Distribution of per-and polyfluoroalkyl substances in blood,Serum,and urine of patients with liver cancer and associations with liver function biomarkers

Studies have shown that per-and polyfluoroalkyl substances(PFASs)may be hepatotoxic in animals or humans.However,data on clinical epidemiology are very limited.In this study,21PFASs were determined in patients with liver diseases,with the highest median concentrations detected in the Serum sample(26.7 ng/mL),followed by blood(10.7 ng/mL)and urine(5.02 ng/mL).Higher total PFAS concentrations were found in hepatocellular carcinoma(HCC)patients compared to non-HCC patients,with significant discrepancies in Serum and blood samples.Besides,significant correlations were also found among PFAS concentrations and age,gender,body mass index(BMI),and liver function biomarkers levels.For example,PFAS concentrations are significantly higher in males than in females;Several Serum PFASs concentrations increase with age and BMI,while the Serum perfluorohexane sulfonic acid(PFHxS)concentrations are negatively correlated with age.In addition,multiple regression models adjusted for age,gender and BMI found that increased Serum perfluorobutane sulfonic acid(PFBS),perfluoroheptane sulfonic acid(PFHpS)and perfluorohexylphosphonic acid(PFHxPA)conentrations are correlated with elevated alkaline phosphatase(ALP),aspartate aminotransferase(AST),and alpha-fetoprotein(AFP)(p<0.05).Our results provide epidemiological support for the future study on the potential clinical hepatotoxicity of PFAS.

Dietary intake and Serum levels of copper and zinc and risk of hepatocellular carcinoma:A matched case-control study

Background:Copper and zinc are involved in the development of multiple malignancies;yet,epidemiological evidence on hepatocellular carcinoma(HCC)is limited.This study aimed to investigate the association between dietary intake and Serum levels of copper and zinc with the risk of HCC.Methods:A total of 434 case-control pairs matched for sex and age(±1 year)were included in this study.Cases with newly diagnosed HCC were from the Guangdong Liver Cancer Cohort(GLCC)study,and healthy controls were from the Guangzhou Nutrition and Health Study(GNHS).A semi-quantitative 79-item food frequency questionnaire(FFQ)was used to assess habitual dietary intakes of copper and zinc.Serum levels of copper and zinc were measured by using inductively coupled plasma mass spectrometry.The copper(Cu)/zinc(Zn)ratio was computed by dividing copper levels by zinc levels.Conditional logistic regression models were performed to calculate the odds ratio(OR)and 95%confidence intervals(CI)for per 1 standard deviation increase(per-SD increase)in copper and zinc levels.Results:Higher dietary intake(OR_(per-SD increase)=0.65,95%CI:0.44,0.96,P_(trend)=0.029)and Serum levels of zinc(OR_(per-SD increase)=0.11,95%CI:0.04,0.30,P_(trend)Serum zinc levels).Serum copper levels(OR_(per-SD increase)=2.05,95%CI:1.39,3.03,P_(trend)=0.020)and Serum Cu/Zn ratio(OR_(per-SD increase)=6.53,95%CI:2.52,16.92,P_(trend)<0.001)were positively associated with HCC risk,while dietary copper intake and dietary Cu/Zn ratio were not associated with HCC risk.Conclusion:Zinc may be a protective factor for HCC,especially among men,but the effects of copper on HCC risk are not clear.

Serum metabolic profile and metabolome genome‑wide association study in chicken

Background Chickens provide globally important livestock products.Understanding the genetic and molecular mechanisms underpinning chicken economic traits is crucial for improving their selective breeding.Influenced by a combination of genetic and environmental factors,metabolites are the ultimate expression of physiological processes and can provide key insights into livestock economic traits.However,the Serum metabolite profile and genetic archi-tecture of the metabolome in chickens have not been well studied.Results Here,comprehensive metabolome detection was performed using non-targeted LC–MS/MS on Serum from a chicken advanced intercross line(AIL).In total,7,191 metabolites were used to construct a chicken Serum metabo-lomics dataset and to comprehensively characterize the Serum metabolism of the chicken AIL population.Regula-tory loci affecting metabolites were identified in a metabolome genome-wide association study(mGWAS).There were 10,061 significant SNPs associated with 253 metabolites that were widely distributed across the entire chicken genome.Many functional genes affect metabolite synthesis,metabolism,and regulation.We highlight the key roles of TDH and AASS in amino acids,and ABCB1 and CD36 in lipids.Conclusions We constructed a chicken Serum metabolite dataset containing 7,191 metabolites to provide a refer-ence for future chicken metabolome characterization work.Meanwhile,we used mGWAS to analyze the genetic basis of chicken metabolic traits and metabolites and to improve chicken breeding.

To the Editor:The risk factors for the development of radiographic axial spondyloarthritis(SpA)are still uncertain.Previous studies found that a high concentration of Serum uric acid(SUA)leads to deposition of monosodium urate(MSU)in different joints of other rheumatic arthritis diseases except gout.Furthermore,it has been reported that gout could mimic SpA.Thus,our study explored whether the development of radiographic axial SpA is related to high SUA concentration.

Serum triglycerides concentration in relation to total and cardiovascular mortality in an elderly Chinese population

OBJECTIVE To investigate Serum triglycerides in relation to all-cause,cardiovascular,and non-cardiovascular mortality in an elderly Chinese population.METHODS The study participants(n=3565)were elderly(≥60 years)community dwellers living in a suburban town of Shanghai.Hypertriglyceridemia was defined as a Serum triglycerides concentration≥2.30 mmol/L(definite)and≥1.70 mmol/L(borderline),respectively.RESULTS The prevalence of definite and borderline hypertriglyceridemia at baseline was 7.5%and 29.5%,respectively.It was higher in women(n=1982,9.0%and 33.8%,respectively)than men(n=1583,6.2%and 27.9%,respectively),in obese and over-weight participants(n=1566,10.5%and 36.4%,respectively)than normal weight participants(n=1999,5.6%and 27.1%,respect-ively),and in diabetic participants(n=177,11.9%and 39.0%,respectively)than non-diabetic participants(n=3388,7.5%and 30.8%,respectively).During a median of 7.9 years follow-up,all-cause,cardiovascular and non-cardiovascular deaths occurred in 529,216 and 313 participants,respectively.In analyses according to the quintile distributions of Serum triglycerides concentration,the sex-and age-standardized mortality rate was lowest in the middle quintile for all-cause,cardiovascular and non-cardiovascu-lar mortality(18.6,7.8 and 11.9 per 1000 person-years,respectively,versus 21.5,10.5 and 12.7 per 1000 person-years,respectively,in the two lower quintiles and 21.7,9.5 and 14.0 per 1000 person-years,respectively,in the two higher quintiles).The fully adjus-ted hazard ratios(95%CI)for the middle quintile versus the combined two lower with two higher quintiles were 0.85(95%CI:0.67-1.07,P=0.17),0.81(95%CI:0.54-1.19,P=0.28)and 0.87(95%CI:0.64-1.17,P=0.35)for all-cause,cardiovascular and non-cardiovascular mortality,respectively.CONCLUSIONS Our study showed high prevalence of hypertriglyceridemia,especially when defined as borderline and in obese and overweight participants,and mildly but non-significantly elevated risks of cardiovascular mortality relative to the middle level of Serum trig

According to current guidelines,patients with chronic hepatitis B virus(HBV)infection whose HBVDNA is positive but whose alanine aminotransferase(ALT)levels are normal do not currently need antiviral treatment, However,studies have found that even chronic HBV infection'patients withnormal ALTmay,progress to cirrhosis or hepatocellular carcinoma(HCC).Furthermore,almost half of patients with normal ALT levels were found to have moderate to severe inflammation and/or significant fibrosis[2]Currently there were only a few studies on the treatment of chronic HBV patients with normal ALT levels,and the factors that affect their virological response(VR)are still unclear.An increasing number of studies have proven that Serum HBV RNAcan be used as a non-invasive indicator to predict a VR,i3i In this study,weexplored the predictivevalue of HBV RNA in anticipating a VR in normal ALTchronic HBV patients.

Yolk free egg substitute improves the Serum phospholipid profile of mice with metabolic syndrome based on lipidomic analysis

In this study,the impacts of egg consumption on mice model of metabolic syndrome(Met S)were comparatively investigated.Mice were divided into five groups(n=8):normal diet group(ND),high-fat diet group(HFD),HFD with whole egg group(WE),HFD with free-yolk egg substitute group(YFES),and HFD with lovastatin group(Lov).Main biochemical indexes and a non-targeted lipidomic analysis were employed to insight the lipid profile changes in Serum.It was revealed that WE could significantly improve Serum biochemical indexes by reducing body weight,low-density lipoprotein cholesterol(LDL-C)and total cholesterol(TC),while increasing high-density lipoprotein cholesterol.YFES exhibited remarkably better performance in increasing phosphatidylglycerol and phosphatidic acids,while decreasing phosphatidylinositol than WE.A total of 50 differential lipids biomarkers tightly related to glycerophospholipids metabolism were screened out.Carnitine C18:2 and C12:1,SM(d18:0/12:0),and SM(d18:1/14:1)were significantly upregulated in YFES compared to WE.YFES reduced expression of SREBP-1c and Cpt1a,while did not affect the expression of PPAR-α.Sphingomyelin biomarkers were positively related to the TC(|r|>0.6),while PPAR-αwas negatively correlated with triglyceride and LDL-C levels.To sum up,YFES attenuated HFD-induced Met S by improving the Serum phospholipids,which account for its modulation of glycerophospholipid metabolism.
