Potentiation of the lateral habenula-ventral tegmental area pathway underlines the Susceptibility to depression in mice with chronic pain

Chronic pain often develops severe mood changes such as depression.However,how chronic pain leads to depression remains elusive and the mechanisms determining individuals’responses to depression are largely unexplored.Here we found that depression-like behaviors could only be observed in 67.9%of mice with chronic neuropathic pain,leaving 32.1%of mice with depression resilience.We determined that the spike discharges of the ventral tegmental area(VTA)-projecting lateral habenula(LHb)glutamatergic(Glu)neurons were sequentially increased in sham,resilient and susceptible mice,which consequently inhibited VTA dopaminergic(DA)neurons through a LHbGlu-VTAGABA-VTADA circuit.Furthermore,the LHbGlu-VTADA excitatory inputs were dampened via GABAB receptors in a pre-synaptic manner.Regulation of LHb-VTA pathway largely affected the development of depressive symptoms caused by chronic pain.Our study thus identifies a pivotal role of the LHb-VTA pathway in coupling chronic pain with depression and highlights the activity-dependent contribution of LHbGlu-to-VTADA inhibition in depressive behavioral regulation.

Susceptibility of patients with chronic obstructive pulmonary disease(COPD)to cardiovascular autonomic dysfunction asso-ciated with exposure to metals in ambient fine particles(PM2.5,particulate matter with aerodynamic diameter≤2.5µm)remains poorly evidenced.Based on the COPDB(COPD in Beijing)panel study,we aimed to compare the associations of heart rate(HR,an indicator of cardiovascular autonomic function)and exposure to metals in PM2.5 between 53 patients with COPD and 82 healthy controls by using linear mixed-effects models.In all participants,the HR levels were significantly associated with interquartile range increases in the average concentrations of Cr,Zn,and Pb,but the strength of the associations differed by exposure time(from 1.4%for an average 9 days(d)Cr exposure to 3.5%for an average 9 d Zn exposure).HR was positively associated with the average concentrations of PM2.5 and certain metals only in patients with COPD.Associations between HR and exposure to PM2.5,K,Cr,Mn,Ni,Cu,Zn,As,and Se in patients with COPD significantly differed from those in health controls.Furthermore,association between HR and Cr exposure was robust in COPD patients.In conclusion,our findings indicate that COPD could exacerbate difference in HR following exposure to metals in PM2.5.

To the Editor:Adipose tissue occurs in at least two different entities in mammals and humans:brown adipose tissue(BAT)and white adipose tissue(WAT).BAT is characterized by a unique uncoupling protein 1(UCP1)in the mitochondria that enables the uncoupling of the respiratory chain from adenosine triphosphate synthesis.Thus,energy is dissipated as heat to reduce fat accumulation.BAT is also considered a highly heterogeneous tissue with abundant oxygen,blood supply,and iron-rich mitochondria.[1,2]Activation of BAT via exposure to a cold environment is considered to be a means of reducing triglycerides to fight obesity.[3]The alterations in cells and tissues of activated BAT include increased iron content and UCP1 expression in mitochondrial,blood perfusion,and lipid utilization.[4]Therefore,accurate identification and quantitative analysis of inactive and activated BAT are of great significance for the treatment of metabolic diseases that target BAT,such as obesity.

New measurement method of displacement Susceptibility based on time‐domain current and potential combined spectra

Displacement Susceptibility is a very important parameter to characterise the displacement polarisation of dielectrics.Two methods based on the time‐domain current and potential combined spectra are proposed to measure the displacement Susceptibility.One is based on the time‐domain polarisation current and decay potential combined spectra,and the other is based on the time‐domain depolarisation current and recovery potential com-bined spectra.The principle of the proposed methods are verified by simulation.In addition,a measurement system for the two proposed methods is built and is used to measure the displacement Susceptibility of transparent organic glass and opaque nonlinear field grading materials.The experimental results are compared with those of the present optical method and the frequency‐domain method.Simulation and experimental results show that the proposed methods for the measurement of displacement Susceptibility are feasible and practical,which lays a foundation for the research on the polarisation characterisation of dielectrics.

Objective:Gliomas are the most common tumors in the central nervous system.The cancer Susceptibility candidate 15(CASC15)gene has been reported to be a Susceptibility gene for several types of cancer.No studies have been carried out on the predisposing effect of CASC15 gene single nucleotide polymorphisms(SNPs)on glioma risk.Methods:In order to determine whether CASC15 gene SNPs are involved in glioma Susceptibility,the first association study in a relatively large sample,which consisted of 171 patients and 228 healthy controls recruited from China,was performed.The contribution of SNPs(rs6939340 A>G,rs4712653 T>C and rs9295536 C>A)to the risk of glioma was evaluated by multinomial logistic regression,based on the calculation of the odds ratio(OR)and 95%confidence interval(CI).Results:In the single locus and combined analysis,it was revealed that the genetic risk score had no significant associations between CASC15 gene SNPs and glioma risk.However,in the stratified analysis,a significant decrease in risk of glioma was observed in subjects of<60 months old with the rs4712653 TT genotype,when compared to those with the CC/CT genotype(OR=0.12,95%CI=0.02-0.91,P=0.041).Conclusion:The present study provides referential evidence on the association between the genetic predisposition of the CASC15 gene and glioma risk in Chinese children.However,more well-designed case-control studies and functional experiments are needed to further explore the role of CASC15 gene SNPs.

Wheat production is often affected by different types of rust diseases,in which stripe rust is triggered by a fungal pathogen,Puccinia striiformis f.***.tritici(Pst),and remains the biggest challenge to plant breeders and the farming community.Nevertheless,pathogens exploit its effector proteins for altering the Susceptibility(S)genes in the host plant,rendering host defense inactivated.Thus,altering the S genes by employing precise genome editing tools provides a great opportunity to develop broad-spectrum and durable,resistant crops.Recently,Wang et al.,2022 have demonstrated the TaPsIPK1 as an ideal S gene target to confer stripe rust resistance in wheat,providing an opportunity to deliver broad-spectrum,rust-resistant crop varieties.

An hourly shallow landslide warning model developed by combining automatic landslide spatial Susceptibility and temporal rainfall threshold predictions

Landslide warning models are important for mitigating landslide risks.The rainfall threshold model is the most widely used early warning model for predicting rainfall-triggered landslides.Recently,the rainfall threshold model has been coupled with the landslide Susceptibility(LS)model to improve the accuracy of early warnings in the spatial domain.Existing coupled models,designed based on a matrix including predefined rainfall thresholds and Susceptibility levels,have been used to determine the warning level.These predefined classifications inevitably have subjective rainfall thresholds and Susceptibility levels,thus affecting the probability distribution information and eventually influencing the reliability of the produced early warning.In this paper,we propose a novel landslide warning model in which the temporal and spatial probabilities of landslides are coupled without predefining the classified levels.The temporal probability of landslides is obtained from the probability distribution of rainfall intensities that triggered historical landslides.The spatial probability of landslides is then obtained from the Susceptibility probability distribution.A case study shows that the proposed probability-coupled model can successfully provide hourly warning results before the occurrence of a landslide.Although all three models successfully predicted the landslide,the probability-coupled model produced a warning zone comprising the fewest grid cells.Quantitatively,the probabilitycoupled model produced only 39 grid cells in the warning zone,while the rainfall threshold model and the matrix-coupled model produced warning zones including 81 and 49 grid cells,respectively.The proposed model is also applicable to other regions affected by rainfall-induced landslides and is thus expected to be useful for practical landslide risk management.

Background Depression,one of the most frequent complications after stroke,increases the disease’s burden and physical disability.Poststroke depression(PSD)is a multifactorial disease with genetic,environmental and biological factors involved in its occurrence.Genetic studies on PSD to date have mainly focused on the monoamine system and brain-derived neurotrophic factors.However,understanding is still limited about the influence of the single nucleotide polymorphism(SNP)of other neurotrophic factors on PSD.Aims The present study aimed to investigate the relationship between seven vascular endothelial growth factor(VEGF)family gene variants that occur with PSD.Methods A multicentre candidate gene study from five hospitals in Jiangsu Province from June 2013 to December 2014 involved 121 patients with PSD and 131 patients with non-PSD.Demographic characteristics and neuropsychological assessments were collected.Theχ^(2)test was used to evaluate categorical variables,while the independent t-test was applied to continuous variables.SNPs in seven genes(VEGFA,VEGFB,KDR,FLT-1,IGF-1,IGF-1R and PlGF)were genotyped.Single-marker association for PSD was analysed byχ^(2)tests and logistic regression using SPSS and PLINK software.Results Patients with PSD included more women and those with lower education levels,lower body mass indexes,lower Mini-Mental State Examination scores,and higher scores on the 17-item Hamilton Depression Rating Scale than non-PSD patients.Ninety-two SNPs with seven genes were genotyped and passed quality control.The rs7692791 CC genotypes,the C allele of KDR and the rs9282715 T allele of IGF-1R increased the risk for PSD(χ^(2)=7.881,p=0.019;χ^(2)=4.259,p=0.039;χ^(2)=4.222,p=0.040,respectively).In addition,the SNP rs7692791 of KDR was significantly associated with PSD by the logistic regression of an additive model(p=0.015,OR=9.584,95%CI:1.549 to 59.31).Conclusions Patients with rs7692791 C allele carriers or the CC genotype of KDR and the rs9282715 T allele of IGF-1R may have PSD Susceptibility.Findin

Lupus nephritis(LN) has a high incidence in systemic lupus erythematosus(SLE) patients, but there is a lack of sensitive predictive markers. The purpose of the study was to investigate the association between the CD4^(+)CD8^(+)double positive T(DPT) lymphocytes and LN. The study included patients with SLE without renal impairment(SLE-NRI), LN, nephritic syndrome(NS), or nephritis. Peripheral blood lymphocyte subsets were analyzed by flow cytometry. Biochemical measurements were performed with peripheral blood in accordance with the recommendations proposed by the National Center for Clinical Laboratories. The proportions of DPT cells in the LN group were significantly higher than that in the SLE-NRI group(t=4.012, PSusceptibility, and the interference of NS and nephritis could be effectively excluded when assessing the risk of renal impairment during SLE with DPT cell proportion.

Proteomic Profiling and Protein-Protein Interaction Network Reveal the Molecular Mechanisms of Susceptibility to Drought Stress in Canola(Brassica napus L.)

Drought stress is one of the most important abiotic stresses that plants face frequently in nature.Under drought conditions,many morphological,physiological,and molecular aspects of plants are changed and as a result plants experience a remarkable reduction in growth,yield,and reproduction.To expand our understanding of the molecular basis of the plant response to drought stress,the proteomic profile and protein-protein network of canola(Brassica napus L.)were studied.The focus was to show molecular mechanisms related to canola Susceptibility to drought stress.The experiment used a completely randomized design,implemented in a hydroponic system under greenhouse conditions.To impose drought stress,plants were exposed to Hoagland’s solution supplemented with polyethylene glycol(PEG)6000 for 7 days.The drought stress resulted in 161reproducible protein spots in twodimensional electrophoresis of canola leaves.The t-student test showed 21 differentially abundant proteins(DAP),of which 2 and 19 were up and down accumulated,respectively.Two spots identified as 1-aminocyclopropane-1-carboxylate oxidase and D-2-hydroxyglutarate dehydrogenase showed an increased abundance of 2.11 and 1.77,respectively.The extended protein-protein interaction of differentially abundant proteins and KEGG analysis showed 47 pathways directly and indirectly associated with canola response to drought stress.DAPs with increased abundance were associated with amino acid and signaling processes,whereas DAPs with decreased abundance were mostly connected with pathways responsible for energy production.The results of the study will help to elucidate further the molecular events associated with the Susceptibility to drought stress in canola.
