Senescence-Inducible Expression of Isopentenyl Transferase Extends Leaf Life,Increases Drought Stress Resistance and Alters Cytokinin Metabolism in Cassava

Cassava （Manihot esculenta Crantz） sheds its leaves during growth, especially within the tropical dry season. With the production of SAG12-1PT transgenic cassava we want to test the level of leaf retention and altered cytokinin metabolism of transgenic plants via the autoregulatory senescence inhibition system. After confirmation of transgene expression by molecular analysis and phenotype examination in greenhouse plants, two transgenic plant lines, 529-28 and 529-48, were chosen for further investigation. Detached mature leaves of 529-28 plants retained high levels of chlorophyll compared with wild-type leaves after dark-induced senescence treatment. Line 529-28 showed significant drought tolerance as indicated by stay-green capacity after drought stress treatment. Field experiments proved that leaf senescence syndrome was significantly delayed in 529-28 plants in comparison with wild-type and 529-48 plants. Physiological and agronomical characterizations of these plants also revealed that the induced expression of IPT had effects on photosynthesis, sugar allocation and nitrogen partitioning. Importantly, the 529-28 plants accumulated a high level of trans-zeatin-type cytokinins particularly of corresponding storage O-glucosides to maintain cytokinin homeostasis. Our study proves the feasibility of prolonging the leaf life of woody cassava and also sheds light on the control of cytokinin homeostasis in cassava leaves.

Characterization of a Novel Rice Metallothionein Gene Promoter:Its Tissue Specificity and Heavy Metal Responsiveness

The rice （Oryza sativa L.） metallothionein gene OsMT-I-4b has previously been identified as a type I MT gene. To elucidate the regulatory mechanism involved in its tissue specificity and abiotic induction, we isolated a 1 730 bp fragment of the OsMT-I-4b promoter region. Histochemical /^-glucuronidase （GUS） staining indicated a precise spacial and temporal expression pattern in transgenic Arabidopsis. Higher GUS activity was detected in the roots and the buds of flower stigmas, and relatively lower GUS staining in the shoots was restricted to the trichomes and hydathodes of leaves. No activity was observed in the stems and seeds. Additionally, in the root of transgenic plants, the promoter activity was highly upregulated by various environmental signals, such as abscisic acid, drought, dark, and heavy metals including Cu2＋, Zn2~, Pb2~ and Al3＋. Slight induction was observed in transgenic seedlings under salinity stress, or when treated with Co2＋ and Cd2＋. Promoter analysis of 5＇-deletions revealed that the region -583/-1 was sufficient to drive strong GUS expression in the roots but not in the shoots. Furthermore, deletion analysis indicated important promoter regions containing different metal- responsive c/s-elements that were responsible for responding to different heavy metals. Collectively, these findings provided important insight into the transcriptional regulation mechanisms of the OsMT-I- 4b promoter, and the results also gave us some implications for the potential application of this promoter in plant genetic engineering.

LjCYC Genes Constitute Floral Dorsoventral Asymmetry in Lotus japonicus

Previous study shows that LjCYC2,a CYC-like TCP （TB1,CYC and PCFs） gene in the model legume,Lotus japonicus,is involved in dorsal petal development,which together with the other two homologous genes,LjCYC1 and LjCYC3,belongs to an LjCYC gene *** this report,we modified the transformation system in ***,and constructed different RNAi transgenes to target different LjCYC *** expression of three endogenous LjCYC genes was specifically suppressed by different specific RNAi transgenes,and a chimerical RNAi transgene that contains the specific sequences from LjCYC1 and LjCYC2 was found to downregulate the expression of both endogenous genes *** of silencing three LjCYC genes were mainly restricted on either dorsal or lateral petals,demonstrating their dorsalizing and lateralizing activities during the development of zygomorphic ***,abolishing the expression of three LjCYC genes could give rise to complete loss of dorsoventral （DV） differentiation in the flower whose petals all resembled the ventral one in the wild type and displayed intact organ internal （IN） *** data demonstrate that during zygomorphic flower development,the DV asymmetry is constituted by the LjCYC genes,while the floral organ IN asymmetry is independently determined by other genetic factors.

In vivo imaging is crucial for developmental biology and can further help to follow cell development/differentiation in normal and pathological *** advances in optical imaging techniques has facilitated tracing of the developmental dynamics of a specific organ,tissue,or even a single *** zebrafish is an excellent model for imaging of hematopoiesis due to its transparent embryo at early stage;moreover,different zebrafish hematopoietic stem cells(HSCs)transgenic lines have been demonstrated as very useful tools for illustrating the details of the HSC developmental *** this review,we summarize recent studies related to the non-invasive in vivo imaging of HSC transgenics,to show that zebrafish transgenic lines are powerful tools for developmental biology and *** the end of the review,the perspective and some open questions in this field will be discussed.

Evaluation of Transgenic <i>Nicotiana tabacum</i>with <i>deh</i>E Gene Using Transposon Based IRAP Markers

In the present study, five genetically modified herbicide tolerant Nicotiana tabacum cv. TAPM24 plants with a constructed vector pCAMBIA1301a carrying dehalogenase E (dehE) gene were compared with three non-transgenic controls using Tto1 retrotransposon specific IRAP markers. dehE gene was originally characterized in Rhizobium sp. and it produced an enzyme which degraded the Dalapon herbicide. IRAP protocol was applied on transgenic and non-transgenic plants to investigate the retrotransposon based genetic variation which may appear during transformation. Polymorphism rates were calculated as 0%-20% from IRAP-PCR products among all plant samples. These results show that transformation of tobacco plant with the dehE gene may cause Tto1 retrotransposon alterations appearing as different band profiles. The findings are expected to contribute to genetic engineering studies to obtain better results and also to understand how transposons contribute to features such as transgenesis. In our knowledge, this is one of the first experimental data of transgenic N. tabacum engineered with dehE gene originated Rhizobium sp. in terms of retrotranposon based variation.

＜正＞Nkx2.2 homeodomain transcriptional factor is initially expressed in differentiating oligodendrocyte precursor cells（OPCs） and plays a critical role in regulating the differentiation *** studies showed that Nkx2.2 expression in oligodendrocytes is rapidly down-regulated as OPCs undergo terminal differentiation, but regained in mature myelinating oligodendrocytes in mature *** biphasic pattern of Nkx2.2 expression in oligodendrocyte lineage suggests that Nkx2.2 may have distinct functions during oligodendrocyte lineage progression. To define the role of Nkx2.2 in oligodendrocyte differentiation and maturation,we have used tetracycline-inducible expression system to specifically induce Nkx2.2 expression in oligodendrocyte cells at different developmental stages. Our results demonstrated that Nkx2.2 promoted OPC differentiation at embryonic stages,but repressed myelin protein production at neonatal stages,indicating that regulation of oligodendrocyte differentiation and myelin gene expression by Nkx2.2 is stage-dependent.
