The unicellular green alga Chlamydomonas reinhardtii(hereafter Chlamydomonas)possesses both plant and animal attributes,and it is an ideal model organism for studying fundamental processes such as photosynthesis,sexual reproduction,and life cycle.N^(6)-methyladenosine(m^(6)A)is the most prevalent mRNA modification,and it plays important roles during sexual reproduction in animals and plants.However,the pattern and function of m^(6)A modification during the sexual reproduction of Chlamydomonas remain unknown.Here,we performed transcriptome and methylated RNA immunoprecipitation sequencing(MeRIP-seq)analyses on six samples from different stages during sexual reproduction of the Chlamydomonas life cycle.The results show that m^(6)A modification frequently occurs at the main motif of DRAC(D=G/A/U,R=A/G)in Chlamydomonas mRNAs.Moreover,m^(6)A peaks in Chlamydomonas mRNAs are mainly enriched in the 30 untranslated regions(30 UTRs)and negatively correlated with the abundance of transcripts at each stage.In particular,there is a significant negative correlation between the expression levels and the m^(6)A levels of genes involved in the microtubule-associated pathway,indicating that m^(6)A modification influences the sexual reproduction and the life cycle of Chlamydomonas by regulating microtubule-based movement.In summary,our findings are the first to demonstrate the distribution and the functions of m^(6)A modification in Chlamydomonas mRNAs and provide new evolutionary insights into m^(6)A modification in the process of sexual reproduction in other plant organisms.

Stylonema alsidii(Zandardini)Drew f.ramosum Zheng et Li is a common species in the genus Stylonema,but there are limited reports on morphological taxonomy and a lack of studies on life history and the regulation of early development.The early development,discoid formation,and erect thallus growth of Stylonema alsidii f.ramosum were observed under laboratory conditions.In addition,the influence of main environmental factors such as temperature and light intensity on its development was briefly studied,and various stages of its life history were observed.The result shows:(1)there are two reproductive modes:asexual reproduction and sexual reproduction;(2)the morphology of this species varies in different degrees under laboratory culture conditions;(3)sexual reproduction and asexual reproduction can coexist under suitable conditions.Five pathways of life history were inferred from experimental phenomena.Mature sporophytes form mature gametophytes by producing initially isolated or initially attached monospore,and mature gametophytes form mature sporophytes by fertilization of eggs and sperms.The sexual and asexual reproduction processes of Stylonema alsidii f.ramosum were reported for the first time,and this is also the first report on sexual reproduction in the genus Stylonema.

Regain of sex determination system and sexual reproduction ability in a synthetic octoploid male fish

Polyploids in vertebrates are generally associated with unisexual reproduction,but the direct consequences of polyploidy on sex determination system and reproduction mode remain unknown.Here,we synthesized a group of artificial octoploids between unisexual gynogenetic hexaploid Carassius gibelio and sexual tetraploid Carassius auratus.The synthetic octoploids were revealed to have more than 200 chromosomes,in which 50 chromosomes including the X/Y sex determination system were identified to transfer from sexual tetraploid C.auratus into the unisexual gynogenetic hexaploid C.gibelio.Significantly,a few synthetic octoploid males were found to be fertile,and one octoploid male was confirmed to regain sexual reproduction ability,which exhibits characteristics that are the same to sexual reproduction tetraploid males,such as 1:1 sex ratio occurrence,meiosis completion and euploid sperm formation in spermatogenesis,as well as normal embryo development and gene expression pattern during embryogenesis.Therefore,the current finding provides a unique case to explore the effect of sex determination system incorporation on reproduction mode transition from unisexual gynogenesis to sexual reproduction along with genome synthesis of recurrent polyploidy in vertebrates.

The sexual and asexual reproductive features of the graminoid species Arundinella hirta growing at riversides of the Jialing River were analyzed.It was found that the total seed mass,seed number per plant,and sexual reproductive allocation of A.hirta decreased with decreasing bank elevation,and plants growing at the lowest elevations of banks subjected to intense flooding did not show sexual reproduction.The total plant biomass and the number of ramifications per plant increased with flooding intensity,which implies that,contrary to sexual reproduction,asexual reproduction of A.hirta was enhanced by flooding.

sexual reproduction is prevalent in eukaryotic organisms and plays a critical role in the evolution of new traits and in the generation of genetic diversity.Environmental factors often have a direct impact on the occurrence and frequency of sexual reproduction in fungi.The regulatory effects of atmospheric relative humidity(RH)on sexual reproduction and pathogenesis in plant fungal pathogens and in soil fungi have been extensively investigated.However,the knowledge of how RH regulates the lifecycles of human fungal pathogens is limited.In this study,we report that low atmospheric RH promotes the development of mating projections and same-sex(homothallic)mating in the human fungal pathogen Candida albicans.Low RH causes water loss in C.albicans cells,which results in osmotic stress and the generation of intracellular reactive oxygen species(ROS)and trehalose.The water transporting aquaporin Aqy1,and the G-protein coupled receptor Gpr1 function as cell surface sensors of changes in atmospheric humidity.Perturbation of the trehalose metabolic pathway by inactivating trehalose synthase or trehalase promotes same-sex mating in C.albicans by increasing osmotic or ROS stresses,respectively.Intracellular trehalose and ROS signal the Hog1-osmotic and Hsf1-Hsp90 signaling pathways to regulate the mating response.We,therefore,propose that the cell surface sensors Aqy1 and Gpr1,intracellular trehalose and ROS,and the Hog1-osmotic and Hsf1-Hsp90 signaling pathways function coordinately to regulate sexual mating in response to low atmospheric RH conditions in C.albicans.

Pheromones emitted by both female and male moths regulate coordination between the sexes for Agriphila aeneociliella (Lepidoptera: Crambidae)

The complex and efficient sex pheromone communication system in insects is essential for reproduction and for reproductive isolation of species.In moths,sex pheromone communication starts with male attraction to compounds emitted by females;only a few species act in the reverse.However,how the pheromones that are emitted by both sexes co-regulate and coordinate mate finding and mating remains unknown.Here,we identified both the male and female pheromones of Eastern Grass Veneer moth,Agriphila aeneociliella(Lepidoptera:Crambidae),and demonstrated their efficiency in manipulating behavioral responses of the opposite sex.Combining data from analysis of gas chromatography-electroantennogram detection,gas chromatography-mass spectrometry,and olfactory behavior assays,the female pheromone of A.aeneociliella was identified as(Z,Z,Z)-9,12,15-octadecatrienal and(Z)-9-hexadecenyl acetate,while the male pheromone was determined to be 1-nonanal.Both the 2 individual components of the female pheromone and their binary mixture were significantly attractive to males,and the 1-nonanal male pheromone induced strong electrophysiological responses in females and induced attraction of females in a Y-tube olfactory test.Depending on the concentration of 1-nonanal,its addition to the binary mixture of the female pheromone either enhanced(10^(−3) or 10^(−2)μg/μL)or reduced(1μg/μL)the aphrodisiac effect of the mixture on males.In wind-tunnel bioassays,different concentrations of pheromones,including the binary mixture of female pheromone and the mixture of male and female pheromones,had significant effects on male behavior.Our findings suggested that the blend of both female and male pheromones plays a significant role in the sexual communication system in some moths.

Identification and functional analysis of arabinogalactan protein expressed in pear pollen tubes

Arabinogalactan proteins(AGPs)are widely distributed in the plant kingdom and play a vital role during the process of plant sexual reproduction.In this study,we performed a comprehensive identification of the PbrAGPs expressed in pear pollen and further explored their influences on pollen tube growth.Among the 187 PbrAGPs that were found to be expressed in pear pollen tubes,38 PbrAGPs were specifically expressed in pollen according to the RNA-seq data.The PbrAGPs were divided into two groups of highly expressed and specifically expressed in pear pollen.We further tested their expression patterns using RT-PCR and RT-qPCR.Most of the PbrAGPs were expressed in multiple tissues and their expression levels were consistent with reads per kilobase per million map reads(RPKM)values during pollen tube growth,implying that PbrAGPs might be involved in the regulation of pear pollen tube growth.We also constructed phylogenetic trees to identify the functional genes in pear pollen tube growth.Therefore,19 PbrAGPs(PbrAGP1 to PbrAGP19)were selected to test their influences on pollen tube growth.Recombinant proteins of the 19 PbrAGP-His were purified and used to treat pear pollen,and 11 of the PbrAGP-His recombinant proteins could promote pear pollen tube growth.Additionally,pollen tube growth was inhibited when the expression levels of PbrAGP1 and PbrAGP5 were knocked down using an antisense oligonucleotide assay.PbrAGP1 and PbrAGP5 were localized in the plasma membrane and might not alter the distribution of pectin in the pollen tube.In summary,this study identified the PbrAGPs expressed in pear pollen and lays the foundation for further exploring their functions in pollen tube growth.

Genetic basis and breeding application of clonal diversity and dual reproduction modes in polyploid Carassius auratus gibelio

A unisexual species is generally associated with polyploidy, and reproduced by a unisexual reproduction mode, such as gyno- genesis, hybridogenesis or parthenogenesis. Compared with other unisexual and polyploid species, gibel carp (Carassius au- ratus gibelio) has a higher ploidy level of hexaploid. It has undergone several successive rounds of genome polyploidy, and experienced an additional, more recent genome duplication event. More significantly, the dual reproduction modes, including gynogenesis and sexual reproduction, have been demonstrated to coexist in the polyploid gibel carp. This article reviews the genetic basis concerning polyploidy origin, clonal diversity and dual reproduction modes, and outlines the progress in new va- riety breeding and gene identification involved in the reproduction and early development. The data suggests that gibel carp are under an evolutionary trajectory of diploidization. As a novel evolutionary developmental (Evo-Devo) biology model, this work highlights future perspectives about the functional divergence of duplicated genes and the sexual origin of vertebrate animals.

Ciliated protists are ideal material for studying the origin and evolution of sex,because of their nuclear dimorphism(containing both germline micronucleus and somatic macronucleus in the same cytoplasm),special sexual processes(conjugation and autogamy),and high diversity of mating-type systems.However,the study of sexual process is limited to only a few species,due to the difficulties in inducing or observing conjugation.In the present study,we investigate the conjugation process in Paramecium multimicronucleatum:(1)of the three prezygotic divisions,all micronuclei undergo the first two divisions(meiosis I,II),while a variable number of nuclei undergo the third division(mitosis);(2)the synkaryon divides three times after fertilization,giving rise to eight products that differentiate into four macronuclear anlagen and four micronuclei;(3)cells restore the vegetative stage after two successive cell fissions during which the macronuclear anlagen are distributed into daughter cells without division,while micronuclei divide mitotically;(4)the parental macronucleus begins to fragment following the first meiotic division and finally degenerates completely;(5)the entire process takes about 110 h,of which about 85 h are required for macronuclear development.In addition,we describe for the first time the process of genomic exclusion occurring between amicronucleate and micronucleate cells of P.multimicronucleatum,during which the micronucleate cell contributes a pronucleus to the amicronucleate cell,resulting in both exconjugants being homozygotes.These results provide new insights into the diversity of sexual processes and lay an important cytological basis for future in-depth studies of mating systems in ciliates.

Many fungal pathogens possess multiple reproductive modes,but the role of sexual reproduction is often neglected because of its rarity and complexity.Puccinia striiformis f.***.tritici（Pst） is the causal agent of wheat stripe rust that occurs in most wheat-growing areas throughout the world and is a severe threat to wheat production.sexual reproduction of Pst on the alternate host barberry has been found recently,and the near-Himalayan regions are thought as the hotspot of sexual reproduction.However,the contribution of sexual recombination to the evolution of Pst,such as the emergence of the Yr26-virulent（V26） races in China,remains largely unknown.In the present study,we resequenced the genomes of 42 worldwide isolates of Pst,including 32 isolates from China.Population structure and phylogenetic analysis indicated that all Chinese V26 isolates clustered into one population P4.The significantly low heterozygosity levels,together with the results from the reticulation of the phylogenetic neighbor-net,PHI tests and LD simulations analyses revealed sexual recombination predominantly in P4 and P3,another population from China that was closely related to P4 except for the virulence to Yr26.Selective sweep analysis revealed 24 regions spanning 94 selected genes.Combined with association analysis,45 genes,including six genes encoding predicted secreted proteins,were identified as AvYr26 candidates.Our results provide new evidence for the important role of sexual reproduction in the variation of Pst virulence and demonstrate how sexual reproduction impacts the coevolution of fungal pathogens and their hosts.
