Detection of SARS-CoV-2 Antibodies in Oral Fluid Using a Magnetic Particle-Based Chemiluminescence Immunoassay-Beijing Municipality,China,2021

Introduction:Oral fluids(OFs)have been broadly used as non-invasive samples for evaluating protective IgG antibodies from natural infection or vaccination,especially in pediatric populations.Methods:Paired OF and serum were collected from both individuals who received a booster dose of the inactive coronavirus disease 2019(COVID-19)vaccine as well as those who did not have a history of COVID-19 vaccination and infection(as the control group).The total human IgG antibody(HIgG)content was evaluated as a marker of OF sampling quality.An in-house adapted magnetic particle-based chemiluminescence immunoassay was used for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)IgG antibody detection in the OF.The SARS-CoV-2 IgG antibody in the serum samples was detected using a commercial immunoassay.Results:In total,579 paired OF and serum samples were collected.An additional 172 OF samples were collected from preschool children.The results indicated that the HIgG concentration in qualified OF samples should be higher than 0.3μg/mL.Compared to the serum assay,the in-house OF immunoassay for detecting IgG antibodies against SARS-CoV-2 had 95.06%accuracy,95.03%sensitivity,and 100%Specificity.Conclusions:Overall,the in-house immunoassay for detecting SARS-CoV-2 IgG antibodies in OF showed high potential for application towards serological surveillance and immunization effect assessment after large-scale,inactive COVID-19 vaccination in China.

Methods of manipulating protein in cells are of great value in research and clinical therapies.Indirect methods,such as plasmid and mRNA transfection,and virus-based delivery,have been important skills of life science and medical researchers.Although extensively applied,these nucleic-acids-based approaches are limited by many shortcomings[1].It is very difficult to precisely manipulate the expression level and timeframe of the objective protein in host cells in these methods.The intracellular stabilities of the plasmids or mRNAs that encode the objective genes are affected by several factors,including gene sequence,three-dimensional structure,host nucleases and their delivery processes.These indirect methods require the host transcriptional and translational systems for protein synthesis.However,the transcription and translation machines are tightly regulated by host signaling.In addition,the immunogenicity of DNAs and viral vectors,and the safety of the lipid nanoparticles for mRNA transfection are obstacles of these indirect methods in many clinical applications[2].In contrast,direct protein delivery with target-cell Specificity is more efficient in controlling the quantity and function of the protein of interest.However,due to the hydrophobicity of plasma membrane and high molecular weights of proteins,it remains a great challenge to direct deliver protein into target cells across the intact plasma membrane.

Specificity of Various Mitochondrial DNA (mtDNA), ND5, D-Loop, and Cty-bDNA Primers in Detecting Pig (Sus scrofa) DNA Fragments

Polymerase Chain Reaction (PCR) is an accurate, simple and fast analytical method. This technique is widely used in the identification of meat adulteration and meat-based processed food products. Three Mitochondrial DNA (mt-DNA) primers NADH Dehydrogenase sub unit 5 (ND5), D-Loop, and Cytochrome b (Cyt-b) were tested for their Specificity in detecting of pig (Sus scrofa) DNA fragments. DNA genome from 6 meat samples (pork, beef, goat, lamb, and chicken) was amplified by PCR technique using three pairs of primers (ND5, D-Loop, and Cyt-b) and sequenced. The results of amplification using the three primers produced specific DNA bands with the lengths of 232 bp, 951 bp, and 404 bp, respectively. Comparison results with ND5, D-Loop, and Cyt-b gene sequences resulted in similarity values of 100%, 97%, and 99%, respectively. These showed that the mt-DNA primers of ND5, D-Loop, and Cyt-b genes can be recommended as specific primers in detecting pig (Sus scrofa) DNA fragments.

Identification of BRAF V600E mutation in odontogenic tumors by high-performance MALDI-TOF analysis

Odontogenic tumors are rare lesions with unknown etiopathogenesis.Most of them are benign,but local aggressiveness,infiltrative potential,and high recurrence rate characterize some entities.The MAP-kinase pathway activation can represent a primary critical event in odontogenic tumorigenesis.Especially,the BRAF V600E mutation has been involved in 80%–90%of ameloblastic lesions,offering a biological rationale for developing new targeted therapies.The study aims to evaluate the BRAF V600E mutation in odontogenic lesions,comparing three different detection methods and focusing on the Sequenom MassARRAY System.81 surgical samples of odontogenic lesions were subjected to immunohistochemical analysis,Sanger Sequencing,and Matrix-Assisted Laser Desorption/Ionization-Time of Flight mass spectrometry(Sequenom).The BRAF V600E mutation was revealed only in ameloblastoma samples.Moreover,the presence of BRAF V600E was significantly associated with the mandibular site(ρ=0.627;P valueSpecificity of the driving mutations of mandibular ameloblastomas,providing a biological rational for developing new targeted therapies.Finally,the high diagnostic accuracy of Sequenom was confirmed.

The CRISPR-Cas9 system,serving as a powerful genome-editing technology,has revolutionized the life sciences.However,it exhibits off-target activities that may present severe problems in clinical applications.Although a great number of in silico models have been developed to predict CRISPR targeting efficiency and Specificity,they are running into a bottleneck with the lack of a mechanistic understanding of the on-and off-target activities of Cas9.

Multiscale structural design of MnO_(2)@GO superoxide dismutase nanozyme for protection against antioxidant damage

Rational design of metallic active sites and its microenvironment is critical for constructing superoxide dismutase(SOD)nanozymes.Here,we reported a novel SOD nanozyme design,with employing graphene oxide(GO)as the framework,andδ-MnO_(2)as the active sites,to mimic the natural Mn-SOD.This MnO_(2)@GO nanozyme exhibited multiscale laminated structures with honeycomb-like morphology,providing highly specific surface area for·O_(2)−adsorption and confined spaces for subsequent catalytic reactions.Thus,the nanozyme achieved superlative SOD-like catalytic performance with inhibition rate of 95.5%,which is 222.6%and 1605.4%amplification over GO and MnO_(2)nanoparticles,respectively.Additionally,such unique hierarchical structural design endows MnO_(2)@GO with catalytic Specificity,which was not present in the individual component(GO or MnO_(2)).This multiscale structural design provides new strategies for developing highly active and specific SOD nanozymes.

＜正＞ Twenty - three of 57 sera （40 % ） of Chinese heroin abusers had positive evidence of anti - morphine antibody using original ELISA method. The polyclonal antimorphine antibody from the abusers was purified by affinity chromatography method. The affinity Specificity between the antibody and opiates was investigated respectively by competitive ELISA to determine the formation of antigenic determinants that the antibody recognized. The antibody showed high cross reactivity between heroin, codeine. Morphine - 3 - glucucoid and morphine （maximum ratios of inhibition range: 80% - 100%, values of IC50 range: 10 -6 - 10-4 mol· L-1） and lower cross reactivity between Methadone. oxycodone, etorphine and morphine （maximum ratios of inhibition range: 50% - 75%, values of IC50 range: 10 -5 - 10 -3 mol· L-1）;Naloxone and naltrexone had nearly no inhibitory effect. The results suggest the antibody had a "group Specificity", the active groups （N - ） of agonists to opiate receptors may be the dominant domain that recognized by the antibody, while some substituent on morphine skeleton （3 - , 6 - ） would affect recognition of the antibody.

An adjusted ELISpot-based immunoassay for evaluation of SARS-CoV-2-specific T-cell responses

Like antibody evaluation,using an effective antigen‐specific T‐cell immunity assessment method in coronavirus disease 2019(COVID‐19)patients,survivors and vaccinees is crucial for understanding the immune persistence,prognosis assessment,and vaccine development for COVID‐19.This study evaluated an empirically adjusted enzyme‐linked immunospot assay for detecting severe acute respiratory syndrome coronavirus 2(SARS‐CoV‐2)‐specific T‐cell immunity in 175 peripheral blood samples from COVID‐19 convalescents and healthy individuals.Results of viral nucleic acid were used as the gold standard of infection confirmation.The SARS‐CoV‐2M peptide pool had higher sensitivity of 85%and Specificity of 71%for the single peptide pool.For combined peptide pools,the parallel evaluation(at least one of the peptide pools is positive)of total peptide pools(S1&S2&M&N)had higher sensitivity(up to 93%),and the serial evaluation(all peptide pools are positive)of total peptide pools had higher Specificity(up to 100%).The result of the serial evaluation was better than that of the parallel evaluation as a whole.The detection efficiency of M and N peptide pool serial evaluation appeared the highest,with a sensitivity of 80%and Specificity of 93%.This T‐cell immunity detection assay introduced in this report can achieve high operability and applicability.Therefore,it can be an effective SARS‐CoV‐2‐specific cellular immune function evaluation method.

Antibody-modified integrated microfluidic terahertz biosensor for detection of breast cancer biomarkers

Breast cancer is the most common malignant tumor in women,which seriously threatens the physical and mental health of women worldwide.The existing detection methods have problems,such as large sample consumption,time-consuming sample preparation,expensive equipment,and low sensitivity.In order to solve these problems,this paper proposes a method for quickly detecting breast cancer using surface-functionalized terahertz metamaterial biosensors.The use of PIK3CA-modified sensors enhances the detection sensitivity and Specificity of exosomes.Based on the red shift of the sensor absorption peak caused by exosomes,breast cancer patients can be distinguished from healthy controls.This study demonstrates that exosome detection is effective for the repeatable and non-invasive diagnosis of breast cancer patients.The terahertz metamaterial biosensor designed in this paper has high Specificity,repeatability,and sensitivity,and has great potential for application in the development of modern diagnostic instruments.
