Microbial population and enzyme activities are the significant indicators of soil *** microbial dynamics characterize microbial population and enzyme *** present study explores the development of efficient predictive modeling systems for the estimation of specific soil microbial dynamics,like rock phosphate solubilization,bacterial population,and ACC-deaminase *** specifically,optimized subtractive clustering(SC)and Wang and Mendel's(WM)fuzzy inference systems(FIS)have been implemented with the objective to achieve the best estimation accuracy of microbial *** measurements were performed using controlled pot experiment using minimal salt media with rock phosphate as sole carbon source inoculated with phosphate solubilizing microorganism in order to estimate rock phosphate solubilization potential of selected *** experimental parameters,including temperature,pH,and incubation period have been used as inputs SC-FIS and *** better performance of the SC-FIS has been observed as compared to the WM-FIS in the estimation of phosphate solubilization and bacterial population with the maximum value of the coefficient of determination(0.9988)2 R=in the estimation of previous microbial dynamics.

To isolate the over-expressed genes in human renal cell carcinoma (RCC) and analyze its molecular basis of carcinogenesis, we used the mRNA from human RCC tissues as tester and that from the matched normal kidney tissues as driver to construct the suppression subtractive hybridization library. 379 of the subtracted clones were arrayed onto a nylon mem-brane and the over-expressed genes were then screened by hybridizing the filter with radioac-tively labeled cDNA from RCC and matched normal kidney tissues. 67 clones over-expressed in RCC by a factor of 6 or more were sequenced and its identities were analyzed in GenBank da-tabase. 4 clones were previously unknown fragments and 2 clones represent KIAA genes. The rest clones were the known genes and some of them were RCC-related, including vascular en-dothelial growth factor, vimentin and tissue factor. Most of the known genes were the RCC-related genes previously unknown, including zinc ribbon domain-containing 1 protein (ZNRD1), pituitary tumor transforming gene1 (PTTG1). Northern blot and semi-quantitative RT-PCR confirmed that the mRNA levels of the 3 novel fragments and 1 KIAA and 3 known genes were significantly higher in RCC than in the matched normal kidney tissues. Immunohis-tochemical and Western blot analysis for PTTG1 and ZNRD1 revealed increased protein level in RCC. The over-expressed genes in RCC are the potential molecular targets for diagnosis and therapy and it is very important to understand the molecular mechanism of RCC through the profile of over-expressed genes.

Using suppression subtractive hybridization, a renal cell carcinoma (RCC) cDNA subtractive library which only contains differently expressed cDNAs between human RCC and normal kidney has been constructed. 200 clones were picked out randomly to perform enzyme digest analysis, a part of them underwent sequence analysis and Northern blot to identify RCC specially expressed genes. Results showed that 190 clones contain 50-400 bp inserts respectively. Sequence analysis was performed in 10 clones. All the 10 sequences were unknown before and derived from 6 unique novel genes among which the cDNA insert RCC18 has five copies. Northern blot analysis showed that RCC18 cDNA expressed highly in RCC, but there was no signal detected in normal kidney, and the full length of RCC18 was about 2.5 kb. The constructed cDNA subtractive library of human RCC is a highly efficient one and lays the solid foundation for large-scale screening and cloning new and specific oncogenes or tumor suppressor genes of RCC. A subtracted cDNA library of rice (Oryza sativa L.) inflorescence meristem (IM) was constructed using the suppression subtractive hybridization (SSH) method. The cDNAs of the rice shoot apical meristem (SAM) were used as 'driver' and inflorescence meristem (IM) as 'tester' in the experiment, respectively. Forty of 250 randomly chosen cDNA clones were identified by differential screening, which were IM-specific or IM-highly expressed. Most of the rice IM cDNAs cloned by SSH appear to represent rare transcripts, 40% of which were derived from truly differentially expressed genes. Of all the forty sequenced cDNA inserts, eleven contain the regions with 60%-90% identity to their homolog in GenBank, eighteen are expected to be new genes, only two correspond to published rice genes.

'Bainong 3217 × Mardler' BC5F4 wheat line at the initial stage of inoculation with powdery mildew pathogen (Erysiphe graminis DC) was used to construct a suppression subtractive hybridization (SSH) cDNA library. Totally 760 ESTs were obtained through sequencing. Similarity analysis of ESTs based on BLASTn and BLASTx with the sequences in GenBank, in combination with macroarray differential screening, revealed that 199 ESTs of 65 kinds were known to be functionally disease resistance related. Based on the gene expression profiling in the present study, it is postulated that salicylic acid (SA) and MAP-related signal transduction pathways were involved in powdery mildew resistance in wheat. System acquired resistance genes were predominant in terms of kinds and quantity. With the initiation of cell defense reaction, the genes conferring anti-oxidation substances were largely expressed and thus cell protection mechanism was activated. [Objectives]This study was conducted to construct forward and reserve libraries of suppression subtractive hybridization (SSH) in the testis of male tilapia under the stress of methomyl by using SSH technology.[Methods]Using male tilapia as the test animal,the forward and reserve libraries of SSH in the testis of tilapia under the stress of methomyl were constructed by using the SSH technology.[Results]45 expressed sequence tags (ESTs) were obtained,and 25 expressed sequence tags were successfully noted,including 13 forward libraries and 12 reverse *** genes with confirmed functions were classified into five *** related to catalytic activity and cell characteristics were up-regulated,while genes related to structural molecule's activity and biological process were *** expression amount of integrin β1 was up-regulated,while serine/threonine protein kinase pim-3,Ca^2+-ATPase,Na^+-K^+-ATPase and ribosomal protein L22 were down-regulated.[Conclusions]The research results could lay a foundation for revealing the molecular mechanism of methomyl's reproductive toxicity to tilapia.

In the face of a growing number of large-scale data sets, affinity propagation clustering algorithm to calculate the process required to build the similarity matrix, will bring huge storage and computation. Therefore, this paper proposes an improved affinity propagation clustering algorithm. First, add the subtraction clustering, using the density value of the data points to obtain the point of initial clusters. Then, calculate the similarity distance between the initial cluster points, and reference the idea of semi-supervised clustering, adding pairs restriction information, structure sparse similarity matrix. Finally, the cluster representative points conduct AP clustering until a suitable cluster *** results show that the algorithm allows the calculation is greatly reduced, the similarity matrix storage capacity is also reduced, and better than the original algorithm on the clustering effect and processing speed.

“Cut-and-paste” method for the rapid prototyping of soft electronics

Unlike wafer-based rigid electronics, soft electronics have many unique advantages including thinness, flexibility, stretchability,conformability, lightweight, large area, as well as low cost. As a result, they have demonstrated many emerging capabilities in healthcare devices, soft robotics, and human-machine interface. Instead of conventional microfabrication, there is an evergrowing interest in the freeform or digital manufacture of soft electronics. This review provides a survey for a cost-and timeeffective subtractive manufacturing process called the "cut-and-paste" method. It employs a mechanical cutter plotter to form patterns on various electronically functional membranes such as sheets of metals, functional polymers, and even two-dimensional(2D) materials, supported by a temporary tape. The patterned membranes can then be pasted on soft substrates such as medical tapes or even human skin. This process is completely dry and desktop. It does not involve any rigid wafers and is hence capable of making large-area electronics. The process can be repeated to integrate multiple materials on a single *** circuits(ICs) and rigid components can be added through a "cut-solder-paste" process. Multilayer devices can also be fabricated through lamination. We therefore advocate that the "cut-and-paste" method is a very versatile approach for the rapid prototyping of soft electronics for various applications.

Cloning and characterization of a novel deletion mutant of heterogeneous nuclear ribonucleoprotein M4 from human dendritic cells

To identify differentially expressed genes from antigen-stimulated human dendritic cells (DC), subtractive cloning was adopted and more than ten novel genes differentially expressed were cloned. One is a deletion mutant of heterogeneous nuclear ribonucleoprotein (hnRNP) M4 in which the residues from 159 to 197 of hnRNP M4 have been absent. The deletion mutant was shown to- be co-expressed with hnRNP M4 in cell lines. The mutant was expressed in antigen-stimulated DC but not in normal DC. Northern blot analysis revealed the presence of a major hnRNP M4 deletion mutant mRNA transcript of 2.4 kilobase with the highest levels in peripheral lymphocytes, lung, liver and spleen. It was also expressed in bone marrow-derived stromal cells (BMSC), BMSC treated with several cytokines but not in BMSC treated with TNF-a. The results revealed a new member of hnRNP family and suggested that hnRNP would participate in antigen process and presentation.
