Since the existing prediction methods have encountered difficulties in processing themultiple influencing factors in short-term power load forecasting,we propose a bidirectional long short-term memory(BiLSTM)neural network model based on the temporal pattern attention(tpa)mechanism.Firstly,based on the grey relational analysis,datasets similar to forecast day are obtained.Secondly,thebidirectional LSTM layermodels the data of thehistorical load,temperature,humidity,and date-type and extracts complex relationships between data from the hidden row vectors obtained by the BiLSTM network,so that the influencing factors(with different characteristics)can select relevant information from different time steps to reduce the prediction error of the model.Simultaneously,the complex and nonlinear dependencies between time steps and sequences are extracted by the tpa mechanism,so the attention weight vector is constructed for the hidden layer output of BiLSTM and the relevant variables at different time steps are weighted to influence the input.Finally,the chaotic sparrow search algorithm(CSSA)is used to optimize the hyperparameter selection of the model.The short-term power load forecasting on different data sets shows that the average absolute errors of short-termpower load forecasting based on our method are 0.876 and 4.238,respectively,which is lower than other forecastingmethods,demonstrating the accuracy and stability of our model.

Winter jujube(Ziziphus jujuba‘Dongzao’)is an excellent late maturing variety of fresh-eating jujube in China.Fruit texture is an important indicator of sensory quality.To investigate the correlations among texture indices and establish an evaluation system for winter jujube texture,we used the TMS-Touch instrument to perform a texture profile analysis(tpa)on 1150 winter jujubes from three major producing areas in China.Eight indices and their best-fit distribution were obtained,including fracture(Pearson),hardness(InvGauss),adhesive force(Weibull),adhesiveness(LogLogistic),cohesiveness(LogLogistic),springiness(BetaGeneral),gumminess(InvGauss),and chewiness(InvGauss).Based on the best-fit distribution curves,each index was divided into five grades(lower,low,medium,high and higher)by the 10th,30th,70th and 90th percentiles.Among the texture indices,82%of the correlation coefficients were highly significant(P<0.01);meanwhile,chewiness was significantly(P<0.01)and positively correlated with springiness and gumminess,of which the correlation coefficients were up to 0.8692 and 0.8096,respectively.However,adhesiveness was significantly(P<0.01)and negatively related to adhesive force with a correlation coefficient of–0.7569.Among hardness,cohesiveness,springiness,gumminess,and chewiness,each index could be well fitted by a multiple linear regression with the remaining four indices,with the coefficients above 0.94 and the mean fitting error and mean prediction error lower than 10%.A comprehensive evaluation model was consequently established based on factor analysis to evaluate the texture quality of winter jujube.The results demonstrated that winter jujube with higher comprehensive scores generally exhibited higher springiness and chewiness,but had lower adhesive force and adhesiveness.We used factor analysis and clustering analysis to divide the eight studied texture into four groups(cohesive factor,adhesive-soft factor,tough-hard factor,and crispness factor),whose representative indices were springiness,adhesiveness,ha

The splicing of many alternative exons in the precursor messenger RNA (pre-mRNA) is regulated by extracellular factors but the underlying molecular bases remain unclear. Here we report the differential regulation of Bcl-x pre-mRNA splicing by extracellular factors and their distinct requirements for pre-mRNA elements. In K562 leukemia cells, treatment with interleukin-6 (IL-6) or granulocyte-macrophage colony stimulating factor (GM-CSF) reduced the proportion of the Bcl-xL variant mRNA while treatment with 12-O-tetradecanoylphorbol 13-acetate (tpa) had no effect. In U251 glioma cells, however, tpa efficiently increased the Bcl-xL level. These regulations were also seen for a transfected splicing reporter mini-gene. Further analyses of deletion mutants indicate that nucleotides 1-176 of the downstream intron are required for the IL-6 effect, whereas additional nucleotides 177-284 are essential for the GM-CSF effect. As for the tpa effect, only nucleotides 1-76 are required in the downstream intron. Thus,

In the present study, gonadotropin and gonadotropin-releasing hormone (GnRH) regulation of tpa and PAI-1 expression in PMSG-primed granulosa cells has been investigated, (i) Addition of go-nadotropins (FSH and LH) and GnRH agonist (GnRHa) or PMA to the culture increases tpa activity) FSH (or LH) plus GnRHa (or PMA) in the culture further enhances the enzyme production to such an extent that a more obvious effect than the additive effect caused by these hormones used alone has been observed; (ii) in contrast, FSH and LH decrease PAI-1 activity, whereas GnRHa and PMA alone markedly increase PAI-1 mRNA level and PAI-1 activity. Because FSH and LH stimulate tpa production and have no significant effect on PAI-1 mRNA induction, the observed inhibition of PAI-1 activity by gonadotropins may be due to the occurrence of neutralization of PA and PAI-1 proteins in the conditioned media by the formation of complexes between PA and PAI-1 ; (iii) increases in PAI-1 mRNA level and activity by GnRH and PMA are

In recent years, one of the most exciting advances in the researches of pituitary adenomas is the discovery that 30 %-40 % of human pituitary somatotrophinomas carry somatic mutations of the gene for the α subunit of the stimulatory GTP binding protein, G s (G sα). These mutations, termed gsp oncogenes, may play an important role in the tumorigenesis of pituitary adenomas. Of 10 somatotrophinomas examined, 3 (30 %) were proved to be gsp positive, as determined by sequence analysis of DNA generated by the polymerase chain reaction (PCR). GHRH exerted a significant stimulatory effect on GH secretion in 2 of 3 gsp positive and 4 of 7 gsp negative tumors. Moreover, phorbol ester, 1, 2 tetradecanoylphorbol 13 acetate (tpa), enhanced stimulation of lated the GH secretion effect exerted by GHRH in gsp positive somatotrophinomas, whereas this effect was not observed in gsp negative tumors. This result suggests that the protein kinase C signal system as well as adenylyl cyclase cAMP protein kinase A intracellular signal transduction system plays a pivotal role in GH secretory control of GHRH, which may work together via a cross talk mechanism.

We have demonstrated for the first time that (ⅰ) mouse Sertoli cells predominantly secrete tpa under the action of FSH and cAMP-generating agents, whereas Leydig cells mainly produce uPA; (ⅱ) Sertoli cells are also capable of secreting PAI-1, as well as FSH, growth factors and GnRH increase PAI-1 gene expression; (ⅲ) the increases in tpa and PAI-1 activities by different hormones in the conditioned media of Sertoli cells correspond to the increases in the levels of tpa and PAI-1 mRNA in the cultured cells, suggesting that the synthesis of the activator-inhibitor in mouse Sertoli cells is regulated at a transcriptional level and the tpa secreted by Sertoli cells may be involved in the spermatogenesis.

In this study we have demonstrated that both granulosa and theca-interstitial cells of hy-pophysectomized rat ovaries are capable of synthesizing tpa and PAI-1.Injection of a GnRH agonist canmarkedly induce these gene expressions in the ovary in a cell-specific and time-coordinated manner,so that asurge of tpa mRNA and its activity in both granulosa and theca-interstitial cells was obtained just prior toovulation.Theca-interstitial cells make PAI-1 become the most active in the ovary.Both the amount PAI-1mRNA and its activity in the cells reach the maximum level 6 h before the tpa peak.By contrast,granulosacells produce only a little amount of PAI-1 (most increase tpa activity),and both PAI-1 mRNA and activityin the cells reach the maximum after ovulation.The coordinated regulation of tpa and PAI-1 in the ovarymay fine-tune the peak of tpa activity which may be important for the regulation of the ovulatory process.The changes of tpa and PAI-1 in the ovarian cells of hypophysectomized rats during GnRHa-induced ovula-tion are similar to that in intact rats during hCG-induced ovulation,suggesting that the ovulatory process canbe modulated by different regulatory signals mediated by influencing the coordinated expression of both tpaand PAI-1.
