Therapeutic effects of herbal formula Huangqisan on metabolic disorders via SREBF1,SCD1 and AMPK signaling pathway

Objective:Metabolic syndrome is a complex medical condition that has become an alarming epidemic,but an effective therapy for this disease is still *** use of the herbal formula Huangqisan(HQS)to treat diabetes is documented in the chinese medical literature as early as 1117 A.D.;however,its therapeutic effects and underlying mechanisms remain ***:To investigate the beneficial effects of HQS on metabolic disorders,high-fat diet-induced obesity(DIO),leptin receptor dysfunction(db/db)and low-density lipoprotein receptor-knockout(LDLR^(-/-))mice were *** mice were treated with either HQS or ***,liver tissue,white fat tissue and brown adipose tissue were harvested at the end of the *** disease-related parameters were evaluated to test effects of HQS against diabetes,obesity and *** arches from LDLR^(-/-)mice were analyzed to investigate the effects of HQS on ***-sequence,quantitative real-time polymerase chain reaction and Western blot were performed to investigate the mechanisms of HQS against metabolic ***:HQS lowered body weight,fasting blood glucose and serum lipid levels and improved glucose tolerance and insulin sensitivity in DIO mice and db/db mice(P<0.05).HQS also blocked atherosclerotic plaque formation in LDLR^(^(-/-)) *** suppressed de novo lipid synthesis by reducing the expression of messenger RNA for sterol regulatory element-binding factor 1,stearyl coenzyme A desaturase 1 and fatty acid synthase,and enhancing adenosine 5’-monophosphate-activated protein kinase signaling in both in vivo and in vitro experiments,indicating potential mechanisms for HQS’s activity against ***:HQS is effective for reversing metabolic disorder and has the potential to be used as therapy for metabolic syndrome.

At present,tradition chinese medicine(TCM)industry in China is in the stage from the empirical development to industrial *** infrared(NIR)spectroscopy has been widely used in the quality control of TCM's modernization with its characteristics incuding rapidness,nondestruction,simplicity,economy,and so *** this study,as one type of a portable micro NIR spectrometer,Micro NIR 1700 was used to establish the qualitative models for identification of geographical region and authenticity of Radix codonopsis based on discriminant analysis(DA)*** of the DA models had better predictive ability with 100%*** addition,a method for rapid quantitative analysis of polysaccharide in Radix codonopsis was also developed based on Micro NIR 1700 spectrometer with partial last-squares(PLS)*** the PLS calibration model,the *** of samples were pretreated with different preprocessing methods and the spectral region was selected with different variable selection methods as *** performance of the final PLS model was evaluated according to correlation cofficient of calibration(Rc),correlation coefficient of prediction(Rp),root mean squared error of cross validation(RMSECV),and root mean squared of prediction(RMSEP).The values of Re,Rp,RMSECV,and RMSEP were 0.9775,0.9602,2.496,and 2.734 g/mL,*** work demonstrated that micro infrared spectrometer could be more convenient and rapid for quality control of Radix codonopsis,and the presented models would be a u1seful reference for quality control of other simi lar raw materials of TCM.

本文对中医明目要药决明(Cassia tora L.)进行了核型分析。结果表明:决明的染色体数目为2n=26;13对同源染色体中,第5、6、8、10、11对,为近中部着丝点,其余几对均为中部着丝点;第5对长臂具次缢痕,染色体的绝对长度在2.14—3.48微米之间,相对长度在5.90%—9.60%之间。决明的核型公式为:K(2n)=26=16m+8sm+2sm(sc)。本实验为中药决明的药材鉴别在染色体水平上提供了更准确的细胞学依据。
