Molecular characteristics and temperature tolerance function of the transient receptor potential in the native Bemisia tabaci AsiaII3 cryptic species

Insects are poikilothermic animals,and temperature is one of the most important abiotic factors affecting their spread and distribution.For example,differences in thermal tolerance may underlie the significant differences in geographical distributions between the native AsiaII3 and invasive MED(Mediterranean)cryptic Bemisia tabaci species in China.transient receptor potential(TRP)channels are key components of the insect temperature perception system and act as molecular thermometers since they can be activated by specific changes in temperature.In this study,we cloned and characterized the AsiaII3 BtTRP gene and revealed its functions in the response to thermal stress.The full-length cDNA of BtTRP was 3821 bp,with a 3501-bp open reading frame encoding a 132.05-kDa protein.Comparing the deduced amino acid sequences of AsiaII3 BtTRP and MED TRP revealed five amino acid differences.In situ hybridization indicated that BtTRP might be widely expressed throughout the AsiaII3 adult body.BtTRP mRNA expression reached the highest levels after exposure to mild thermal stimuli(12 and 35°C),showing that BtTRP expression can be induced by temperature stress.Furthermore,the thermal tolerance of AsiaII3 after BtTRP dsRNA feeding was significantly lower than that of the control.Taken together,the present study highlights the importance of TRP channels for B.tabaci thermal resistance,and allows us to infer that the differences in amino acids between AsiaII3 and MED might cause the differences in thermal tolerance of these two cryptic species.This study provides a new direction for investigating geographic distribution differences between invasive and native insects.

Breathing is an intrinsic natural behavior and physiological process that maintains life.The rhythmic exchange of gases regulates the delicate balance of chemical constituents within an organism throughout its lifespan.However,chronic airway diseases,including asthma and chronic obstructive pulmonary disease,affect millions of people worldwide.Pathological airway conditions can disrupt respiration,causing asphyxia,cardiac arrest,and potential death.The innervation of the respiratory tract and the action of the immune system confer robust airway surveillance and protection against environmental irritants and pathogens.However,aberrant activation of the immune system or sensitization of the nervous system can contribute to the development of autoimmune airway disorders.transient receptor potential ion channels and voltage-gated Na+channels play critical roles in sensing noxious stimuli within the respiratory tract and interacting with the immune system to generate neurogenic inflammation and airway hypersensitivity.Although recent studies have revealed the involvement of nociceptor neurons in airway diseases,the further neural circuitry underlying airway protection remains elusive.Unraveling the mechanism underpinning neural circuit regulation in the airway may provide precise therapeutic strategies and valuable insights into the management of airway diseases.

Pulmonary artery hypertension(PAH) is associated with profound vascular remodeling and alterations in Ca homeostasis in pulmonary arterial smooth muscle cells(PASMCs).Recent studies show that chronic hypoxia upregulates expression of canonical transient receptor potential(TRPC) genes,which encode store-operated and receptor-operated cation channels,and enhances both store-and receptor-operated Ca entries in PASMCs.However, whether the phenomenon was existed in monocrotaline(MCT)-induced PAH has not been determined.Here PAH was induced in SD rats by a single subcutaneous injection of MCT at a dose of 60 mg/kg.We measured contractile tension of pulmonary arteries(PAs) rings in control and MCT rats.In comparison to the controls,right ventricular systolic pressure,mean right ventricular pressure and the ratio of right ventricle(RV) to left ventricle plus septum (LV+S) weight were markedly elevated in MCT rats.Depletion of SR Ca stores with cyclopiazonic acid caused dramatic increase in PAs contraction;while application of 1-oleoyl-2-acetyl-sn-glycerol(OAG) to directly activate receptor-operated channels did not cause strikingly increase.In MCT rats,the curve was shifted to the left that Endothelin 1(ET-1)-induced concentration-dependent vasoconstriction of PAs and inhibition rate of 30 mmol/L lanthanum on ET- 1-induced plateau was significantly increased.Our results show store-operated channels of PASMCs are upregulated and contribute to the enhanced sensitization of vascular tone induced by ET-1 in MCT models.
