[Objectives]To analyze the chemical components of Meconopsis integrifolia(Maxim.)Franch total flavonoids(MITF).[Methods]The chemical components of MITF were identified by UPLC-Q-Exactive Orbitrap *** column was performed using ACQUITY UPLC HSS T3(100 mm×2.1 mm,1.8μm)with 0.1%formic acid water(A)-acetonitrile(B)with gradient elution at 30℃,injection volume of 2μL,and flow rate of 0.3 mL/*** ion source adopted positive and negative ion detection mode with scanning range m/z 100-1000.[Results]A total of 93 compounds were identified from MITF,including 57 flavonoids,25 phenolic acids,9 alkaloids and 2 *** them,dimethoxytaxifolin was identified as a possible new compound by SciFinder search,and 67 compounds were first identified in ***.[Conclusions]This study provides a scientific foundation for clarifying the material basis of the efficacy of *** and improving the quality standards.

Anti-inflammatory Activity and Mechanism of total flavonoids from the Phloem of Paulownia elongate S.Y. Hu in LPS-stimulated RAW264.7 Macrophages

Exposure to ambient particulate matter in fine rages(PM) is associated with the increased risk of metabolic syndrome,including non-alcoholic fatty liver disease(NAFLD).Loquat Leaf possesses several pharmacological actions,especially the effect on metabolic *** the main biological active ingredients,the potential therapeutic role of total flavonoids(TF) isolated from Loquat Leaf in PM-induced NAFLD model remains *** present study was designed to explore the pharmacological effect of TF in PM2.5-induced NAFLD mice with its related mechanisms of *** were exposed to PM2.5 for 35 days to induce NAFLD,and their body weight,the ratio of liver to body weight and the blood lipids increased significantly compared with the control *** was found that TF significantly reduced the body weight,fat deposition in the liver and blood lipids in PM2.5-induced NAFLD *** with mice under PMexposure,TF treatment alleviated oxidative stress by preventing the accumulation of oxidative product malondialdehyde(MDA) and strengthening the anti-oxidative capacity of Superoxide Dismutase(SOD).TF was also found to reduce the alanine aminotransferase(ALT) and aspartate aminotransferase(AST) activity in PM2.5-induced NAFLD *** addition,TF repaired the PM2.5-induced decline of insulin receptor substrate-1(IRs-1) and protein kinase B(Akt)***,it was found to suppress the expression of cytochrome P4502 E1(CYP2 E1) and the phosphorylation of c-jun terminal kinase(JNK) in PM-induced *** together,the current study demonstrated TF might be a potential therapeutic agent for PM-induced NAFLD via regulation of IRs-1/Akt and CYP2 E1/JNK.

total flavonoids extracted from large-leaved Kuding tea(Ilex latifolia Thunb.) were investigated to develop anti-UV sun-screening agents. The results showed that total flavonoids of I. latifolia Thunb.(TFILT) showed maximum absorb peak at 325 nm between UVB and UVA by wavelength scanning spectrum. TFILT could inhibit the decrease of cell viability induced by UVB irradiation by CCK-8 method, and 0.25 mg/ml of TFILT showed the most significant protective activity on cells. The dermal thickness and collagen fiber density as skin structural characteristic were analyzed by Histological staining. The results showed that TFILT protected mice skin by significantly attenuating the dermal thickness and inhibiting collagen fiber degradation. The mechanism in vivo needs to be further confirmed in future.

[Objectives]Optimum extraction conditions of total flavonoids from Fructus Aurantii Immaturus(TFFAI)and its resistance activity to ultraviolet radiation were investigated in present research.[Methods]The optimal extraction conditions of TFFAI were determined by single factor and orthogonal experiments,and the survival rate of TFFAI on HaCaT cells irradiated with UVB rays was *** s antioxidant capacity was determined by ABTS.[Results]The results showed that the highest yield of TFFAI was obtained with 70%ethanol at a solid-to-liquid ratio of 1:50(w/v)and 40℃for 1.5 h by single-factor and orthogonal *** flavonoids(0.25-1.00 mg/ml)could significantly improve the survival rate of HaCaT cell ***,the maximum absorption peak of TFFAI was found at 283 nm,and in-vitro antioxidant experiment identified that TFFAI had a good clearance rate to *** suggestes that TFFAI could protect the cells from UVB damage by absorption of UVB rays and anti-oxidation.[Conclusions]TFFAI played a protective role on UVB irradiated cells through UVB ultraviolet absorption and antioxidant pathways.

The aim of the present work is to assess the value of Detarium Senegalense by determining the content of total phenols, total flavonoids and total anthocyanins, and by evaluating the free radical scavenging activity of Detarium Senegalense extracts. For this purpose, sequential extraction using solvents of increasing polarity was essential. The various extracts obtained underwent phytochemical and biochemical analyses. Phytochemical screening revealed the presence of flavonoids, alkaloids, tannins, polyphenols, anthocyanins and steroids/terpenes. Quantitative analysis of total polyphenols, total flavonoids and total anthocyanins yielded the following results: total flavonoids (0.803 ± 0029 mg EQ/100g P for acetone extract of roots and 0.871 ± 0.401 mg EQ/100g P for methanol extract of leaves);total polyphenols (23.298 ± 12.68 mg EAG/100g P for acetone extract of roots and 24.69 ± 0.49 401 mg EAG/100g P for methanol extract of leaves);total monomeric anthocyanins (44.697 ± 0.939 mg EC3G/100g P and 16.699 ± 0.193 mg EC3G/100g P respectively for acetone and methanol extracts of stem bark). DPPH free radical scavenging activity was 1.674 ± 0.023 mg/mL for the acetone extract and 0.934 ± 0.24 mg/mL for the methanol extract of roots. .

In this study, bioassay-guided isolation, identification and biological evaluation of antioxidant components from total flavonoids of Chinese herbal plant Mimenghua were performed. 1, 1-Diphenyl-2-picrylhydrazyl(DPPH) radical scavenging test was adopted as the bioassay-guided method, Sephadex LH-20 column chromatography and high performance liquid chromatography were used as the purification tools for the acquisition of antioxidant components. One compound was obtained and identified as acteoside by its physicochemical properties and spectral characteristics. Antioxidant activity in vitro of acteoside was determined by evaluation of the scavenging activity on DPPH radical and hydroxyl radical, reducing power and total antioxidant *** results showed that acteoside had significant antioxidative activity and possessed higher activity with the increase of concentration, which provides the potential application of acteoside in food, pharmaceutical and cosmetic industries.

[Objective] This study aimed to optimize the ultrasonic extraction process of total flavonoids from Angelica keiskei Koidzumi. [Method] Using the yield of total flavonoids as an indicator, the ultrasonic extraction process of total flavonoids from A. keiskei was optimized by orthogonal experimental design. [ Result ] Among four factors in orthogonal experimental design, extraction time exhibited the most significant effect on the extraction result; ethanol concentration exhib- ited a significant effect on the extraction result; ultrasonic power and solid-to-liquid ratio had no significant effect. The optimal conditions for ultrasonic extraction of total flavonoids from A. keiskei were extraction time of 25 min, ethanol concentration of 80%, ultrasonic power of 60 W, solid-to-liquid ratio of 1：20 （g/ml）, under which the yield of total flavonoids reached 1.56%. [ Conclusion] This study provides the basis for further development and utilization of total flavonoids from A. keiskei.

[Objectives]To optimize the extraction process of total flavonoids of Zhuang medicine Clerodendrum japonicum(Thumb.)Sweet and establish a method for its extraction and content determination.[Methods]The total flavonoids of *** were extracted by reflux extraction *** a single factor experiment,the effects of extraction method,extraction solvent concentration,extraction volume and extraction time on the total flavonoids content of medicinal materials were investigated to select the optimal extraction process of the total flavonoids of ***.[Results]The optimal extraction process of the total flavonoids of *** was 75%ethanol,1∶20 solid-to-liquid ratio,and 50 min reflux extraction time.[Conclusions]This method can effectively determine the content of total flavonoids in *** and provide a certain scientific basis for the study of the quality standard of this Zhuang *** method has high *** is stable and feasible in extraction of total flavonoids from Zhuang medicine ***.
