SERS-based lateral flow immunoassay of troponin I by using gapenhanced Raman tags

The lateral flow immuno assay (LFIA) has emerged as a powerful tool for rapid scree ning owing to its simplicity and flexibility for detection of various biomarkers. However, conventional LFIA strips have several disadvantages, including limits in quantitative analysis and low sensitivity. Here we developed a novel surface-e nhanced Rama n scatteri ng LFIA based on non spherical gap-e nhanced Raman tags (GERTs), with Raman molecules (RMs) embedded in a 1-nm gap between Au nanorod core and Au shell. Such tags have a strong and uniform surface-enhanced Rama n scattering (SERS) resp on se, an order of mag nitude higher than that of other comm on SERS tags such as Au nano rods, nano stars, Au nano shells with surface-adsorbed RMs, or spherical GERTs with embedded RMs. The feasibility of the tags was dem on strated by the semiqua ntitative and sen sitive detecti on of the heart disease biomarker cardiac tropo nin I (cTnI). GERTs were conjugated with mono clonal antibodies and used for LFIA in the same way as ordinary functionalized colloidal gold. The presenee of the target antigen, cTnI, was identified by Raman microscopy mapping of the test zone. With the SERS-based LFIA, the limit of cTnI detection was about 0.1 ng/mL. This value is within the diagnostic range of cTnI in the blood serum of patients with heart infarction and is 30 times lower than that of the colorimetric LFIA test using the same antibodies and either GERTs or colloidal gold as labels.

Identification of protein targets for the antidepressant effects of Kai-Xin-San in Chinese medicine using isobaric tags for relative and absolute quantitation

Kai-Xin-San consists of Ginseng Radix, Polygalae Radix, Acori Tatarinowii Rhizoma, and Poria at a ratio of 3:3:2:2. Kai-Xin-San has been widely used for the treatment of emotional disorders in China. However, no studies have identified the key proteins implicated in response to Kai-Xin-San treatment. In this study, rat models of chronic mild stress were established using different stress methods over 28 days. After 14 days of stress stimulation, rats received daily intragastric administrations of 600 mg/kg Kai-Xin-San. The sucrose preference test was used to determine depression-like behavior in rats, while isobaric tags were used for relative and absolute quantitation-based proteomics to identify altered proteins following Kai-Xin-San treatment. Kai-Xin-San treatment for 2 weeks noticeably improved depression-like behaviors in rats with chronic mild stress. We identified 33 differentially expressed proteins: 7 were upregulated and 26 were downregulated. Functional analysis showed that these differentially expressed proteins participate in synaptic plasticity, neurodevelopment, and neurogenesis. Our results indicate that Kai-Xin-San has an important role in regulating the key node proteins in the synaptic signaling network, and are helpful to better understand the mechanism of the antidepressive effects of Kai-Xin-San and to provide objective theoretical support for its clinical application. The study was approved by the Ethics Committee for Animal Research from the Chinese PLA General Hospital(approval No. X5-2016-07) on March 5, 2016.

Sensory and motor nerve fibers of peripheral nerves have different anatomies and regeneration functions after injury. To gain a clear understanding of the biological processes behind these differences, we used a labeling technique termed isobaric tags for relative and absolute quantitation to investigate the protein profiles of spinal nerve tissues from Sprague-Dawley rats. In response to Wallerian degeneration, a total of 626 proteins were screened in sensory nerves, of which 368 were upregulated and 258 were downregulated. In addition, 637 proteins were screened in motor nerves, of which 372 were upregulated and 265 were downregulated. All identified proteins were analyzed using the Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis of bioinformatics, and the presence of several key proteins closely related to Wallerian degeneration were tested and verified using quantitative real-time polymerase chain reaction analyses. The differentially expressed proteins only identified in the sensory nerves were mainly relevant to various biological processes that included cell-cell adhesion, carbohydrate metabolic processes and cell adhesion, whereas differentially expressed proteins only identified in the motor nerves were mainly relevant to biological processes associated with the glycolytic process, cell redox homeostasis, and protein folding. In the aspect of the cellular component, the differentially expressed proteins in the sensory and motor nerves were commonly related to extracellular exosomes, the myelin sheath, and focal adhesion. According to the Kyoto Encyclopedia of Genes and Genomes, the differentially expressed proteins identified are primarily related to various types of metabolic pathways. A Genetic Algorithm-Based Smart Antenna Technique for Anti-Collision of Multiple SAW ID-tags

In the radio frequency identification (RFID) system based on surface acoustic wave (SAW) technique, some tags often locate in the field of a transceiver at the same time. These tags produce simultaneous echo signals which "collide" when they arrive back at the transceiver, which leads to difficult identification. In this paper, smart antenna technique is presented to implement anti-collision in SAW RFID system. The direction of arrivals (DOAs) are used to denote the locations of tags, and genetic algorithm (GA) is suggested to find the optimal estimates of the DOAs in complex multimodal search spaces. Once the DOAs are obtained, the array weights are formed and the signals of tags are recovered to implement decoding. The experimental results show that the GA-based smart antenna technique works well in some occasions.

Research on Social tags Recommendation Techniques Based on Content

With the rapid development of the Internet,more and more people begin to pay attention to social tagging which is a flexible and efficient method for classification. How to retrieve tags from the huge tag library becomes a hot topic to research. Firstly,the existing systems of social tags and its recommendation principles used in Web 2. 0 are introduced in this paper. Secondly,the existing techniques about tag recommendation are summarized,and their merits and demerits are analysed. In most techniques for tag recommendation only two dimensions "resource-user " are considered. But there are three dimensions "resource-user-tag " in recommendation system based on social tags. A new method of social tag recommendation based on content-Feature Vote Tagging ( FVT) is proposed in this paper. Finally,several kinds of evaluation methods are used to assess the return results of methods. The experiment results show that the method proposed in this paper can satisfy the expectation of the user for the recommendation results.

Generation and analysis of expressed sequence tags from the medicinal plant Salvia miltiorrhiza

Milk thistle(Silybum marianum)is a crucial medicinal plant containing a large amount of *** the study,the changes in storage oil during seed germination and seedling transition from heterotrophic phases were *** results showed that seed oil decreased from 19.53%to 0.88%on the 7th day of seedling *** hydrolysis continued until the 4th day of germination with a low slope,but then increased the use of oils in seed germination end seedling growth *** results indicated that the quantitative changes in fatty acids,presented at lower amount,were relatively higher than dominant fatty *** were decreasing phenolic content in the developing seedlings,but overall,lowest level of total phenolic content can be attributed to the control(30.52 mg⋅100 g⋅Oil^(-1)).In contrast,the maximum peroxide value(2.58 meq⋅kg Oil^(-1))in the developing seedling was observed on the last day of the *** results showed that there was a significant correlation between saturated fatty acid,unsaturated fatty acid,and lipase ***,the correlation between lipase activity and polyunsaturated fatty acids was significantly higher than between lipase activity and monounsaturated fatty acids(R^(2)=90%and R^(2)=77%,respectively).Therefore,the lipolysis process acts selectively in milk thistle *** to the results,C12:0 exhibits a greater impact on the early seedling growth rather than on the germination process and is one of the determining factors in the transition from heterotroph to ***,it can be a marker for tags breakdown.

Recharging RFID tags for Environmental Monitoring Using UAVs: A Feasibility Analysis

RFID tags are used for different purposes. One of the major problems to be addressed, particularly for monitoring purposes, is their limited power autonomy. tags must perform different tasks with limited power consumption and their batteries capacities are often too low, even if low power consumption techniques are implemented. In these operational situations tags should be kept in operation for long periods of time and the common solution is to go directly where they are installed and recharge them manually or change their batteries;alternatively, when possible, small photovoltaic (PV) panels may be adopted. This paper proposes a feasibility analysis of how it is possible to recharge a multipurpose RFID tag using a UAV (Unmanned Aerial Vehicle), which is programmed to go above the tags and recharge them. This possibility is analyzed from an energetic point of view assuming to recharge a Wireless Sensor Network (WSN) using a common commercial UAV adequately instrumented using the wireless power transfer technique.

Analysis of Simple Sequence Repeats Information from Floral Expressed Sequence tags Resources of Papaya (<i>Carica papaya</i>L.)

Papaya (Carica papaya L.) is one of the most economically, medicinally and nutritionally important tropical fruit crops. Expressed sequence tags (ESTs) derived simple sequence repeat (SSR) markers are more valuable as they are derived from conserved genic portion. Development of EST-SSRs markers through in silico approach is cheaper, less time consuming and labour-intensive. In this study, we aimed to mine SSRs and developed EST-SSR primers from papaya floral ESTs. A total of 75,846 papaya floral ESTs were downloaded from public database National Centre for Biotechnology Information (NCBI). A total of 26,039 floral unigenes (7961 contigs and 18,078 singletons) were generated after assembly of these ESTs. From these floral unigenes, 433,782 perfect SSRs, 204,968 compound SSRs and 6061 imperfect SSRs were mined, respectively. In perfect SSRs, mononucleotide repeats were most abundant (94.7%) followed by tri- (3.1%) and di-nucleotide repeats (1.7%). The frequencies of tetra-, hexa- and penta-nucleotide repeats accounted for only (0.17%), (0.04%) and (0.03%), respectively. In mononucleotide repeats, the most abundant motif was A/T (69.3%) and in di- and tri-nucleotide repeats were AG/CT (61%) and AAG/CTT (31%), respectively. In imperfect SSRs, mononucleotide repeats (56.5%) were most abundant. 176 different types of motifs were identified. A total of 3807 primer pairs for floral papaya ESTs were successfully designed. These developed EST-SSR primers are being used for the genetic improvement of papaya such as study of cross-transferability across genera/species, evaluation of genetic diversity, and identification of sex-specific markers. These EST derived SSRs can also be used in filling gaps in existing linkage maps in papaya.

A Comparison of Languages on the Chinese and English Clothing tags from the Cross-cultural Perspective

With the rapid growth of China's marketing economy, a lot of China-made clothes sweep into the foreign markets in recent years, so the internationalization, standardization and normalization of the languages of the clothing tags seem more important than they probably are. From the cross-cultural perspective and with the help of stylistic analysis, the author studies the languages of 10 Chinese-version clothing tags and 10 original English-version clothing tags respectively in lexis, syntax and discourse. The English version of Chinese clothing tags inclines to use the imperative sentences with strong tone of command, and their corresponding sentence structures and those of the original English clothing tags are not alike. Comparatively the latter give a more static feeling in a milder tone. In the perspective of lexis and discourse, clothing tags, both home and abroad, use the simple words and add the words with pictures. Tentatively, the author gets a general conclusion of the skills and characteristics of English clothing tags' language. Besides, according to the current problems existing in the Chinese clothing tags' translation,the author gives the relative advice to the Chinese clothing company in order to promote the international commercial communication, besides, the author expects more studies on this field with a new view point.
