(D-Ser2) oxyntomodulin recovers hippocampal synaptic structure and theta rhythm in Alzheimer’s disease transgenic mice

In our previous studies,we have shown that(D-Ser2)oxyntomodulin(Oxm),a glucagon-like peptide 1(GLP-1)receptor(GLP1R)/glucagon receptor(GCGR)dual agonist peptide,protects hippocampal neurons against Aβ1-42-induced cytotoxicity,and stabilizes the calcium homeostasis and mitochondrial membrane potential of hippocampal neurons.Additionally,we have demonstrated that(D-Ser2)Oxm improves cognitive decline and reduces the deposition of amyloid-beta in Alzheimer’s disease model mice.However,the protective mechanism remains unclear.In this study,we showed that 2 weeks of intraperitoneal administration of(D-Ser2)Oxm ameliorated the working memory and fear memory impairments of 9-month-old 3×Tg Alzheimer’s disease model mice.In addition,electrophysiological data recorded by a wireless multichannel neural recording system implanted in the hippocampal CA1 region showed that(D-Ser2)Oxm increased the power of the theta rhythm.In addition,(D-Ser2)Oxm treatment greatly increased the expression level of synaptic-associated proteins SYP and PSD-95 and increased the number of dendritic spines in 3×Tg Alzheimer’s disease model mice.These findings suggest that(D-Ser2)Oxm improves the cognitive function of Alzheimer’s disease transgenic mice by recovering hippocampal synaptic function and theta rhythm.

Modulation of Neuronal Activity and Saccades at theta rhythm During Visual Search in Non-human Primates

Active exploratory behaviors have often been associated with theta oscillations in rodents,while theta oscillations during active exploration in non-human primates are still not well understood.We recorded neural activities in the frontal eye field(FEF)and V4 simultaneously when monkeys performed a free-gaze visual search task.Saccades were strongly phase-locked to theta oscillations of V4 and FEF local field potentials,and the phase-locking was dependent on saccade direction.The spiking probability of V4 and FEF units was significantly modulated by the theta phase in addition to the time-locked modulation associated with the evoked response.V4 and FEF units showed significantly stronger responses following saccades initiated at their preferred phases.Granger causality and ridge regression analysis showed modulatory effects of theta oscillations on saccade timing.Together,our study suggests phase-locking of saccades to the theta modulation of neural activity in visual and oculomotor cortical areas,in addition to the theta phase locking caused by saccade-triggered responses.

Chronic intermittent hypoxia（IH）occurs in obstructive sleep apnea（OSA）,a very common sleep and breathing disorders that is associated with central nervous system dysfunction,including learning and memory impairment.Despite the obvious importance,the question of exactly what happens to neuronal activities in different brain regions during intermittent hypoxia has never been addressed.We envision that long-term recording of the firing activities of neurons and brain rhythms in vivoduring and after the IH will provide direct information to address this question and thus considerable insight into the cause of cognitive dysfunctions in OSA.Based on a well-established OSA model in which the rats were subject to daily 8-hr cycling of oxygen between 21%and 10%every 90s,the firing activities of neurons in the CA1region of the hippocampus were followed for 14 days by chronically implanted multi-electrode arrays.We found that both CA1 principal neurons and interneurons tended to increase their firing activities during the first week of IH treatment,particularly apparent at the late hours（at 8 h）but not early hours（at 5 min and3 h）of daily IH paradigm.The hyper-excitability,however,was followed by gradual suppression of firing in the second week.At Day 14,the firing rates of principal neurons and interneurons were typically less than60%and 25%,respectively of those at Day 1.These profiles paralleled the increase in long-term potentiation magnitude at Day 2 and its suppression in the second week of IH treatment measured from acutely prepared hippocampal slices.Partial recovery of the neuronal activities was found after 1-week recovery in normoxia but confined to principal neurons only.Interestingly,analysis of the field potential revealed that the hippocampal theta rhythm（4-8 Hz）was diminished by chronic IH with no sign of recovery 1 week after the IH treatment.Together,these data suggest that hippocampal neurons respond to short-term hypoxia treatment by boosting their activities.However,long term IH impairs the firing activ
