Dynamic DNA nanodevices have gained tremendous attention due to their extraordinary inherent functionality and advantages,however,dynamic DNA nanodevices-based biosensors are still challenging due to their high reliance on proteases and limited amplification capabilities.Herein,exploiting bispecific aptamer as initiators for the first time,we developed a three-dimensional(3D)DNA nanomotor biosensor powered by DNAzyme and entropy-driven circuit for sensitive and specific detection of lysozyme,in which walking and rolling strategies are efficiently integrated to achieve excellent signal amplification capability.Benefiting from the high selectivity of bispecific aptamer,the 3D DNA nanomotor biosensor can respond to lysozyme with high Specificity and operate at high speed to release signals.The whole process is independent of protease,avoiding the influence of adverse environment on the operation stability.Under optimal conditions,it can achieve a limit of detection as low as 0.01 pg/mL with an excellent linear range of 0.05 pg/mL–500 ng/mL for lysozyme.Furthermore,the proposed strategy revealed high accuracy in the analysis of real samples,indicating a great potential for the application of nanomotor biosensors to the detection of non-nucleic acid targets.

Although nanozyme has become an emerging area of research attracting extensive attention recently,the activity and Specificity of currently reported nanozymes are generally lower than those of natural enzymes.Developing highly active and specific nanozymes is therefore extremely necessary and also remains a great challenge.Superoxide dismutase(SOD)catalyzes the disproportionation of cytotoxic O_(2)·^(−)into hydrogen peroxide and oxygen,and plays an important role in reducing human oxidative stress.In this work,we prepare Cu single-atom catalysts(Cu/GO SACs,GO=graphene oxide)through a simple and low-cost strategy at room temperature using Cu foam and graphene oxide.Cu/GO SACs can maintain excellent catalytic activity under harsh environment.Compared with the natural enzyme,SOD-like Cu/GO SAC nanozyme has higher catalytic activity and meanwhile,it does not possess the common properties of other mimic enzymes often existing in nanomaterials.Based on the excellent SOD-like enzyme activity of Cu/GO SACs,it successfully eliminates the active oxygen in cigarette smoke.This work not only provides a new idea for the design and synthesis of nanozymes with excellent SOD mimetic properties,but also is promising in the treatment of lung injury and inflammatory diseases related to free radical production.

The significance and ability for receptor targeted nanoliposomes(tNLs)to bind to their molecular targets in solid tumors in vivo has been questioned,particularly as the efficiency of their tumor accumulation and selectivity is not always predictive of their efficacy or molecular Specificity.This study presents,for the first time,in situ near-infrared(NIR)molecular imaging-based quantitation of the in vivo Specificity of tNLs for their target receptors,as opposed to tumor selectivity,which includes influences of enhanced tumor permeability and retention.Results show that neither tumor delivery nor selectivity(tumor-to-normal ratio)of cetuximab and IRDye conjugated tNLs correlate with epidermal growth factor receptor(EGFR)expression in U251,U87,and 9L tumors,and in fact underrepresent their imaging-derived molecular Specificity by up to 94.2%.Conversely,their in vivo Specificity,which we quantify as the concentration of tNL-reported tumor EGFR provided by NIR molecular imaging,correlates positively with EGFR expression levels in vitro and ex vivo(Pearson’s r=0.92 and 0.96,respectively).This study provides a unique opportunity to address the problematic disconnect between tNL synthesis and in vivo Specificity.The findings encourage their continued adoption as platforms for precision medicine,and facilitates intelligent synthesis and patient customization in order to improve safety profiles and therapeutic outcomes.

The genome of cells is constantly challenged by DNA damages from endogenous metabolism and environmental agents.These damages could potentially lead to genomic instability and thus to tumorigenesis.To cope with the threats, cells have evolved an intricate network, namely DNA damage response(DDR) system that senses and deals with the lesions of DNA.Although the DDR operates by relatively uniform principles, different tissues give rise to distinct types of DNA damages combined with high diversity of microenvironments across tissues.In this review, we discuss recent findings on specific DNA damage among different tissues as well as the main DNA repair way in corresponding microenvironments, highlighting tissue Specificity of DDR and tumorigenesis.We hope the current review will provide further insights into molecular process of tumorigenesis and generate new strategies for cancer treatment.

Molecular Mechanism of the Specificity of Protein Import into Chloroplasts and Mitochondria in Plant Cells

Plants possess both types of endosymbiotic organelles, chloroplasts and mitochondria. Transit peptides and presequences function as signal sequences for specific import into chloroplasts and mitochondria, respectively. However, how these highly similar signal sequences confer the protein import Specificity remains elusive. Here, we show that mitochondrial- or chloroplast-specific import involves two distinct steps, Specificity determination and translocation across envelopes, which are mediated by the N-terminal regions and functionally interchangeable C-terminal regions, respectively, of transit peptides and presequences. A domain harboring multiple-arginine and hydrophobic sequence motifs in the N-terminal regions of presequences was identified as the mitochondrial Specificity factor. The presence of this domain and the absence of arginine residues in the N-terminal regions of otherwise common targeting signals confers Specificity of protein import into mitochondria and chloroplasts, respectively. AtToc159, a chloroplast import receptor, also contributes to determining chloroplast import Specificity. We propose that common ancestral sequences were functionalized into mitochondrial- and chloroplast-specific signal sequences by the presence and absence, respectively, of multiple-arginine and hydrophobic sequence motifs in the N-terminal region.

Diagnostic tests are usually based on some quantitative biomarkers.Two key parameters used to characterise the quality of a test are test sensitivity and Specificity.Predictive values of the disease status based on test results are also of interest in medical research and public health management.In this paper,we study the relations among sensitivity,Specificity and predictive values of the test.The core concept is risk function,which is assumed to be an increasing function of the biomarker.Our results show that test sensitivity and Specificity change in opposite directions.The positive predictive value and the sensitivity also change in opposite directions.Likewise,the negative predictive value and the Specificity change in opposite directions.

Role of helicity of a-helical antimicrobia peptides to improve Specificity

A major barrier to the use of antimicrobial peptides as antibiotics is the toxicity or ability to lyse eukaryotic cells. In this study, a 26-residue amphipathic a-helical antimicrobial peptide A12L/A20L （Ac-KWKSFLKTFKSLK KTVLHTLLKAISS-amide） was used as the framework to design a series of D- and L-diastereomeric peptides and study the relationships of helicity and biological activi- ties of o-helical antimicrobial peptides. Peptide helicity was measured by circular dichroism spectroscopy and demonstrated to correlate with the hydrophobicity of peptides and the numbers of D-amino acid substitu- tions. Therapeutic index was used to evaluate the selectivity of peptides against prokaryotic cells. By introducing D-amino acids to replace the original L-amino acids on the non-polar face or the polar face of the helix, the hemolytic activity of peptide analogs have been significantly reduced. Compared to the parent peptide, the therapeutic indices were improved of 44-fold and 22-fold against Gram-negative and Gram- positive bacteria, respectively. In addition, D- and L-diastereomeric peptides exhibited lower interaction with zwitterionic eukaryotic membrane and showed the significant membrane damaging effect to bacterial cells. Helicity was proved to play a crucial role on peptide Specificity and biological activities. By simply replacing the hydrophobic or the hydrophilic amino acid residues on the non-polar or the polar face of these amphipathic derivatives of the parent peptide with D-amino acids, we demonstrated that this method could have excellent potential for the rational design of antimicrobial pep- tides with enhanced Specificity.

Mechanism of the acupoints of the governor vessel in treatment of post-stroke depression on the base of the Specificity of meridian points

Focusing on the effect Specificity of meridian points along the running course of meridian,by taking the treatment of post-stroke depression(PSD)with the acupoints of the governor vessel as an example,the correlation between meridian points and disease was explored.The specific regulatory effect of governor vessel,Baihui(百会GV20)and Shenting(神庭GV24)in physiology and pathology in PSD and the material basis of the Specificity effect were analyzed.Finally,the specific relationship between the acupoints of the governor vessel and PSD was explained.
