Gli1 promotes epithelial-mesenchymal transition and metastasis of non-small cell lung carcinoma by regulating Snail transcriptional activity and stability

Metastasis is crucial for the mortality of non-small cell lung carcinoma(NSCLC) patients.The epithelial-mesenchymal transition(EMT) plays a critical role in regulating tumor metastasis.Glioma-associated oncogene 1(Gli1) is aberrantly active in a series of tumor tissues. However, the molecular regulatory relationships between Gli1 and NSCLC metastasis have not yet been identified. Herein,we reported Gli1 promoted NSCLC metastasis. High Gli1 expression was associated with poor survival of NSCLC patients. Ectopic expression of Gli1 in low metastatic A549 and NCI-H460 cells enhanced their migration, invasion abilities and facilitated EMT process, whereas knock-down of Gli1 in high metastatic NCI-H1299 and NCI-H1703 cells showed an opposite effect. Notably, Gli1 overexpression accelerated the lung and liver metastasis of NSCLC in the intravenously injected metastasis model. Further research showed that Gli1 positively regulated Snail expression by binding to its promoter and enhancing its protein stability, thereby facilitating the migration, invasion and EMT of NSCLC. In addition, administration of GANT-61, a Gli1 inhibitor, obviously suppressed the metastasis of NSCLC. Collectively, our study reveals that Gli1 is a critical regulator for NSCLC metastasis and suggests that targeting Gli1 is a prospective therapy strategy for metastatic NSCLC.

Background The area of Oncomelania hupensis Snail remains around 3.6 billion m^(2),with newly emerging and reemergent habitats continuing to appear in recent years.This study aimed to explore the long-term dynamics of Snail density before and after the operation of Three Gorges Dam(TGD).Methods Data of Snail survey between 1990 and 2019 were collected from electronic databases and national schistosomiasis surveillance.Meta-analysis was conducted to estimate the Snail density.Joinpoint model was used to identify the changing trend and inflection point.Inverse distance weighted interpolation(IDW)was used to determine the spatial distribution of recent Snail density.Results A total of 3777 Snail survey sites with a precise location of village or beach were identified.For the downstream area,Snail density peaked in 1998(1.635/0.11 m^(2),95%CI:1.220,2.189)and fluctuated at a relatively high level before 2003,then declined steadily from 2003 to 2012.Snail density maintained lower than 0.150/0.11 m^(2) between 2012 and 2019.Joinpoint model identified the inflection of 2003,and a significant decreasing trend from 2003 to 2012 with an annual percentage change(APC)being−20.56%(95%CI:−24.15,−16.80).For the upstream area,Snail density peaked in 2005(0.760/0.11 m^(2),95%CI:0.479,1.207)and was generally greater than 0.300/0.11 m2 before 2005.Snail density was generally lower than 0.150/0.11 m^(2) after 2011.Snail density showed a significant decreasing trend from 1990 to 2019 with an APC being−6.05%(95%CI:−7.97,−7.09),and no inflection was identified.IDW showed the areas with a high Snail density existed in Poyang Lake,Dongting Lake,Jianghan Plain,and the Anhui branch of the Yangtze River between 2015 and 2019.Conclusions Snail density exhibited a fluctuating downward trend in the Yangtze River basin.In the downstream area,the operation of TGD accelerated the decline of Snail density during the first decade period,then Snail density fluctuated at a relatively low level.There still exists local areas with a high Snail density.L

DEAR EDITOR,Bellamya purificata(Gastropoda:Caenogastropoda;Architaenioglossa;Viviparidae:Bellamyinae;Sinotaia),a homotypic synonym of S.purificata,is widely distributed in freshwater habitats in Asia.It is an economically important edible Snail and plays a vital function in freshwater wetland ecology.However,genomic resources for this Snail are lacking and no reference genome has been released.In this study,we constructed the first chromosome-level genome of B.purificata using PacBio long-read sequencing and high-throughput chromosome conformation capture(Hi-C)technology.

Methyltransferase like 13(METTL13),a kind of methyltransferase,is implicated in protein binding and synthesis.The upregulation of METTL13 has been reported in a variety of tumors.However,little was known about its potential function in head and neck squamous cell carcinoma(HNSCC)so far.In this study,we found that METTL13 was significantly upregulated in HNSCC at both mRNA and protein level.Increased METTL13 was negatively associated with clinical prognosis.And METTL13 markedly affected HNSCC cellular phenotypes in vivo and vitro.Further mechanism study revealed that METTL13 could regulate EMT signaling pathway by mediating enhancing translation efficiency of Snail,the key transcription factor in EMT,hence regulating the progression of EMT.Furthermore,Snail was verified to mediate METTL13-induced HNSCC cell malignant phenotypes.Altogether,our study had revealed the oncogenic role of METTL13 in HNSCC,and provided a potential therapeutic strategy.

Combination therapy with miR34a and doxorubicin synergistically inhibits Doxresistant breast cancer progression via downregulation of Snail through suppressing Notch/NF-kB and RAS/RAF/MEK/ERK signaling pathway

Resistance to breast cancer(BCa) chemotherapy severely hampers the patient’s prognosis.MicroRNAs provide a potential therapeutic prospect for BCa.In this study,the reversal function of microRNA34 a(miR34 a) on doxorubicin(Dox) resistance of BCa and the possible mechanism was investigated.We found that the relative level of miR34 a was significantly decreased in Dox-resistant breast cancer cell MCF-7(MCF-7/A) compared with Dox-sensitive MCF-7 cells.Transfection with miR34 a significantly suppressed the invasion,migration,adhesion of MCF-7/A cells without inhibiting their growth obviously.The combination of miR34 a and Dox could significantly inhibit the proliferation,migration,invasion and induce the apoptosis of MCF-7/A cells.The synergistic effect of this combination on resistant MCF-7/A cells has no obvious relation with the expressions of classical drug-resistant proteins P-GP,MRP and GST-π,while closely related with the down-regulation on TOP2 A and BCRP.Moreover,we found both protein and mRNA expression of Snail were significantly up-regulated in MCF-7/A cells in comparison with MCF-7 cells.Transfection with small interfering RNA(siRNA) of Snail could inhibit the invasion,migration and adhesion of drug-resistant MCF-7/A cells,while highexpression of Snail could remarkably promote the invasion,migration and adhesion of MCF-7 cells,which might be related with regulation of N-cadherin and E-cadherin.Transfection with miR34 a in MCF-7/A cells induced a decrease of Snail expression.The potential binding sites of miR34 a with 3’UTR of Snail were predicted by miRDB target prediction software,which was confirmed by luciferase reporter gene method.Results showed that the relative activity of luciferase was reduced in MCF-7/A cells after co-transfection of miR34 a and wild type(wt)-Snail,while did not change by co-transfection with miR34 a and 3’ UTR mutant type(mut) Snail.Combination of miR34 a and Dox induced a stronger decrease of Snail in MCF-7/A cells in comparison to miR34 a or Dox treatment alone.What’ more,for

食管鳞癌组织Snail与PDL-1和CD8^(+)T表达及预后分析

目的探讨食管鳞癌(ESCC)组织中Snail与细胞程序性死亡配体1(PDL-1)、CD8^(+)T之间的相关性及预后观察。方法收集222例2014-02-01-2015-12-30就诊于新乡医学院第一附属医院,手术后经病理确诊为ESCC的癌组织。应用免疫组织化学法检测Snail、PDL-1和CD8^(+)T表达,Spearman相关分析三者的相关性,Kaplan-Meier法绘制生存曲线,Cox回归模型进行预后因素分析。结果Spearman相关分析显示,Snail与PDL-1蛋白表达呈低度正相关,r=0.210,P=0.002;与CD8^(+)T浸润呈低度负相关,r=-0.174,P=0.009。Cox单因素分析显示,远处转移(HR=4.060,95%CI为2.472~6.668,PSnail蛋白高表达(HR=3.781,95%CI为2.421~5.906,PSnail蛋白高表达CD8^(+)T低浸润组(HR=4.130,95%CI为2.033~8.390,PSnail蛋白高表达CD8^(+)T高浸润组(HR=2.872,95%CI为1.385~5.954,P=0.005)患者,总生存期(OS)较差。多因素分析显示,TNM分期(HR=3.335,95%CI为2.249~4.945,PSnail蛋白高表达(HR=1.968,95%CI为1.058~2.906,P=0.046)、Snail蛋白高表达同时CD8^(+)T低浸润(HR=2.616,95%CI为1.249~5.487,P=0.011)是影响ESCC患者预后的独立危险因子。Kaplan-Meier生存分析显示,Snail蛋白高表达组的OS低于低表达组,PSnail与CD8^(+)T联合分析结果显示,Snail高CD8^(+)T低、Snail低CD8^(+)T高及Snail低CD8+T低组患者的OS均高于Snail高CD8^(+)T低表达组,差异有统计学意义,PSnail与PDL-1、CD8^(+)T存在相关性,联合检测Snail与CD8^(+)T对ESCC患者预后预测具有一定参考价值。

研究背景在全球范围内,肝癌的发病率在恶性肿瘤中居于第六位,死亡率排在第四位,严重威胁人类的生命健康。肝细胞癌是一种最常见的恶性肿瘤,在肝癌中占比达到了90%。肝癌患者的早期症状不明显,多数患者被确诊时已经进入晚期阶段。对于晚期肝癌患者,手术治疗等在早期阶段获益大的治疗方式已不适用,主要采用索拉非尼、乐伐替尼等肝癌一线治疗药物进行靶向治疗,从而延长肝癌患者的生存时间。但是由于肿瘤的异质性,药物靶点在不同患者肿瘤组织中的表达存在差异,所以会出现药物不响应和继发性耐药等问题。因此,我们亟需阐明肝癌发生发展的机制,发现新的肝癌药物靶点。牛痘相关激酶1(Vaccinia-related kinase 1,VRK1)是细胞核内的丝氨酸/苏氨酸染色质激酶,参与细胞周期调控、DNA损伤修复、细胞核膜组装、基因转录等重要的细胞生命活动。VRK1在肺癌、乳腺癌、胶质母细胞瘤和食管癌等多种癌症中高表达并与它们的不良预后相关。现有研究表明,VRK1通过调节细胞周期进展促进肝癌细胞的增殖,但VRK1在肝癌迁移和进展中的作用和机制目前尚未有详细阐述。我们利用敲低VRK1的肝癌细胞系,研究VRK1对肝癌细胞增殖和迁移能力的影响,并在裸鼠异种移植瘤模型中验证VRK1对肝癌生物学功能的影响。机制方面,通过RNA测序筛选VRK1调控的下游靶基因以及细胞信号通路,免疫沉淀结合质谱分析确定VRK1的相互作用蛋白。结合上述关键分子的筛选实验阐明VRK1促进肝癌进展的机制。研究目的1.研究VRK1对肝癌细胞增殖和迁移能力的影响。2.揭示VRK1在肝癌中调控的关键下游靶基因以及细胞信号通路。3.探究VRK1调控关键下游靶基因并促进肝癌进展的机制。研究方法1.利用TCGA数据库分析VRK1在肝癌患者肿瘤组织中的表达情况以及与肝癌患者生存期和肿瘤分期之间的关系。2.利用CCK8、克隆形成和Transwell实验检测VRK1对肝癌细胞增殖、迁移能力的影响。3.利用裸鼠异种移植瘤模型检测VRK1对体内肿瘤生长的影响。4.利用RNA测序和GSEA,分析VRK1所调控的下游靶基因和细胞信号通路。5.在敲低VRK1的细胞系中回补靶蛋白Snail,利用EdU和Transwell实验检测VRK1是否通过Snail促进肝癌细胞的增殖和迁移。6.免疫沉淀结合质谱分析确定VRK1的相互作用蛋白。7.利用qRT-PCR和Western blotting检测VRK1的相互作用蛋白CHD1L是否介导了VRK1对SNAI1(编码Snail)表达的调控。8.利用Western blotting检测VRK1是否磷酸化修饰CHD1L。9.利用CHD1L的磷酸化位点失活突变体(CHD1L S122A)研究VRK1是否通过CHD1L S122位点的磷酸化调控Snail的表达。研究结果1.VRK1在肝癌中高表达,与肝癌患者预后不良相关TCGA数据库数据分析显示VRK1在肝癌患者肿瘤组织中高表达,并且VRK1高表达的肝癌患者生存期短。随肝癌患者肿瘤分级和分期的升高,VRK1在肿瘤组织中的表达也升高,VRK1与肝癌患者肿瘤恶性程度具有一定相关性。VRK1在肝癌细胞系中的表达也显著高于正常肝细胞。2.敲低VRK1抑制肝癌细胞的增殖和迁移能力与对照组相比,敲低VRK1组Huh7和HepG2细胞的增殖活力降低、克隆形成数目减少,并且细胞迁移率也降低。3.敲低VRK1抑制体内肿瘤生长与对照组相比,敲低VRK1组裸鼠的瘤体积较小,瘤重较轻,裸鼠体重未有明显波动,表明敲低VRK1抑制体内肿瘤的生长。4.VRK1调控SNAI1表达RNA测序结果显示,对照组与敲低VRK1组之间共存在325个差异表达基因,其中151个基因在敲低VRK1组表达下调,171个基因在敲低VRK1组表达上调。GSEA分析结果显示,差异基因显著富集在上皮-间充质转化通路上,其中间充质主要标志物SNAI1(编码Snail)在敲低VRK1组表达显著下调。qRT-PCR和Western blotting结果证实VRK1调控SNAI1mRNA和蛋白的表达。5.VRK1通过Snail促进肝癌细胞的增殖和迁移挽救实验检测敲低VRK1后回补靶蛋白Snail对肝癌细胞增殖和迁移能力的影响,结果显示VRK1通过Snail促进肝癌细胞的增殖和迁移。6.VRK1与CHD1L相互作用通过免疫沉淀结合质谱分析发现,与对照组相比,在VRK1过表达组中富集的蛋白有175个,其中CHD1L是结合肽段最多的蛋白。经过免疫沉淀实验验证,确认VRK1与CHD1L之间存在相互作用。7.VRK1通过CHD1L调节Snail的表达与敲低VRK1组相比,Snail在敲低VRK1后过表达CHD1L组中蛋白和mRNA的表达量升高,说明VRK1通过CHD1L调节Snail的表达。8.VRK1磷酸化CHD1L S122位点与对照组相比,敲低VRK1组中CHD1L的总体磷酸化水平降低,说明VRK1对CHD1L进

目的探讨沉默信息调节因子1(SIRT1)诱导食管癌EC-9706和Eca-109细胞上皮间质转化(EMT)发生中的作用和机制。方法转染3条化学合成SIRT1 siRNA入食管癌EC-9706和Eca-109细胞,以空白细胞和转染阴性对照siRNA细胞作为对照,real-time PCR和western blot检测转染效果。Transwell侵袭小室和划痕实验检测各组细胞的侵袭转移能力变化。Real-time PCR和Western blot检测各组细胞上皮间质转化相关分子E-cadherin和vimentin的表达变化,同时进一步检测E-cadherin转录抑制因子Snail、twist1和ZEB的表达变化。结果与空白组和阴性对照组细胞相比,转染SIRT1 siRNA1和SIRT1 siRNA3的EC-9706和Eca-109细胞SIRT1 mRNA和蛋白表达均明显降低,差异有统计学意义(PSnail表达下降,与空白组和阴性对照组细胞相比差异有统计学意义(PSnail和twist1表达下降,与空白组和阴性对照组细胞相比差异有统计学意义(PSnail表达诱导EMT发生从而在食管癌细胞侵袭转移中发挥作用。

Background:With the closure of the Three Gorges Dam in 2003 the hydrology of Poyang Lake was altered dramatically leading to significant changes in the environment.In order to assess the impact on schistosomiasis this study assessed the spatial and temporal patterns of the Snail intermediate host,Oncomelania hupensis in the Poyang Lake tributaries.The results of the study have important implications for future Snail control strategies leading to disease elimination.Methods:The marshland area surrounding Poyang Lake was divided randomly into 200×200 m vector grids using ArcGIS software,and the surveyed grids were randomly selected by the software.The Snail survey was conducted in each selected grid using a survey frame of 50×50 m with one sideline of each grid serving as the starting line.No less than ten frames were used in each surveyed grid with Global Positioning System(GPS)recordings for each.All Snails in each frame were collected to determine infection status by microscopy.Altitude data for all frames were extracted from a lake bottom topographic map in order to analyze the average altitude.All Snail survey data were collected and statistically analyzed with SPSS 20.0 software in order to determine the difference of the percentage of frames with living Snails and mean density of living Snails in different regions of Poyang Lake.The altitude of the Snail-infested marshlands and Snail dens were subsequently identified.Results:A total of 1159 potential Snail sampling grids were surveyed,of which 15231 frames(0.1 m2/frame)were investigated.1241 frames had live Oncomelania Snails corresponding to 8.15%of the total number of frames.The mean density of living Snails was 0.463/0.1 m2 with a maximum of 57 Snails per frame.The percent of frames with Snails in the southern sector(8.13%)of Poyang Lake did not differ statistically from the north(8.21%).However,the mean density of live Snails in the northern sector(0.164/0.1 m2)of the lake was statistically higher(F=6.727;P=0.010)than the south(0.141/0.1 m2).In the south

目的探讨核转运蛋白a2(KPNA2)和锌指转录因子Snail在结直肠癌组织中的表达其临床价值。方法选择广东医科大学附属医院2020年5月至2023年5月病理学确诊的123例结直肠癌组织标本及其癌旁组织标本,应用免疫组织化学法检测上述组织中KPNA2和Snail的表达,采用χ^(2)检验分析两者表达与结直肠癌临床病理因素的关系。结果结直肠癌组织中KPNA2表达率为69.11%(85/123),明显高于癌旁组织中KPNA2表达率22.76%(28/123),差异有统计学意义(χ^(2)=53.181,P0.05)。结直肠癌组织中Snail表达率为64.23%(79/123),明显高于癌旁组织中Snail表达率17.89%(22/123),差异有统计学意义(χ^(2)=54.575,PSnail表达水平与结直肠癌患者浸润深度、淋巴结转移、TNM分期明显相关(χ^(2)=4.513、5.192、5.811,PSnail表达水平与结直肠癌患者组织学分化程度、性别、年龄、肿瘤部位无明显相关(χ^(2)=0.015、0.030、0.226、0.022,P>0.05)。结论结直肠癌组织中KPNA2和Snail的表达均升高,进行KPNA2和Snail检测可判断结直肠癌患者病情及预测结直肠癌患者的预后。
