation.The activity of eEF2 is negatively regulated by its kinase eEF2K that phosphorylates eEF2 at threonine-56(Thr56).
